Abstract:Objective:To analyze the composing experience of professor YAN Zheng-hua for treating insomnia by traditional Chinese medicine(TCM)inheritance support system. Method: The prescriptions for insomnia by YAN Zheng-hua were collected,then the data were entered into TCM inheritance support system and analyzed the composing principles by using datamining methods such as revised mutual information,complex system entropy cluster and unsupervised hierarchical cluster. Result: Based on the analysis of 170 prescriptions, the frequency of each herb and association rules among the herbs were computed, 44 core combinations and 7 new prescriptions were mined from the database. Conclusion: Professor YAN Zheng-hua is good at nourishing the heart to calm the mind and tranquillizatting with heavy to relieve insomnia,TCM inheritance support system can be used to analyze clinical experiences of senior TCM doctors.

Abstract:Objective:To conduct a review to explore the efficacy of Shengmai injection in treating ischemic stroke. Method: All randomized controlled trails (RCTs) with regard to the effectiveness of the Shengmai injection for ischemic stroke were collected. The trails were analyzed and evaluated strictly by means of meta-analysis. The relative risk (RR) for effective rate of the Shengmai Injection for ischemic stroke was calculated. Result: Totally 12 RCTs including 960 cases were analyzed. The cumulative cases employed Shengmai Injection for ischemic stroke were 493, in which 444 cases were effective,while in control groups the cases treated without Shengmai injection was 467, in which 351 were effective. The pooled RR was 1.20, with a 95% confidence interval of 1.13 to 1.27. Conclusion: Shengmai injection is effective for ischemic stroke and better than the control treatment, and it can improve the defects in nervous function.

Abstract:Objective:Through screening and analyzing of clinical research literature on vascular cagnitive impairment(VCI), objectively evaluate the efficacy and the safety of the therapy of invigorating kidney, removing phlegm and blood stasis, to provide clinical evidence of evidence-based medicine. Method: With the method of cochrane systematic review, searching randomized controlled trial(RCT) reports of PubMed, Cochrane, CBM, CNKI, VIP, Wanfang (2001-2011), collecting the researches met inclusion criteria by two independent evaluators, analyzing data using Revman 5.1. Result: According to the inclusion criteria, screened out 18 articles of RCT qualified reports from 569 of Chinese medicine treatment of VCI, of which 18 reports had relative high quality. The results showed that: ①The therapy of invigorating kidney and removing phlegm and blood stasis had a better efficacy than the control group by mini-mental state examination(MMSE),BBS(P<0.05);②There was no significant difference shown by ADL;③ There was no statistical significant difference in safety, indicating that the two groups of incidence of adverse reaction no significant differences by Chi-square test. Conclusion: Available evidences manifests: the therapy has a better efficacy and no significant differences in safety. Due to defects of experimental design and selection of the outcome indexes, should have the process of syndrome differentiation of traditional Chinese medicine(TCM) and make and use unified syndrome curative effect evaluation standard according to syndrome differentiation of TCM, and need to push in more the rational design, the execution strictly, the multi-central and large sample, long time follow-up and has Chinese medicine characteristics double-blind RCT.

Abstract:Objective:To evaluate clinical efficacy of Kangfuxin liquid in the treament of hand-foot-mouth disease. Method: Literatures were retrieved from Cochrane library,PubMed,CNKI,VIP,WanFang.and Meta-analysis was performed. Result: Fourteen RCTs and 1 792 patients were included.The meta-analysis showed that Kangfuxin liquid-treated group was better than control group in terms of defervescence time, oral herpes or ulcer improvement time,the skin rash regression time and the total clinical efficacy.Serious adverse drug reactions(ADRs) induced by Kangfuxin liquid hadn't been reported. Conclusion: Actual clinical evidence indicated that efficacy and safety of Kangfuxin liquid in the treament of Hand-Foot-Mouth Disease.The reliability of systematic review is affected by low quality of included study.More randomized controlled trial of high-quality large-scale multiple-centered should be carried out.

Abstract:Objective:To study on dynamic change rule of chemical components from Aconitum carmichaelii during decoction process after and before compatibility with Glycyrrhiza uralensis,in order to determine optimum decoction conditions of compatibility of G.uralensis and A. carmichaelii. Method: To determine optimum decoction conditions by determining the contents of total alkaloids and 3 kinds of double ester alkaloid in different decoction time,which were from single decoction liquid of A. carmichaelii and mixed decoction liquid of G.uralensis and A.carmichaelii.And rats cardiac toxicity of soup with optimum boiling technology was investigated. Result: Optimum boiling technology of G. uralensis-A. carmichaelii was:after boiling with a blaze,kept boiling 30 min with small fire.During 0-90 min decoction process,it showed different change trend of the contents of total alkaloids and 3 kinds of double ester alkaloid before and after compatibility of G. uralensis and A. carmichaelii,it reached to both comprehensive peak value in 30 min;both single decoction liquid of A. Carmichaelii and mixed decoction liquid of G. uralensis and A. carmichaelii showed some cardiac toxicity on rats in boiling 30 min,but cardiac toxicity of the latter was relative less. Conclusion: Optimized compatibility boiling technology could provide reference for 'reducing toxicity and storing efficacy’of A. carmichaelii in clinical application and traditional Chinese medicine compound.

Abstract:Objective:To optimize matrix formulation of Sanhuang Sanyu cataplasm. Method: Taking appearance and adhesion of cataplasm as evaluation indexes,matrix formulation of Sanhuang Sanyu cataplasm was optimized by single factor test and orthogonal test. Result: Optimized matrix formulation of Sanhuang Sanyu cataplasm was NP-700-K90-glycerol(0.5∶0.4∶6).0.02 g·mL-1 solution was received by adding carbomer-u10NF into mixed extraction concentrated liquid of Rheum palmatum and Scutellaria baicalensis,swelling overnight, as A phase,0.5 g sodium polyacrylate and 0.4 g PVP were added to 6 g glycerol as B phase,aluminum chloride and citric acid were added to water as C phase,single extraction concentrated liquid of Phellodendron chinense was as D phase,four phases was fully mixed with A-B-C-D(10∶3∶3∶5), stirred 15 min, coating,covered with anti-sticking layer. Conclusion: Optimized matrix formulation of Sanhuang Sanyu cataplasm was simple,stable and feasible,which could be applied to industrial production.

