    目的: 观察预防性给予血塞通(三七总皂苷)和金纳多(银杏叶提取物)注射液对大鼠脑缺血再灌注损伤的效果. 方法: SD雄性大鼠130只,分两部分进行实验,第一部分为低剂量(25 mg·kg-1)比较,设假手术组、模型组、血塞通组、金纳多组;第二部分为高剂量(50 mg·kg-1)比较,也分别设假手术组、模型组、血塞通组、金纳多组.通过研究脑梗死体积、行为学评分、死亡率、血清超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽-S转移酶(GSH-S-transferase)、一氧化氮(NO)、一氧化氮合酶(NOS)、C-反应蛋白(CRP)、黄嘌呤氧化酶(XOD)进行评价. 结果: 血塞通低剂量对4,24 h行为学评分,死亡率,SOD,NO,NOS无明显影响,但能显著降低脑梗死体积(P<0.05)、体重下降(P<0.05)、MDA含量(P<0.01);高剂量时,对4 h行为学评分,死亡率,SOD,MDA,NO,NOS,XOD,CRP无明显影响,但能显著降低24 h行为学评分(P<0.05)、脑梗死体积(P<0.05),GSH(P<0.01)含量.金纳多低剂量时,对4,24 h行为学评分、死亡率、MDA,SOD,GSH,NOS无明显影响,但能显著降低脑梗死体积(P<0.01),NO含量(P<0.05);高剂量时,对 MDA,SOD,GSH,NO,CRP无明显影响,但能显著降低脑梗死体积(P<0.05),4,24 h行为学评分(P<0.05),NOS(P<0.01),XOD活性(P<0.01). 结论: 高剂量的血塞通和金纳多对大鼠脑缺血再灌损伤治疗效果比低剂量可能更有效,相同剂量时,血塞通与金纳多疗效并无统计学差异.

    Abstract:

    Objective: To observe the preventive effects of Xuesaitong and Ginaton injection on cerebral ischemia reperfusion(I/R) injury in rats. Method: Hundred and thirty SD male rats were divided into low dosage (25 mg· kg-1) and high dosage (50 mg· kg-1), of Xuesaitong and Ginaton injection, sham-operation and model groups.The volume of cerebral infarction, behavioral score, weight, mortality, superoxide dismutase(SOD), malondialdehyde(MDA), glutathione-S-transferase(GSH-S-transferase), nitrogen oxide(NO), NO synthase(NOS), xanthine oxidase(XOD), C-reactive proein(CRP) were determined to evaluate preventive effect. Result: Low Xuesaitong dosage had no effect on volume of cerebral infarction,behavioral score at 4, 24 h, mortality, SOD, GSH, NO and NOS, but it could reduce MDA content significantly. High Xuesaitong dosage had no effect on behavioral score at 4 h, mortality, SOD, MDA, NO, NOS, CRP, but it could reduce behavioral score at 24 h, volume of cerebral infarction, GSH. The low Ginaton dosage had no effect on weight,behavioral score at 4,24 h, mortality, MDA, SOD, GSH, NOS, but it could reduce volume of cerebral infarction, NO content significantly;the high dosage had no effect on MDA, SOD, GSH, NO, CRP. It could reduce behavioral score at 4, 24 h, volume of cerebral infarction, NOS and XOD activities significantly. Conclusion: The high dose Xuesaitong and Ginaton might be more effective than low dosage in reducing cerebral ischemia reperfusion injury. No significant difference was observed between Xuesaitong and Ginaton in a same dosage.
