Objective: To establish a HPLC method for simultaneous determination of rutin, forsythoside A and phillyrin of Forsythiae Fructus and its extract. Method: The procedure of HPLC-PDA was performed on the chromatographic column of Sunfire C18 (4.6 mm×150 mm, 5 μm), and the mobile phase in the gradient elution (0 min, 14%A; 15 min, 18%A; 40 min, 30%A) was acetonitrile(A)-0.2% acetic acid solution (B). The flow rate was 1.0 mL·min-1; the temperature was kept at 40℃, and the detection wavelength was set at 354, 329 nm and 277 nm respectively. Result: Rutin, forsythoside A and phillyrin were linear in the range of 0.1-2.0, 0.12-2.4 μg and 0.05-1.0 μg respectively. The average recoveries were 98.63%, 98.97% and 102.18% respectively (RSD 7.94%, 0.93% and 1.63%). Conclusion: The method is accurate with higher repeatability, which can be used in the quality control of Forsythiae Fructus and its extract.