Abstract:Objective:To investigate attenuating toxicity mechanism of processing for Arisaema erubescens. Method: Toxicity of raw products,processed products,raphides and different extraction site of A. erubescens were compared by acute toxicity test in mice and rabbit conjunctival irritation test;Change condition of special calcium oxalate raphides and impact of related ion from alum on morphological change of raphides before and after processing of A. erubescens were observed by scanning electron microscopy. Result: Acute toxicity test results showed that LD50 of raw products powder group,processed products powder group,raphides group of A. erubescens were 1062,2788,42.53 mg·kg-1.Results of rabbit conjunctival irritation test showed that raw powder group and raphides group had low and moderate irritation,but other various extracts had no irritation.Electron microscopy showed that ultrastructure of raphides occurred significantly change before and after processing of A. erubescens,and it was destroyed after soaked in KAl (SO4)2·12H2O or AlCl3 solution for 12 h,but no big change after soaked in Na2SO4 or KCl solution,the tip still existed. Conclusion: Raphides from A. erubescens was main material foundation of irritating toxicity,irritation toxicity of raphides from A. erubescens was related with its morphology, mechanism of attenuating toxicity of processed A. erubescens by alum was that Al3+ may accelerate destruction of calcium oxalate raphides morphology.

Abstract:Objective:To determine dissolution of evodiamine dispersible tablets and evodiamine which were processed by different methods. Method: According to the second dissolution method(slurry method) in 2010 edition 'Chinese Pharmacopoeia',1%SDS solution of 900 mL was used as dissolution medium,rotation speed was 100 r·min-1,HPLC was as quantitative method to determine dissolution,evodiamine-poloxamer 188 solid dispersions,evodiamine ultrafine powder and evodiamine ultrafine powder-poloxamer 188 solid dispersions were prepared. Result: Ddissolution of evodiamine dispersible tablets was more than 60% at 45 min;Dissolution of evodiamine ultrafine-poloxamer 188 solid dispersions was higher than others. Conclusion: This established dissolution determination method of evodiamine dispersible tablets was simple and rapid,which could be as dissolution detection method of evodiamine dispersible tablets;Evodiamine was almost insoluble in water,but ultrafine grinding combined with solid dispersion technology could significantly improve dissolution of it.

Abstract:Objective:To study on elution property of pigment contained in fructo-oligofructose from Morinda officinalis suitable by resin adsorption. Method: With decolorization rate and retention rate of oligosaccharide as indexes, suitable resin was chosen,and static and dynamic adsorption tests were carried out to investigate decolorization effect of fructo-oligofructose from M. officinalis by single factor test.The content of nystose was determined by HPLC-ELSD. Result: D941 resin was chosen,optimum technology conditions were as follows:room temperature(25 ℃),oligosaccharide solution 4 BV(pH 5.88,15 g·L-1), flow rate 4 BV·h-1,diameter high ratio 1∶10.Under these conditions, decolorization rate and retention rate of oligosaccharide were 92.54%,82.76%,respectively,and the content of nystose increased from 22.46% to 29.94%. Conclusion: D941 resin had good decolorization effect on fructo-oligofructose from M. officinalis.

Abstract:Objective:To optimize matrix prescription of Guyanxiao cataplasm. Method: Matrix prescription was optimized by orthogonal test with initial viscosity,sustained adhesive strength and general sensory as comprehensive evaluation index,the amount of NP700,CMC-Na, glycerol and titania were chosen as factors. Result: Optimal matrix prescription was:NP-700, CMC-Na, glycerol and titania of 3,0.6,16,0.8g,respectively. Conclusion: Guyanxiao catapasm with good appearance and adhesion could be prepared by optimized matrix formulation.

Abstract:Objective:To investigate purification effect of different types of macroporous resin on effective constituents from Fagopyrum dibotrys. Method: With the content of epicatechin and mixture of proanthocyanidins tannic condensation,extract rate as indexes,optimum resin was selected by combining with HPLC fingerprint. Result: Elution rate of epicatechin by AB-8 resin was 89.78%,elution rate of mixture of proanthocyanidins tannic condensation by AB-8 resin was 93.04%.Comparison of the common peak area of HPLC fingerprint showed that AB-8 resin was the most suitable one to purify effective constituents from F. dibotrys. Conclusion: This process of applying AB-8 resin to purity effective constituents from F. dibotrys was feasible, simple and stable and could be extended to large-scale production applications.

Abstract:Objective:To optimize flocculation clarification technology conditions of Yilingjing oral liquid by chitosan. Method: Taking absorbance and retention rate of stilbene glucoside as evaluation indicators,influence of the mass concentration of extraction liquid,chitosan dosage,pH,temperature,stirring speed,stirring time and other factors on flocculation effect were investigated.Based on single factor test,orthogonal design was selected to optimize flocculation technology with the mass concentration of extraction liquid,chitosan dosage,flocculation temperature as factors.Flocculation clarification method and alcohol sedimentation were compared. Result: Optimal clarification procedure condition was that the concentration of materials-extraction solution 0.25 g·mL-1, the adding dosage of chitosan of 1.2 mL·g-1, flocculation temperature of 60 ℃, pH of 5.0-5.5, stirring speed of 300 r·min-1 and stirring time of 10 min. Conclusion: Chitosan flocculation method could be used as clarification technology of Yilingjing oral liquid,which could substitute ethanol precipitation method.

Result: Optimum preparation technology was:7% emulsifier,stirring speed 600 r·min-1,volume fraction of glutaraldehyde 10 mLand 25% cyclohexane,drug loading 6.23%;Average particle size of SM-PVA microcapsules 16 μm,it showed uniform dispersion and smooth surface of spherical.These prepared SM-PVA microcapsules showed good sustained release in simulated gastric and intestinal fluid. Conclusion: SM-PVA microcapsules could be a novel drug delivery system of sinomenine hydrochloride from Chinese materia medica.

