HPLC Fingerprints of Amino Acids Constituents in Cervil Cornus Colla

    Abstract:

    Objective: To establish a method of fingerprint analysis of the amino acids on Cervil Cornus Colla by HPLC. Method: Cervil Cornus Colla was derivated by Phenyl isothiocyanate (PITC). The Chromatographic fingerprints were obtained by the Diamonsil C18 (2) column (4.6 mm×250 mm, 5 μm) with the gradient elution solvent system composed of acetonitrie-natrium aceticum buffer solution (pH 6.5). The detective wavelength was set at 254 nm, the flow rate was 1 mL·min-1 and the column temperature was maintained at 43 ℃. Result: The result showed that the amino acids constituents had a high similarity. 18 kinds of amino acids in Cervil Cornus Colla were detected, of which 13 peaks were identified in common peaks. Conclusion: The method has good reproducibility and stability, and could be used for the research and the quality control of Cervil Cornus Colla.
