HPLC测定淡竹叶中日本当药黄素的含量

HPLC Determination of Swertiajaponin in Leaves of Lophatherum gracile 1,3*

摘要:

目的: 建立中药淡竹叶中日本当药黄素的RP-HPLC含量测定方法。 方法: 高效液相色谱法,采用Cosmosil 5 C₁₈-ms-Ⅱ色谱柱(4.6 mm×250 mm, 5 μm),流动相乙腈-0.05%醋酸水溶液(14:86),流速1.0 mL ·min^-1,分析时间20 min,检测波长350 nm,进样体积10 μL。 结果: 日本当药黄素进样量与峰面积呈良好线性关系,线性范围为0.3~5.0 μg, r=1.000 0 (n=5),回收率为100.2% (n=6)。 结论: 该方法首次测定了8个地区淡竹叶药材中日本当药黄素的含量,该方法准确、简便、重复性好。

Abstract:

Objective: To develop a reversed-phase HPLC method for the determination of swertiajaponin in the leaves of Lophatherum gracile. Method: Cosmosil 5 C₁₈-ms-Ⅱ (4.6 mm×250 mm, 5 μm) column was adopted with the mobile phase of acetonitrile-0.05% acetic acid solution (14:86); the flow rate was 1 mL ·min^-1, and the detection wavelength was set at 350 nm. Result: The calibration curve was linear in the range of 0.3~5.0 μg (r=1.000 0); the average recovery was 100.2 %. Conclusion : The content of swertiajaponin in the leaves of L. gracile from 8 different sources was determined for the first time. The developed method is simple, accurate with good reproducibility.