Abstract:Objective:To optimize extraction technology of lovastatin from Monascus purpureus. Method: With composite score of yield of dry extract and transfer rate of lovastatin as index,taking extraction times, extraction time, the concentration and amount of ethanol as factors, extraction technology of lovastatin from M. purpureus was optimized by L9 (34) orthogonal test. Result: Optimum extraction technology was as follows:reflux extracted 2 times with 6 times the amount of 95% ethanol,0.5 h per time. Conclusion: This optimized extraction technology was feasible, stable and reasonable with high yield of lovastatin.

Abstract:Objective:To study effect of compatibility of traditional Chinese medicine pair on decocting rate of main active ingredient. Method: With pair of Scutellaria baicalensis and Coptis chinensis as study object,the content of baicalin and berberine hydrochloride as indexes,which were determined by HPLC,the amount of water,extraction times,extraction time and ratio of S. baicalensis and C. chinensis were chosen as factors,boiling technology was optimized by mixing uniformity design test.Difference of chemical composition between total decoction(with different proportion of S. baicalensis and C. chinensis) and mixed liquid of single decoction of them was investigated,and two target ingredients were visual contrasted by three dimensional response surface and triangle energy level diagram,dynamic change phenomenon of chemical composition was focused on,which due to mixed decoction of S. baicalensis and C. chinensis. Result: The decocting content of main active ingredient from C. chinensis showed significantly correlation with compatibility proportion of S. baicalensis and C. chinensis;But the decocting content of active ingredient from S. baicalensis was concerned with decoction influencing factors.The contents of two index component were lower with other proportions of S. baicalensis and C. chinensis,except the amount of C. chinensis was 2,3,5 times for S. baicalensis. Conclusion: Clinical efficacy of pair-Chinese materia medica could be revealed by study on dynamical change relationship of different compatibility proportions and their chemical composition.

Abstract:Objective:To investigate effect of gel material ratio on in vitro percutaneous absorption of basil oil transdermal gel. Method: With CMC-Na and PVA as accessories to prepared basil oil transdermal gel.Modified Franz diffusion cell was used,percutaneous penetration test was isolated with mouse dorsal skin as barrier,with the content eugenol as index,percutaneous permeation quantity of basil oil was detected by gas chromatography. Result: Ratio of polyvinyl alcohol and sodium carboxymethyl cellulose showed no significant effect on transdermal ability of basil oil,test determined 2∶4 was the best ratio;But their concentration in gel matrix had a certain influence on transdermal ability of basil oil,the content of them with 25% was optimal. Conclusion: These results suggested that basil oil transdermal gel could greatly improve efficacy of basil oil.

Abstract:Objective:To optimize extraction and purification technology of total triterpene acid from Hedyotis diffusa. Method: With extraction rate of total triterpene acid as index,the amount and concentration of ethanol,reflux time were chosen as factors,extraction technology of total triterpene acid from H. diffusa was optimized by orthogonal test;Resin model was screened by static adsorption and desorption test,purification technology of total triterpene acid was investigated by single factor test. Result: Optimum extraction technology was:reflux extracted 1.5 h with 10-fold 70% ethanol.Optimum purification technology of AB-8 macroporous resin was:sample flow rate 2 BV·h-1,the concentration of sample liquid 0.25 g·L-1, pH 5,the concentration of elution was 80% ethanol,eluting velocity 2 BV·h-1 with pH 7. Conclusion: Optimized technology was simple,stable with good reproduciblity,which was suitable for industrial production of total triterpene acid.

Abstract:Objective:To optimize mixed proportion for prescription of oily acne herbs through response surface method. Method: Bacteriostasis experiment, hyaluronic acid and inhibiting thrombin experiment in vitro and inhibiting human sebum secretion experiment were carried out. Based on the results of these experiments, the indicators were analyzed by response surface method to find the best ratio finally. Result: Simulation and prediction of the best ratios of the herb extracts showed dandelion extract (1.19), Magnolia bark extract (2.19) and ligusticumchuanxiong extract (2.24). The prediction estimated a relative-inhibiting compound fat rate of 38.9%. Because all three kinds of Chinese traditional medicine extract is 0.2 g·mL-1, extracts can be expressed as a proportion of their own quality of Chinese herbal medicines. The real experimental value of the relative-inhibiting compound fat rate was 40.25%. Conclusion: The predicted value of the relative-inhibiting compound fat rate though response surface method is very similar to the experimental value. Response surface method can be employed in the proportion study of compound prescriptions.

Abstract:Objective:To study chemical constituents from Isatis indigotica. Method: Chemical constituents were isolated by many chromatographic separation method,and structure of these were identified by purified by1H,13C-NMR and other spectroscopy technology and physicochemical data. Result: Six compounds were isolated and identified as n-heneicosanoic acid, β-sitosterol, indirubin, adenosine, emodin, Fructopyrano-(1→4)-glucopyranose. Conclusion: N-heneicosanoic acid and Fructopyrano-(1→4)-glucopyranose were first isolated from I. indigotica.

Abstract:Objective:To establish a method of fingerprint analysis on Yiqing soft capsule by HPLC and provide a reference for inner quality evaluation of Yiqing soft capsule. Method: The separation was performed on Xtimate-C18 chromatographic column with mobile phase consisted of water (contains 0.2% phosphoric acid)-acetonitrile (gradient elution) at a flow rate of 0.6 mL·min-1. Detecting wavelength was set at 280 nm and the column temperature was set at 30 ℃. About fifteen samples of Yiqing soft capsule from five manufacturers were determined and the similarity of each chromatogram was analyzed. The adscription of each peak was by comparing nine standard sample and three single herb preparation with Yiqing soft capsule. Result: HPLC-fingerprint of Yiqing soft capsule was established, which contained 18 common peaks, and eight peaks in the fingerprint were identified. The different batch samples made by the same producer had good similarity, but there were great differences among different manufacturers. There was great variability in the content of anthraquinones in individual crude drugs Rhubarb from different manufacturers and batches, but the main component in coptis root and scutellaria is relatively stable. Conclusion: The method with good reproducibility is accurate and reliable, it can be used in quality control of Yiqing soft capsule.

