大防风汤治疗Ⅱ型胶原纤维诱导小鼠类风湿性关节炎的实验研究

Experimental Studies on the Treatment of Type Ⅱ Collagen Induced Arthritis with Da-Fang-Feng-Tang

摘要:

目的 :观察大防风汤对于类风湿性关节炎的治疗效果。 方法 :采用Ⅱ型胶原纤维诱导的NIH小鼠类风湿性关节炎动物模型,从分子生物学、免疫学、病理学等方面对大防风汤颗粒进行研究。 结果 :模型对照组与给药组相比,发病率较高,足部红肿程度较重,症状持续时间较长,血清IgG水平明显增高(P<0.05);IL-1,TNF-α和IFN-γ的mRNA基因表达水平均较高。 结论 :证明大防风汤对于Ⅱ型胶原纤维诱导的类风湿性关节炎动物模型具有明显的抑制作用,实验结果支持大防风汤在临床上治疗类风湿性关节炎有效的结论。

Abstract:

Objective: To observe the effect of Da-Fang-Feng-Tang(DFFT) in treating of Rheumatoid Arthritis(RA). Methods : The animal model of type Ⅱ collagen induced arthritis was used. Granules of the DFFT extract were administered by gastric gavage, studying in genetics, immunology, physiology and clinical. Results : The results indicated a higher onset rate, a severer redness and swelling for arthritic limb and a longer time of clinic manifestation in contract group than treated group, and the IgG level in sera was significantly increased compared contract group to treated group(P<0.05). The gene express of IL-1, TNF-α and IFN-γ were higher in contract group. Conclusion : DFFT express obviously suppressive function on model of type Ⅱ collagen induced arthritis in NIH mouse, and support the belief that DFFT is effective in treating human RA.
