地黄叶总苷对兔IgG加速型大鼠肾毒血清肾炎的作用研究

Effects of Total Saponins Extracted from Leaves of Rehmannia on Accelerated Nephrotoxic Nephritis Induced by Rabbit IgG in Rat

摘要:

目的 :观察地黄叶总苷对兔IgG加速型大鼠肾毒血清肾炎的治疗作用。 方法 :通过制备兔抗大鼠基底膜抗体肾毒血清对大鼠尾静脉进行注射造模后,给予本品口服16 d观察对模型动物血生化、尿蛋白、肾脏病理等指标,评定受试物对肾炎的治疗功效。 结果: 给药后,地黄叶总苷可明显降低模型动物的24 h尿蛋白、血浆总胆固醇、尿素氮,升高外周血浆白蛋白含量,并具有使模型大鼠血液流变性正常化的作用,同时可减轻模型肾脏的病理学变化。 结论 :地黄叶总苷20~40 mg·kg^-1口服16 d,对兔IgG加速型肾毒血清肾炎有明显的治疗作用,并且有一定的量效关系。

Abstract:

Objective: To investigate therapeutic effects of total saponins extracted from leaves of rehmannia on accelerated nephrotoxic nephritis induced by rabbit IgG in rat. Method: Rephrotoxic serum containing antibody to rat basilar membrane was preparcd in rabbits.For establishing accelerated nephrotoxic mephritis in rat,the seruem was obtained and given to rat by injection through tail venule.Total saponins extracted from leaves of rehmannia were orally given to the model rats as treatment for 16 days at doses of 20~40 mg·kg^-1.Blood biochemeical parameters,urine protein,hemorheological parameters and pathological changes of nephritis were employed to evaluate the efficacy the efficacy of the treatment. Results :Compared with the model rats, plasma albumin,cholesterol and urea nitrogen were significantly increased. There was also a tendency of normalization for hemorheological parameters.The pathological changes of nephritis were alleviated. Conclusion :Total saponins extracted from leaves of rehamannia has therapeutic effect with a dose-response manner on accelerated nephrotoxic nephritis induced by rabbit IgG in rat.