Abstract:Objective:RP-HPLC was used to establish a method of fingerprint analysis on Folium Isatidis Pieces. This method can provide a reliable basis for the quality control. Method: Chromatographic column: Inertsil ODS-3 (4.6 mm×250 mm, 5 μm); mobile phase: ACN-0.1%formic acid, gradient elution; flow rate: 1.0 mL·min-1; detection wavelength: 254 nm; column temperature: 30 ℃. Result: The normal characteristic fingerprint of Folium Isatidis Pieces was established. The result showed that 32 peaks were characteristic including four common peaks and one partially detected peak. Conclusion: With high specificity, precision and good repeatability, the established method provides an effective means for overall quality control of Folium Isatidis Pieces.

Abstract:Objective:A rapid method was established for quantitative determination of peptide based on biuret reaction-enzyme-linked immunosorbent detector, and peptide content in the same geographical origin of pre-and-post-processed Trionycis Carapax was analyzed. Method: The developer was 0.02 g·mL-1 sodium potassium tartrate solution (50 μL), 5% NaOH (20 μL), 1% CuSO4·5H2O (50 μL). The wavelength of detection was at 580 nm. Result: The calibration curves were linear in the ranges of 0.00-5.00 g·L-1 for septa-peptide from Trionycis Carapax. Peptide content in vinegar-processed samples was 6.99%, while the same content in crud samples was 1.04%. Conclusion: Peptide content in vinegar-processed samples was significantly higher than the one in crud samples. The content of Trionycis Carapax is increased greatly after vinegar by decoction.

Abstract:Objective:To establish the analytical method for characteristic fingerprint of Scutellaria baicalensis by UPLC-MS, which can be used as a more effective method for its quality assessment. Method: Samples from 10 habitats were extracted in turn by 70% methanol and 100% methanol, and the combined solution was analyzed by UPL-ESI-TOF/MS to acquire their chromatograms. The common peaks were selected according to the retention time and the m/z value of molecular ion, and the peak area was calculated in the corresponding extracted ion chromatogram. Finally the similarity of the relative peak area was calculated by the correlation coefficient and the cosine value of vectorial angle. Result: Twenty three common peaks were selected to establish the UPLC-MS characteristic fingerprint of S. baicalensis. The samples of S. baicalensis from 10 habitats had a good similarity more than 0.9. Conclusion: The UPLC-MS characteristic fingerprint of S. baicalensis is accuracy and reproducible, and the method can be applied to the quality assessment of S. baicalensis.

Abstract:Objective:To establish a microwave-assisted extraction method for the extraction of the linarin from Chrysanthemun indicum. Comparison of the determination of linarin from C. indicum between microwave-assisted extraction and Chinese pharmacopeia method was made. Method: Dikma Diamonsil C18 column(4.6 mm×150 mm,5 μm)was used. The mobile phase cousisted of methanol-water-acetic acid (26∶23∶1).The temperature of column was kept at 30 ℃. The flow rate was 1.0 mL·min-1 and the detection wave-length was at 334 nm. Result: Microwave extractive time was 15 min,the temperature was 100 ℃,the extraction solvent was 50% ethanol. The calibration curve was in good linearity within the range from 0.04 to 1.2 μg(r=0.999 9). The average recovery was 100.13%(n=6). Conclusion: The method is simple,rapid,as well as precise and reliable,and can be used for the determination of linarin from C. indicum.

Abstract:Objective:To establish a method for the determination of paeonol in Eczema Nanoemulsion spray by HPLC. Method: Using HPLC technique,we detected the samples by the C18 ODS column of Phenomenex company under the wave length at 274 nm, the chromatographic conditions were phenomenex C18 -ODS column, mobile phase of methanol-water (55∶45), flow rate of 1.0 mL·min-1 and detection wave-length at 274 nm. Result: The standard curve was linear over the range of 10.0-100.0 mg·L-1,with the correlation coefficient of 0.999 9.It is steady in 48 hour. The average recovery was 99.70% with RSD 1.12%(n=9). The theoretical plates are above 6 000,separating degree is greater than 1.5. Conclusion: The analytical method is simple,rapid,sensitive,accurate; stability and reproducibility are good,and the method could be used to determine content of paeonol in a Chinese medicine Eczema Nanoemulsion spray.

Abstract:Objective:To discuss detection method of the qualitative and quantitative of Erigeron canawdensis. Method: TLC was used to determine quercetin of E. canawdensis. The content of quercetin was determined by HPLC. Result: n-Hexane-butyl acetate-formic (6∶5∶1) is used as TLC deployment system. It may clearly detect quercetin; Shimadzu C18 (4.6 mm×250 mm, 5 μm) column was used in HPLC with mobile phase methanol-0.4% phosphoric acid (47∶53). The detection wavelength was set at 257 nm and the flow rate was 1 mL·min-1; the column temperature was at 30 ℃.The linear range of quercetin were respectively 0.504-1.134 μg, and their average recovery were respectively 97.97%,RSD were respectively 1.2%. Conclusion: This method is used as detection method of the qualitative and quantitative of E. canawdensis.

Abstract:Objective:To establish the determination method and variation of medicinal parts of Juglans mandshurica. Method: Extract the total flavone from Qinglongyi by 75% ethanol, take the aluminum nitrate-nitrite colorimetric, in addition to the pigment by AB-8 macroporous resin and establish the determination method. Result: The content of total flavone in five different acquisition time was 3.25,3.49,3.90,3.98,3.96 mg·g-1. Conclusion: Before the canicular days,the content of total flavones was gradually increased; after the canicular days, the content of total flavones was significantly growth to flatten, the content of total flavones of medicinal parts of J. mandshurica was leaf>bark> stem.

