    摘要:

    目的: 优选治郁颗粒的醇提工艺和水提工艺,为该制剂的工业化生产提供参考. 方法: 采用HPLC测定栀子苷含量,流动相乙腈-水(10:90),检测波长238 nm;以栀子苷提取率为指标,通过正交试验考察乙醇体积分数、加醇量、提取时间及提取次数对醇提工艺的影响.采用UV测定总多糖含量,检测波长490 nm;以总多糖提取率为指标,通过正交试验考察加水量、提取时间及提取次数对水提工艺的影响. 结果: 最佳提取工艺为加6倍量70%乙醇提取2次,每次1.0 h,药渣加10倍量水煎煮3次,每次2.0 h;栀子苷、总多糖提取率分别为0.013 29,0.030 39 g·g-1. 结论: 该优选工艺合理可行,有效成分提取率高.

    Abstract:

    Objective: To optimize alcohol and water extraction processes of Zhiyu granules,in order to provide a reference for commercial production of this preparation. Method: HPLC was employed to determine the content of geniposide with mobile phase of acetonitrile-water(10: 90) and detection wavelength at 238 nm;taking yield of geniposide as index,orthogonal test was adopted to optimize alcohol extraction process with extracting time,extracting times,ethanol concentration and amount as factors.The content of total polysaccharides by UV,detection wavelength was set at 490 nm;orthogonal test was used to optimize water extraction process by taking yield of total polysaccharides as index,the amount of water,extraction time and times as factors. Result: Optimum extraction technology of Zhiyu granules was as following:extracted twice with 6 times the amount of 70% ethanol for 1.0 h of per time,then medicine residue extracted thrice with 10 times the amount of water for 2.0 hours of each time;yields of geniposide and total polysaccharides were 0.013 29 and 0.030 39 g· g-1. Conclusion: This optimized process is reliable and feasible with high extraction rate of effective components.
