    目的: 观察祛痰活血汤对非酒精性脂肪性肝病(nonalcoholic fatty liver disease,NAFLD)模型大鼠肝细胞水通道蛋白9(aquaporin 9,AQP9)表达的影响,探讨祛痰活血汤治疗非酒精性脂肪性肝病的机制. 方法: 选取普通清洁级雄性SD大鼠60只,随机分5组,每组12只,分别为:空白对照组、模型对照组、祛痰活血汤低、中、高剂量组(6,12,16 g·kg-1),高脂饲料喂养10周建立NAFLD模型,造模成功后,ig给药,每日2次,连续2周.采用HE染色法、酶联免疫吸附试验(Elisa)法及反转录PCR(RT-PCR)技术,观察各组大鼠肝脏病理变化、总胆固醇(TC)、甘油三酯(TG)水平及肝细胞中水通道蛋白9(AQP9)的表达. 结果: ①基因及蛋白水平,NAFLD模型组大鼠肝细胞中AQP9的表达均较空白对照组上调(P<0.05).祛痰活血汤3个剂量组大鼠肝细胞AQP9表达均下调,其中12 g·kg-1组AQP9 mRNA表达下调较其他两组明显(P<0.05).②祛痰活血汤3个剂量组大鼠TC,TG水平均较模型组下降(P<0.05),其中16 g·kg-1组TC,TG水平下降较其他两组明显(P<0.05).③3个剂量祛痰活血汤治疗组大鼠肝脏脂肪变性均较NAFLD模型组减轻,16 g·kg-1组脂肪变性较其他两组减轻明显. 结论: 祛痰活血汤可调节NAFLD大鼠肝细胞中AQP9的表达,并显著降低NAFLD大鼠TC,TG水平,改善肝脏脂肪变性程度.

    Abstract:

    Objective: To observe the effect of Qutan Huoxue decocotion (QTHXD) on the expression of aquaporin 9 (AQP9) in the nonalcoholic fatty liver rats, and discuss the mechanism. Method: Sixty SD rats were randomly divided into 5 groups,12 each group:the blank control group, model control group,QTHXD low, medium and high dose groups(6,12,16 g· kg-1),all rats except balnk congtrol group were fed high fat diet for 10 weeks to establish nonalcoholic fatty liver disease(NAFLD) model. After modelingg, the corresponding drugs were ig giventwice a day, for 2 weeks.HE,ELISA and RT-PCR were used to observe the liver pathology changes, total cholesterol(TC),triglycerides(TG) and the AQP9 mRNA expression in liver cells. Result: ① AQP9 expression in liver cells of NAFLD model group compared with blank control group was raised., but decreased in three QTHXD dose 12 g· kg-1group was lower than the other two groups.②TC and TG in three QTHXD dose groups deacreased, 16 g· kg-1group was lower than the other two groups.③Hepatic steatosis in the QTHXD different dose groups was reduced compared with NAFLD model group, and hepatic steatosis in 16 g· kg-1 group reduced more markedly than the other two groups. Conclusion: QTHXD could regulate AQP9 expression of liver cells in NAFLD model rats,reduce TC and TG and improve the degree of hepatic steatosis.
