Effect of Thujone on Human Peripheral Blood γδ T Cells Function in Vitro

    Abstract:

    Objective: To investigate the effect of thujone on human peripheral blood γδ T cells function in vitro. Method: The human peripheral blood γδ T cells were expanded using in vitro isopentenyl pyrosphate method. After culturing with different concentrations of thujone for 48 hours, the growth curve of γδ T cells was tested using CCK-8 method;the cell phenotype, the cell cycle, the cell apoptosis, and the expression of CD107a, perforin, granzyme B were tested using flow cytometer. Result: Thujone could obviously suppress the γδ T cells growth with the concentration range of 300-600 μmol· L-1(P<0.05), while thujone could proliferate the γδ T cells's growth obviously with the range of 0.036 25-150 μmol· L-1(P<0.05). The peak of proliferation for thujone was (37.03±0.52)%, and the expression rate of proforin was increased by (39.94±1.04)% at the concentration of 9.4 μmol· L-1(P<0.05). Conclusion: Thujone can obviously proliferate the γδ T cells and up-regulate the expression of preforin of γδ T cells at the concentration of 9.4 μmol· L-1.
