高效液相色谱法测定风湿定胶囊中欧前胡素和异欧前胡素的含量

Determination of Imperatorin and Isoimperatorin in Fengshiding Capsule by HPLC

摘要:

目的：建立高效液相色谱法测定风湿定胶囊中欧前胡素和异欧前胡素的含量。方法：Waters e2695 HPLC色谱系统（Empower Waters 工作站，Waters 2489 UV/visible Detector检测器，Hypersil ODS2色谱柱（4.6 mm×150 mm，5 μm），柱温25 ℃，流速1.0 mL·min^-1，流动相甲醇-水（62：38），检测波长254 nm，进样量20 μL。结果：欧前胡素在1.064~31.92 mg·L^-1呈良好的线性关系（r=0.999 7），异欧前胡素在1.090~32.70 mg·L^-1呈良好的线性关系（r=0.999 3）。欧前胡素的平均加样回收率100.2%（n=9），RSD 1.47%，异欧前胡素的平均加样回收率为100.0%（n=9），RSD 1.64%，结论：该方法操作简便、准确、稳定、灵敏度高，适用于风湿定胶囊中欧前胡素和异欧前胡素的质量控制。

Abstract:

Objective: To establish a HPLC method for determination of the content of imperatorin and isoimperatorin in Fengshiding capsule. Method: Hypersil ODS2 column (4.6 mm×150 mm, 5 μm) was used. The methanol-water (62:38) was used as mobile phase. The flow rate was 1.0 mL·min^-1, the detection wavelength was set at 254 nm. The column temperature was set at room temperature. The injection volume was 20 μL. Result: The linear range of imperatorin was 1.064-31.92 mg·L^-1 (r=0.999 7). The linear range of isoimperatorin was 1.090-32.70 mg·L^-1 (r=0.999 3). The average recovery of imperatorin was 100.2% (n=9), RSD 1.47%. The average recovery of isoimperatorin was 100.0% (n=9), RSD 1.64%. Conclusion: The method is simple, sensitive, and accurate for the quality control of Fengshiding capsule.
