Abstract:Objective:To optimization preparation technology of sinomenine hydrochloride-polyvinyl alcohol (SM-PVA) microcapsules,and investigate its characterization and in vitro release performance. Method: With drug loading as index,the amount of emulsifier,stirring speed,volume fraction of glutaraldehyde and dosage of cyclohexane(oil phase) were chosen as factor,SM-PVA microcapsules was prepared byW/O/W multiple emulsion interfacial polymerization method,preparation technology of SM-PVA microcapsules was optimized by orthogonal test;drug loading was determined by UV spectrophotometric,morphology of SM-PVA microcapsules was scanned by electron microscopy,particle size of SM-PVA microcapsules was charactered by particle size analysis instrument,and in vitro release performance was investigated. Result: Optimum preparation technology was:7% emulsifier,stirring speed 600 r·min-1,volume fraction of glutaraldehyde 10 mLand 25% cyclohexane,drug loading 6.23%;Average particle size of SM-PVA microcapsules 16 μm,it showed uniform dispersion and smooth surface of spherical.These prepared SM-PVA microcapsules showed good sustained release in simulated gastric and intestinal fluid. Conclusion: SM-PVA microcapsules could be a novel drug delivery system of sinomenine hydrochloride from Chinese materia medica.
