    摘要:

    目的:建立鹿角胶氨基酸类成分的高效液相色谱指纹图谱。方法:采用异硫氰酸苯酯(PITC)柱前衍生法建立高效液相指纹图谱,梯度洗脱进行分离,选用Diamonsil C18(2)(4.6 mm×250 mm,5 μm)色谱柱,流动相为乙腈-醋酸钠缓冲液(pH 6.5),检测波长254 nm,柱温43 ℃,流速1.0 mL·min-1结果:共指认13个共有峰,10批样品具有较高的相似度,其液相色谱指纹图谱可反映鹿角胶所含的18种氨基酸成分。结论:该方法稳定可靠,可为鹿角胶中氨基酸成分质量的全面控制提供依据。

    Abstract:

    Objective: To establish a method of fingerprint analysis of the amino acids on Cervil Cornus Colla by HPLC. Method: Cervil Cornus Colla was derivated by Phenyl isothiocyanate (PITC). The Chromatographic fingerprints were obtained by the Diamonsil C18 (2) column (4.6 mm×250 mm, 5 μm) with the gradient elution solvent system composed of acetonitrie-natrium aceticum buffer solution (pH 6.5). The detective wavelength was set at 254 nm, the flow rate was 1 mL·min-1 and the column temperature was maintained at 43 ℃. Result: The result showed that the amino acids constituents had a high similarity. 18 kinds of amino acids in Cervil Cornus Colla were detected, of which 13 peaks were identified in common peaks. Conclusion: The method has good reproducibility and stability, and could be used for the research and the quality control of Cervil Cornus Colla.