Abstract:Objective:To establish a method of determining quercetin, kaempferide and isorhamnetin in Ganershu pellets by HPLC. Method: An Hedera ODS-2 column(4.6 mm×200 mm,5 μm)was used at 30 ℃ with the mobile phase of 0.4% phosphate (A) and methanol (B)by gradient elution.The detection wavelength was set at 360 nm and the flow rate was 1.0 mL·min-1. Result: The quercetin, kaempferide and isorhamnetin had good linear relationship within the ranges of 0.042 8-0.42 8 μg(r=0.999 8), 0.003 49-0.034 9 μg (r=0.999 1),0.041 8-0.418 μg(r=0.999 2).The average recoveries were 99.88%,99.44% and 97.50% with RSD 0.77%(n=6),1.2%(n=6),1.5%(n=6) respectively. Conclusion: The method was accurate and reproducibility was good. The method can be used for the quality control of Ganershu pellets.

Abstract:Objective:HPLC chromatographic fingerprints for Shuangshan granules were developed in order to evaluate the internal quality. Method: HPLC analysis was performed at 25 ℃ by using a Diamonsil C18 column (4.6 mm×250 mm,5 μm), with the gradient elute of mobile phase consisting of acetonitrile-0.1% glacial acetic acid at the flow rate of 1.0 mL·min-1. The UV detection was set at 340 nm. Result: Chlorogenic acid was used as the reference substance in the HPLC chromatographic fingerprint of Shuangshan Granules, which showed 13 common peaks and the similarity was 0.98 among 10 samples. Conclusion: This method was accurate, repeatable and useful for the quality control of Shuangshan granules.

Abstract:Objective:To establish a method for the determination of safrolglycol in Cinnamomum camphora. Method: Using HPLC with Hypersil ODS column, CH3CN-H2O (14∶86) as mobile phase and detection wavelength at 282 nm. Result: The linearity of the calibration curve was evaluated over the amount range of 1.014-5.07 μg for safrolglycol, and good linearity with correlation coefficient (r) of 0.999 9 was obtained. The recovery and RSD were 99.0% and 1.89% (n=6) respectively. Conclusion: This method is rapid, simple and accurate, thus may be used for the quality control of C. camphora.

Abstract:Objective:To explore quality of Gardeniae Fructus from Bazhong. Method: HPLC and ELSD were used to determine the content of geniposide, crocinⅠ,and ursolic acid. Using t-test and Z-score comprehensive evaluation on quality of Gardeniae Fructus from Bazhong. Result: Results of t- test showed that the content of Geniposide in Gardeniae Fructus from Bazhong is significantly higher than in Gardeniae Fructus from Hubei(P<0.05), outstandingly higher than in Gardeniae Fructus from Jiangxi or Jiangjin(P<0.001); the content of crocinⅠand ursolic acid in Gardeniae Fructus from Bazhong was remarkablly higher than in Gardeniae Fructus from Hubei or Jiangxi or Jiangjin(P<0.001); we established multiple linear regression equation between Z-score and ratios of geniposide, crocinⅠ,ursolic acid, geniposide X1, crocinⅠ X2 and ursolic acid X3, respectively. the multiple linear regression equation was Z=-11.213 6+0.0218 2X1+0.560 2X2+17.331 6X3(R2=0.999 9). Results of Z-score comprehensive evaluation showed that Z-score of Gardeniae Fructus from Bazhong is far ahead of Gardeniae Fructus from Hubei or Jiangxi or Jiangjin. Conclusion: Quality of Gardeniae Fructus from Bazhong is very good;Gardeniae Fructus from Bazhong is well worth focusing by government and society.

Abstract:Objective:To establish a method for the relative recovery (RR) of chlorine-e6 in microdialysis probe and to investigate the effect of the concentration,time and temperature on RR in chlorin-e6 microdialysis probe. Method: Gain method and loss method were used for the study. The concentration of chlorine-e6 in micro-dialysate samples was detected by HPLC and calculated probe RR with the formula used for the probe. Result: At the same concentration, RR decreased with the increase of the flow rate in the two methods. RR detected by gain method was similar to that detected by loss method. At the same flow rate, the concentration had no obvious effect on RR. RR kept steady within 12 hours, and increased obviously with the temperature. Conclusion: The influencing factors on RR in chlorin-e6 microdialysis probe are confirmed. The probe recovery obtained by gain method is similar to that by loss method. Retrodialysis can be used for the determination of the chlorine-e6 in-vivo recovery.

Abstract:Objective:To establish the quality control methods for Triticum aestivum. Method: The pharmacognostical methods were employed for the morphological and histological characters of Triticum aestivum. TLC was used to identify 5-heneicosyl resorcinol and the content of 5-heneicosyl resorcinol was determined by HPLC. Result: The morphological and histological characters of Triticum aestivum were observed. The spots were observed in the TLC with high characterisitic and good repeatability. Under the selected HPLC method, 5-heneicosyl resorcinol was well separated with others. The linear range of 5-heneicosyl resorcinol was 129.5-2 590 ng. Y=5 056.5X+201.5(r=0.999 9).The average recovery of 5-heneicosyl resorcinol was 101.26%, and the RSD was 1.89%. Conclusion: The established method is simple, feasible, reproducible and accurate. It is suitable for identification and evaluation of the quality, and can be used as the quality control standard of Triticum aestivum.

Abstract:Objective:To establish a method for quality control of Biyanling Pill. Method: Cocklebur fruit,Flos magnoliae liliflorae, Mentha haplocalyx and Asarum were identified by TLC, and the content of baicalin in pills was determined by HPLC with C18 column and the mobile phase consisted of methanol-water-phosphoric acid solution (47∶53∶0.2). Detection wavelength was set at 278 nm. Result: Cocklebur fruit, Flos magnoliae liliflorae, Mentha haplocalyx and Asarum could be identified by TLC. The calibration curves of baicalin was in a good linearity in the range of 0.125 6-2.198 μg (r=0.999 9). The average recovery rate was 98.05% with RSD of 1.66% (n=6). Conclusion: The method is simple, accurate and can be applied to the quality control of Biyanling Pill.

