    目的: 探讨参芎化瘀胶囊对血管性痴呆(vascular dementia,VD)模型大鼠海马CA1区细胞凋亡及B细胞淋巴瘤/白血病-2(B cell lymphoma/lewkmia-2,Bcl-2),Bax蛋白表达的影响。 方法: 采用反复夹闭、再通双侧颈总动脉同时ip硝普钠制备大鼠血管性痴呆模型,应用Morris水迷宫检测各组大鼠学习记忆能力,TUNEL法检测细胞凋亡,免疫组织化学SABC法检测Bcl-2,Bax蛋白表达。 结果: ① 与正常组及假手术组相比,模型组大鼠海马CA1区凋亡细胞增多(P<0.01);与模型组比较,参芎化瘀胶囊治疗组大鼠海马CA1区凋亡细胞减少(P<0.01)。② 与正常组及假手术组相比,模型组大鼠海马CA1 区Bcl-2,Bax阳性细胞均增多(均P<0.01),Bcl-2/Bax降低(P<0.01);与模型组比较,参芎化瘀胶囊治疗组大鼠海马CA1区Bcl-2阳性细胞增多(P<0.01),Bax阳性细胞减少(P<0.01),Bcl-2/Bax增加(P<0.01)。 结论: 参芎化瘀胶囊可能通过上调Bcl-2蛋白表达,下调Bax蛋白表达,减少细胞凋亡,从而改善血管性痴呆大鼠的学习记忆能力。

    Abstract:

    Objective: To study the effects of Shenxiong Huayu Capsule on the apoptosis and the expression of Bcl-2 and Bax protein in CA1 Area of hippocampus in vascular dementia rats. Method: The vascular dementia model of rat was established by repeated close and open of the common carotid arteries at both sides and intraperitoneal injection of sodium nitroprusside (SNP). The learning and memory abilities of rats were detected by Morris water maze. The cell apoptosis was detected by TUNEL staining and the expression of Bcl-2 and Bax protein were detected by immunohistochemical SABC staining. Result: ① The number of cell apoptosis in the CA1 area of hippocampus was increased in the VD model group compared with that in the sham-operation group and blank control group (P<0.01). The number of cell apoptosis in the CA1 area of hippocampus was decreased in Shenxiong Huayu Capsule intervention group than in the VD model group (P<0.01). ②The expression of Bcl-2, Bax in the CA1 area of hippocampus was increased in the VD model group compared with that in the sham-operation group and blank control group (all P<0.01), Bcl-2/Bax was decreased (P<0.01). The expression of Bcl-2 in the CA1 area of hippocampus was increased in Shenxiong Huayu Capsule intervention group compared with that in the VD model group (P<0.01), Bax was decreased(P<0.01) and Bcl-2/Bax was increased (P<0.01). Conclusion: Shenxiong Huayu Capsule can inhibit apoptosis by up-regulation of expression of Bcl-2 protein and down-regulation of Bax protein in CA1 area of hippocampus in vascular dementia rats and improve the learning and memory abilities of for the vascular dementia rats.
