HPLC Assaying of Aconitine in Xiaojin Pills

摘要:

目的: 建立高效液相色谱法测定小金丸中乌头碱的含量。 方法: 采用ultimate C₁₈柱(4.6 mm×250 mm,5 μm),以甲醇-0.03 mol·L^-1磷酸二氢钾溶液-三乙胺(45:55:0.04)为流动相,流速1.0 mL·min^-1,检测波长240 nm,柱温25℃。 结果: 乌头碱在0.114~2.28 μg具有良好的线性关系(r=0.999 8),平均回收率为97.79%,RSD 2.31%。 结论: 该方法准确、灵敏,重复性好, 可用于小金丸中乌头碱的含量测定。

Abstract:

Objective: To establish a HPLC method for the assaying of aconitine in Xiaojin pills. Method: C₁₈ (4.6 mm×250 mm,5 μm) was used with methanol-30 mmol·L^-1phosphate solution-triethylamine (45: 55: 0.04) as the mobile phase. The flow rate was 1.0 mL·min^-1 and the detection wavelength was set at 240 nm. Result: Aconitine concentration presented a good linear range of 0.114-2.28 μg (r=0.999 8). The average recovery was 97.79% and RSD was 2.31%. Conclusion: This method is rapid, simple, accurate and suitable for the quality control of the pill.