Abstract:Objective:To establish an HPLC method for determination of geniposide in Yinzhi oral solution. Method: The Agilent Eclipse XDB-C18 (4.6 mm × 150 mm, 5 μm) was used with the mobile phase consisted of methanol-water (25∶75); the flow rate was 1.0 mL·min-1, the detection wavelength was at 238 nm, and the column temperature was at 30 ℃. Result: A good linear correlation of geniposide was observed within the range of 0.010 20-0.408 0 μg. The average recovery was 99.35% with RSD 1.90% (n=6). Conclusion: The method is rapid, simple and accurate. It can be used for the determination of geniposide in Yinzhi oral solution.

Abstract:Objective:To determine the content of total flavonoids in several different sources of Gaoziban. Method: Using rutin as a reference substance, the content of total flavonoids in Gaoziban of Asperugo procumbens from Hetian, Cele, and Yutian, Anchusa italica,Borage officinales, and Cynoglossum officinales were determined at 510 nm by UV spectrophotometry colored with sodium nitrite-aluminum nitrate-sodium hydroxide. Result: Content of total flavonoids in three Gaozibans of Anchusa italica, Borage officinales, Cynoglossum officinales were 3.85, 3.72, 2.35 mg·g-1; and three Gaozibans of A. procumbens from Hetian, Cele and Yutian were 3.14, 4.73, 3.67 mg·g-1 respectively, among which the content of total flavonoids in Gaoziban was highest in A. procumbents from Cele and lowest in C. officinales. Conclusion: The method was accurate, reliable, repeatable, and could be used to determine the content of total flavonoids in Gaoziban. The content of total flavonoids is not much different from A. italica, B. officinales and A. procumbens.

Abstract:Objective:The influence of heating time and temperature on the content of ginsenosides from stems and leaves of ginseng during water decoction process was researched to calculate the term of validity. Method: The HPLC was used to determine the content of ginsenoside Rg1 and ginsenoside Re extracted in different heating conditions.An Agilent Extend-C18 column(4.6 mm×250 mm, 5 μm) was used with acetonitrile-water(19∶81)as the mobile phase at the flow rate of 1.0 mL·min-1,and the column temperature was maintained at 30 ℃ and the detection wavelength was set at 203 nm. The term of validity was caculated by classical homeothermia. Result: When the heat comes to 12 h, ginsenoside Rg1 and ginsenoside Re had different degree of degradation, and they degraded rapidly when temperature is at 100 ℃. It’s only 20 percent of the initial content at 100 ℃,and 60% at 80 ℃, 80% at 60 ℃.With simple linear regression analysis to 1/T as lgK, the regression equations are:lgK=-2 396.2/T+5.587 3 for ginsenoside Rg1, lgK=-2 518.2/T+5.901 6 for ginsenoside Re. The term of validity of ginsenoside Rg1 and ginsenoside Re were calculated as 30.1 h and 37.6 h. Conclusion: The content of ginsenoside Rg1 and ginsenoside Re decreased with time increasing and temperature increasing. The term of validity about stems and leaves of Panax ginseng is in short period.So,intermediate which contains water extract of stems and leaves of P. ginseng should accomplish the preparation within the term of validity.

Abstract:Objective:To determine the content of total aristolochic acid in different extracts of Asarum heterotropoides, and provide the basis for clinical application of A. heterotropoides. Method: A. heterotropoides was extracted with water, ethyl acetate, 30% ethanol, 50% ethanol, 70% ethanol and 95% ethanol as extracting solvent. The ultraviolet spectrophotometry was used with aristolochic acid A as the chemical reference. The UV detector wavelength was set at 390 nm. Result: The content of aristolochic acid A showed a linear relation with the spectrophotometric absorbance within the range of 3.0-30.0 mg·L-1, and the linear equation was Y=19.844X+0.027 2 (r=0.999 6). The average recovery was 98.51%, and RSD was 1.32%. The total aristolochic acid content varied from 1.34%-3.15%. It was the lowest in the water extract and highest in the 95% ethanolic extract. Conclusion: The method is reliable, accurate and suitable for determining the content of the total aristolochic acid in extracts of A. heterotropoides.

Abstract:Objective:To establish an HPLC method for determination of quercetin and kaempferol in Baihua Sheshecao dispensing granule. Method: Orthogonal experiments were used to optimize extraction technology. The samples were separated on a Hypersil ODS C18 (4.6 mm×250 mm, 5 μm) column which was eluted with methanol-0.2% phosphoric acid solution (45∶55) with detective wavelength at 360 nm and flow rate at 1 mL·min-1, column temperature at 30 ℃. Result: The method had good linear relationship within the range of 0.09-0.72 μg (r=0.999 9) for quercetin and 0.034 3-0.274 4 μg (r=0.999 9) for kaempferol. The average recovery rate were 100.80% (RSD 1.79%) and 101.75% (RSD 1.76%). Conclusion: The method is simple, rapid and reliable which can be used to determine quercetin and kaempferol in Baihuashe Shecao dispensing granule.

Abstract:Objective:Use flame atomic absorption spectrometric method for the determination of trace elements content in Dioscoreae Rhizoma. Method: Six Dioscoreae Rhizoma samples of six different sources were purchased and the mixed solution of HNO3 and HClO4 (4∶1)was used to digest the sample of Dioscoreae Rhizoma. The content of Mn, Fe, Cu, Zn, Mg, in the sample solution was determined by FAAS. Result: Dioscoreae Rhizoma medicinal materials contain trace elements, its content from high to low was Mg>Fe>Cu>Zn>Mn. The recovery rate of this method was 95.50%-103.50%. Conclusion: The method is sensitive, rapid, stable, and can be used for determination of trace elements in Dioscoreae Rhizoma.

