Objective: HPLC was adopted to determine the baicalin content of the Scutellariae Radix and Scutellariae Radix powder by using irradiation sterilization, together with herbs without the sterilization. Compared the content changes before and after mirradiation sterilization. Method: Accurasil-C18 chromatographic column(4.6 mm×250 mm,5 μm), methanol-water-phosphoric acid (47: 53: 0.2) as mobile phase, the wavelength detection was 280 nm with flow rate of 1.0 mL·min-1, column temperature 30 ℃, and the injection volume of 10 μL. Result: Baicalin had good linear relationship in the range of 0.163 2-0.816 0 μg. The average recovery was 99.44% with the RSD of 1.6%. Content baicalin in Radix had significant changes before and after sterilization. Conclusion: It indicates that Scutellariae Radix is not suitable for applications of irradiation sterilization,which takes baicalin as index.
