目的 :研究蒺藜提取物对阿霉素(ADR)所致大鼠心肌细胞凋亡的影响。 方法 :将蒺藜提取物灌胃于ADR所致心肌细胞凋亡的大鼠,采用电镜对大鼠心肌细胞进行超微结构观察,采用TUNEL法检测心肌细胞凋亡指数,免疫组化SP法检测心肌细胞Bax,Bcl-2蛋白的表达。 结果 :ADR累积用量20 mg ·kg-1可致大鼠心肌细胞凋亡,蒺藜提取物中、高剂量组(40,60 mg ·kg-1)可减轻心肌细胞超微结构的损伤,蒺藜提取物中、高剂量组较模型组均可显著降低细胞凋亡率( P< 0.05)。蒺藜提取物中、高剂量组可显著上调Bcl-2表达( P <0.01)、下调Bax表达( P <0.05)。 结论: 蒺藜提取物可抑制ADR所致大鼠心肌细胞的凋亡,对阿霉素所致的心脏毒性有保护作用。
Abstract:
Objective : To investigate the effect of extract from Tribulus terrestris on myocardial cell apoptosis caused by adriamycin in rats. Method : Rats were given intraperitoneal injection of adriamycin (ADR) to induce cardiocyte apoptosis. After continuous ig of extract from T. terrestris , myocardial ultrastructure in every experimental group was observed with electron microscope. The apoptotic rate of myocardial cells was determined by terminal deoxynucleotidyl transferaseediated dUTP nick-end labeling (TUNEL), and the expression of Bcl-2 and Bax was detected by using immuno-histochemical method. Result : ADR of 20 mg ·kg-1 induced cardiocyte apoptosis. Extract from T. terrestris middle-and high-dosage groups (40,60 mg ·kg-1) could reduce cardiotoxicity injury by ADR. Extract from T. terrestris middle-and high-dosage groups could significantly increase the Bcl-2 expression ( P <0.01), reduced Bax expression ( P <0.05). Conclusion : Extract from T. terrestris may decrease the cardiocyte apoptosis induced by adriamycin, and it has a protective effect on adriamycin-induced cardio-toxicity.
