Objective: To establish a method for determining polysaccharides in Atractylodis rhizoma, then compare the content of polysaccharides in Atractylodis rhizoma before and after processing from different sources and to investigate the effects on the content of polysaccharides in Atractylodis rhizoma by stir-frying with wheat bran. Method: The samples were extracted with water; the phenol-sulfuric acid method was used to detect the polysaccharide content. Result: The content of polysaccharides in Atractylodis rhizoma from 17 sources was determined, the glucose concentration value C and absorbance A were linear in the range of 0.004 08-0.012 24 mg·L-1 (r=0.999 4), the average recovery was 101.92%, RSD 2.58%. The content of polysaccharides in Atractylodis rhizoma from various cultivation locations varied from 3.45%-24.6%, and 3.34%-17.45% in its processed product. Conclusion: The content of polysaccharides in Atractylodis rhizoma from different sources was different, the content of polysaccharides in Atractylodis rhizoma decreased after being processed.
