Objective: To establish a HPLC method for determination of gallic acid and quercetin in Houerhuan Xiaoyan Jiaonang. Method: The analytical column was Hypersil BDS-C18(4.6 mm×150 mm, 5 μm), and the mobile phase was methanol-phosphate solution with the gradient elution. The flow rate was 1.0 mL·min-1 and the detection wavelength was 254 nm. The column temperature was kept at 30℃, and the sample volumn was 10 μL. Result: A good linearity was obtained in the range of 15.0-180.0 mg·L-1 (r=0.999 6) and 1.25-15.0 mg·L-1 (r=0.999 7) for gallic acid and quercetin respectively, and the average recovery was 98.2% (RSD 0.7%) and 99.2% (RSD 0.5%) respectively. Conclusion: The method is convenient, accurate, precise and can be used in the content determination of gallic acid and quercetin in Pithecellobium clypearia antiphlogistic capsule at the same time, and can be used for the quality control of this capsule.
