    目的: 优选酸枣仁总皂苷的提取工艺,为该成分的工业化生产提供参考。方法: 采用HPLC-UV检测酸枣仁皂苷A和B的含量,流动相乙腈-水(34:66),检测波长204 nm。在单因素试验基础上,通过正交试验考察乙醇体积分数、乙醇用量、提取时间及提取次数对酸枣仁总皂苷提取工艺的影响。结果: 酸枣仁皂苷A,B的线性范围分别为0.24~2.40,0.12~1.20 μg,平均加样回收率分别为100.39%,99.40%。酸枣仁总皂苷最佳提取工艺为加12倍量70%乙醇回流提取2次,每次1.5 h;酸枣仁皂苷A,B的平均质量分数分别为0.606,0.228 mg·g-1结论: 建立的HPLC检测酸枣仁皂苷A,B含量方法准确可靠,优选的酸枣仁总皂苷提取工艺稳定可行,具有节约成本、省时等特点,适用于工业化生产。

    Abstract:

    Objective: To optimize extraction technology of total saponins from Ziziphi Spinosae Semen and provide a reference for industrial production of this component. Method: HPLC-UV method was employed to detect contents of jujuboside A and jujuboside B with mobile phase of acetonitrile-water(34: 66) and detection wavelength at 204 nm.On the basis of single factor tests,effects of ethanol concentration,liquid-sodid ratio,extraction time and extraction times on extraction process were investigated by orthogonal test. Result: Linear ranges of jujuboside A and jujuboside B were 0.24-2.40,0.12-1.20 μg,average recovery of them were 100.39% and 99.40%.The best extraction technology of total saponins from Ziziphi Spinosae Semen was as following:extracted 2 times with 12 times the amount of 70% ethanol for 1.5 h per time;average contents of jujuboside A and jujuboside B were 0.606,0.228 mg·g-1,respectively. Conclusion: This established HPLC method is accurate and reliable to detect contents of jujuboside A and jujuboside B.At the same time,this optimized extraction technology of total saponins from Ziziphi Spinosae Semen is stable and feasible with advantages of energy-saving,time-saving and so on,so it is suitable for industrial production.