Abstract:Objective:To establish quantification methods for determination of four types of active constituents in two sorts of Rhei Radix ex Rhizoma by HPLC, including free anthraquinone, anthraquinone glycoside, sennoside, catechin and gallic acid, in order to evaluate the quality of Rhei Radix ex Rhizoma all-around. Method: HPLC was applied on the determination of four types of active constituents with Agilent Zorbax SB-C18 column (4.6 mm×250 mm,5 μm). Mobile phaseⅠwas 0.1% phosphoric acid-methanol by gradient elution, mobile phaseⅡwas THF-water-acetic acid(2∶80∶1.5)-acetonitrile by gradient elution, and mobile phase Ⅲ was 0.5% acetic acid-methanol by gradient elution; the three kinds of flow rate were 1.0,0.8,1.0 mL·min-1; the wavelengths were set at 254,350,270 nm; the column temperature were all set at 25 ℃. Result: Determine of four types of active constituents in two different sorts of Rhei Radix ex Rhizoma. These components were separated completely within 30 minutes under this chromatographic condition. The average recoveries were from 97.09% to 103.2%, its relative standard deviations were from 0.15% to 3.0% (n=6). Conclusion: The methods with good reproducibility are simple and accurate; it can be used as quality control methods for Rhei Radix ex Rhizoma.

Abstract:Objective:Study on the chemical constituents from Cibotium barometz and their scavenging activities for 2,2-diphenyl-2-picrylhydrazyl (DPPH) radical. Method: The chemical constituent from70% alcohol extract of C. barometz was isolated on chromatograph of silica gel and sephadex LH-20 and HPLC, their structure were elucidated on the basis of spectra data and the compounds were tested for their scavenging activities on DPPH radical. Result: Ten compounds were purified and their structure were identified as 1-O-caffeyl-D-glucopyranose ( 1 ), 6-O-caffeyl-D-glucopyranose ( 2 ), 3-O-caffeyl-D-glucopyranose ( 3 ), 3-hydroxymethyl-2(5H)-furanone ( 4 ), β-miroside( 5 ), cibotiumbaroside A( 6 ), protocatechuic acid ( 7 ), glucose ( 8 ), mannose ( 9 ), Corchoionoside C ( 10 ), kojic acid ( 11 ). Three isolated compounds showed strong scavenging activities on DPPH radical. Conclusion: Compound 1-5 were isolated for the first time from C. barometz. among of them, compound 1 , compound 3 and compound 6 showed significant antioxidant activity, and the scavenging activity of compound 1 was similar to that of Vit C.

Abstract:Objective:To provide a reference of reasonable quality control standards through a comparative study of the four fingerprints of four origin plants of Curcumae Radix. Method: The separation was performed on HP-5 0.32 μm×0.32 mm×30 m column,hydrogen flame ionization detector (FID); injector temperature was 280 ℃, detector temperature was 300 ℃; the column oven was begun at 125 ℃,then programmed from 125 ℃ to 170 ℃ at 3 ℃·min-1, then programmed from 170 ℃ to 260 ℃ at 9 ℃·min-1 and final, held 0 min. Result: To analyse 20 batches of volatile oil, the fingerprints of four origin plants of Curcumae Radix were established. The fingerprint similarity value of the Curcuma longa and C.wenyujin is above 0.95; the fingerprint similarity value of the C.phaeocaulis is above 0.99; the fingerprint similarity value of the C.kwangsiensis is about 0.90. Conclusion: It displayed marked differences between four origin plants of Curcumae Radix volatile oil chemical constituents and everyone's fingerprint could describe own chemical constituents. So we provide the data support for establishing scientific quality control standards of Curcumae Radix.

Abstract:Objective:To study the character of the contents and distribution of chemical composition of Uighur Medicine Coreopsis ticntoria flowers (CTF), and to provide valuable experimental evidence for the quality evaluation and control of CTF. Method: Ten batches of samples,CTF come from different producing areas of Xinjiang were carried out to quantitate by UV and ICP.Principal component analysis and Hierarchical clustering analysis were applied to study on the chemical components were extracted from the original data. The principal component composition and chemical pattern recognition. Result: Three principal analysis results showed that Cu, Zn,Fe,Mn,Ni,Cr and Al may be the characteristic elements. The ten samples could be clustered reasonably into three groups, and the contents of chemical composition of CTF were related to the ecological environment, such as temperature and location. Conclusion: This paper, by measuring the number of content indicators, using principal component analysis and cluster analysis clearly reveals the regularity of the different origins and differences in CTF.

Abstract:Objective:To study the chemical constituents of the ethyl acetate portion of Incarvillea arguta. Method: The ethyl acetate portion were isolated and pured by silica gel column chromatography, HP-20 and Sephadex LH-20.Their chemical structures were elucidated by spectral data(1H-NMR,13C-NMR). Result: Five compounds were isolated from the ethyl acetate portion. They were identified as suberosenol A( 1 ),suberosenol B( 2 ),suberosenone( 3 ),kaempferol-7-O-α-L-rha mnoside( 4 ),ferulic acid( 5 ). Conclusion: All compounds were isolated from this plant for the first time.

Abstract:Objective:To establish a method for the determination of 5 elements (As, Pb,Cu,Cd and Hg) in different pocessed products of pyrite by inductively coupled plasma atomicemission spectrometry/mass spectrometry(ICP-AES/MS), in order to provide a basis for thelimit standards of As, Pb,Cu, Cd and Hg and other harmful elements. Method: Five elements of the digested samples were determined by ICP-AES/MS simultaneously, and the contents were calculated by calibration curves. Result: Good linearity of calibration curves was obtained for ICP-AES/MS assay, the accuracy and the recovery met the requirement of trace analysis. Conclusion: The method is highly sensitive,rapid,accuracy and offered with low LOD.

