    目的: 研究中药清胰化积复方联合化疗药物吉西他滨对人胰腺癌细胞SW1990凋亡及Bcl-2蛋白表达的影响。 方法: 将荷SW1990裸小鼠随机分为模型组、化疗组、化疗联合中药高、中、低剂量组,计算各组治疗后抑瘤率,并采用流式细胞术检测药物作用后肿瘤细胞的凋亡率,采用免疫组化法检测各组Bcl-2蛋白的表达情况。 结果: 化疗联合中药各组的瘤重均小于化疗组,化疗联合中药中剂量组的肿瘤细胞凋亡率高于化疗组,Bcl-2蛋白表达程度下降,有显著性差异。 结论: 清胰化积复方与吉西他滨联合应用,对人胰腺癌细胞SW1990体内生长具有协同抑制作用,下调Bcl-2蛋白的表达,促进细胞凋亡可能是其作用机理之一。

    Abstract:

    Objective: To explore the effect of the herbal decoction Qingyi Huaji formula (QYHJ) combined with gemcitabine on apoptosis and expression of Bcl-2 protein of human pancreatic cancer SW1990 in vivo. Methods: Tumor-burdened nude mice were randomized into control group, Chemo group, groups of Chemo plus QYHJ at different doses. After treatment, inhibiting rates of tumor growth were calculated. Cell apoptosis of each group was extracted by flow cytometry(FCM). Expression of Bcl-2 protein was learned by method of immunohistochemistry. Result: Tumor weight in the Chemo plus QYHJ groups at different doses was significantly lower than that in the Chemo group (P<0.05). Compared to the Chemo group, cell apoptosis rate in the Chemo plus QYHJ groups at middle dose was higher (P<0.05), while the expression of Bcl-2 was lower (P<0.05). Conclusion: QYHJ formula may have synergtic action on Gemcitabine, to inhibit the proliferation of human pancreatic cancer SW1990 in vivo, possibly by inhibiting expression of Bcl-2 protein and inducing cell apoptosis.
