紫花地丁抗炎作用及机制研究

    目的: 观察紫花地丁水提物及醇提物的抗炎作用,并分析其抗炎机制。 方法: 取ICR小鼠随机分模型对照组,布洛芬组(0.08 g·kg-1),紫花地丁水提物高、低剂量(9.0,3.0 g·kg-1)组,紫花地丁醇提物高、低剂量(9.0,3.0 g·kg-1)组,每天1次,连续ig给药7 d,末次给药30 min后,采用二甲苯致小鼠耳肿胀法和角叉菜胶致小鼠足肿胀法观察抗炎作用,并采用紫外分光光度法和酶联免疫吸附法(enzyme-linked immunosorbent assay,ELISA)检测炎性组织中前列腺素E2(PGE2,prostaglandin E2)和血清中肿瘤坏死因子(tumor necrosis factor-alpha,TNF-α)、白细胞介素-1β(interleukin-1β,IL-1β)的含量,探讨抗炎作用机制。 结果: 紫花地丁水提物及醇提物各剂量组(9.0,3.0 g·kg-1)对二甲苯致小鼠耳肿胀及角叉菜胶致小鼠足肿胀均具有显著的抑制作用,与模型对照组比较,差异均具有统计学意义(P<0.05~P<0.01);并可不同程度降低角叉菜胶致炎小鼠血清TNF-α,IL-1β及炎性组织中PGE2含量,高剂量组(9.0 g·kg-1)的作用显著,与模型对照组的差异具有统计学意义(P<0.01)。 结论: 紫花地丁水提物及醇提物均具有显著的抗炎作用,其抗炎作用机制可能与降低TNF-α,IL-1β及PGE2的表达有关。

    Abstract:

    Objective:To observe the anti-inflammatory effects of Viola yedoensis, and analyze the mechanism. Method: The models of ear edema induced by xylene and paw edema induced by carrageenan in mice were used to observe the anti-inflammation effects, the spectrophotometric method and the enzyme-linked immunosorbent assay (ELISA) were used separately to measure the content of prostaglandin E2 (PGE2) in inflammatory tissue and the content of tumor necrosis factor-alpha (TNF-α), interleukin-1β (IL-1β) in serum. Result: Compared with the model control group, the aqueous extraction and alcohol extraction of V. yedoensis (9.0, 3.0 g·kg-1) could significantly inhibit the inflammation caused by xylene or carrageenan (P<0.05-P<0.01), and decreased the content of PGE2 in inflammatory tissue and the content of TNF-α, IL-1β in serum of the mice. induced by carrageenan. There were significant difference between the high dose group (9.0 g·kg-1) and the model control group (P<0.01). Conclusion: The aqueous extraction and alcohol extraction of V. yedoensis have significant anti-inflammation effects, which may be related to the inhibition of the generation of TNF-α, IL-1β and PGE2.