Abstract:Objective:Using chlorzoxazone (CZX) as the 'probe drug’,to establish a rapid and accurate method for evaluating the activities of cytochrome P4502E1(CYP2E1) by the high-performance liquid chromatography-UV detection. Method: In vivo study: 20 mg·kg-1 of CZX was given to the rats via the tail vein. Blood samples were collected at designed time points post dose and plasma samples were obtained. CZX and 6-hydroxy chlorzoxazone (6-OHCZX) were determined by an HPLC method. In vitro study:After incubation, the formation rate of 6-OHCZX was used to evaluate the function of CYP2E1.The analysis was performed on the Agilent Extend-C18 column (4.6 mm×100 mm, 5 μm), with a mobile phase consisting of acetonitrile-0.6% acetic acid in water at a flow rate of 1.0 mL·min-1. Phenacetin was used as the internal standard. The detection wavelength was 290 nm, and column temperature was set at 30 ℃. Result: In vivo and in vitro experiments revealed that the methods had good linear ranges and their intra-and inter-day relative standard deviations, relative recoveries, stabilities met the requirement of biological estimation. Conclusion: Other endogenous substances in the plasma and incubation system did not interfere with the determination. The method is rapid, stable and highly sensitive. It is suitable for determination of CZX and its metabolites 6-OHCZX and can be used for in vivo and in vitro evaluation of CYP2E1 activitise and research of enzyme kinetics.

Abstract:Objective:To determine the oil/water partition coefficient of homoplantaginin and to investigate its absorption characteristics in various intestinal segments in rats. Method: A HPLC(shake flask method) was established to determine n-octanol/water partition coefficient of homoplantaginin in n-octanol(water and n-octanol) different pH solution; the intestinal absorption characteristics were detected by the in situ perfused rat intestinal model, the concentration of phenol red was determined by UV method while those of homoplantaginin was analysed by HPLC method. Result: The allocation ratio of homoplantaginin in octanol/water system was 18.07 (lgP=1.26) and the PH of buffer solution had no influence on its distribution coefficient. The Ka and Peff of homoplantaginin showed jejunum>duodenum>ileum>colon in four regions of intestine. The values of Ka and Peff of different drug concentrations had no significant influence, furthermore, the values of Ka and Peff of different intestines showed to be jejunum>duodenum>ileum>colon. Conclusion: Homoplantaginin can be absorbed in whole intestinal segment, the absorption mechanism may be related to passive diffusion.

Abstract:Objective:To study the protective effects of acanthoic acid (AA) against acute liver injury. Method: Sixty mice were andomly divided into six groups: the normal group, the ethanol model group, silymarin group (100 mg·kg-1), high-dose AA group (30 mg·kg-1),middle-dose AA group (10 mg·kg-1) and low-dose AA group (5 mg·kg-1). Each group received medicine by gavage every 12 hrs for a total of 3 doses.Except for normal group, the other mice were received 60% ethanol (5 g·kg-1) 1 h. After administrayion. The thehepatic pathological and biochemical index changes in rats were observed. Result: The high-dose AA group and the middle-dose AA group can reduce the level of ALT(25±6),(31±8) U·L-1, AST(157±23),(187±18) U·L-1,TNF-α(32±4.62),(41±5.3) μg·L-1 and TG(190±23),(257±23) mmol·L-1, which was compared with the model group, and showed a significant difference(P<0.001). Conclusion: The AA could protect mice from ethanol induced acute liver injury efficiently.

Abstract:Objective:To investigate the protective effect of Wuzi Yanzong decoction on experimental cryptorchidism mice. Method: Five-six weeks old BALB/c mice cryptorchidism was induced and divied into four groups as sham, model, low dose of Wuzi Yanzong decoction (100 mg·kg-1), high dose of Wuzi Yanzong decoction (200 mg·kg-1). At the end of the study, testis tissues were collected for detection of testes weight, sperm density,live rate, histological structure changes of testis, detection intestes glutathione peroxidase(GSH-Px), superoxide dismutase(SOD) activity and the malonyldialdehyde(MDA) content changes. Result: The Wuzi Yanzong decoction treated mice in the cryptorchid group significantly increased the mice testis weight (P<0.01), sperm density (P<0.05), sperm live rate (P<0.01) and showed an improved histologic appearance. Wuzi Yanzong decoction treatment significantly increased the reduced SOD and GSH-Px enzyme activities (P<0.05 or P<0.01) and decreased the elevated tissue malondialdehyde (MDA) levels (P<0.01) in testis tissues samples. Conclusion: Wuzi Yanzong decoction is a potential beneficial agent to reduce testicular damage in cryptorchid mice by decreasing oxidative stress.

Abstract:Objective:To observe the effect of the Yiqi Huoxue recipe on the expression of endothelin(ET-1)/interferon-γ(γ-IFN) in radiation-induced lung injury model rats. Method: 66 Wister rats were randomly divided into irradion plus Chinese herbs group (Yiqi Huoxue Recipe group),the purely irrdiated group(model Group),normal contorl group. From the exposure day, the Yiqi Huoxue Recipe group received ig Yiqi Huoxue Recipe 10 g·kg-1, the model Group and the contorl group received ig distilled water 10 g·kg-1 everyday,utill the rats were killed. At the 4th , 6th, 8th, 12th, 26th weekend after irradiation, six rats were randomly selected from the Yiqi Huoxue Recipe group and model group. At the same time, six rats in normal group were executed at the 8th weekend. After executing the rats, the serum and right lung tissue were collected for examination. Result: The expression and serum levels of the ET-1 in Yiqi Huoxue Recipe group were lower than those in model Group(P<0.05). γ-IFN began to decrease at the 4th week, and drop at the 12th week and rose again by the 26th week. The expression and serum levels of γ-IFN in Yiqi Huoxue Recipe group were higher than those in model group(P<0.05). Conclusion: Yiqi Huoxue Recipe can regulate and control the expression and secretion of ET-1,γ-IFN, and has good effect on the prevention of radiation-induced lung injury.

Abstract:Objective:A new method was established to analyze and evaluate the progress of asthma based on the biological signal acquisition and processing system, which was used to observe the anti-asthmatic effects of Maxing Shigan decoction. Method: The biological signal acquisition and processing system was used to record the difference of breath wave before and after asthma induced by histamine phosphate, asthmatic latency and duration was recorded by the routine method at the same time, amplitude variations between the first or the last convulsive wave and the normal breath wave were determined according to the result observed by naked eye, then the diagnostic standard of asthmatic latency and duration were established preliminaryly. Breath wave of Guinea pigs was recorded 30 minute at the same way after Maxing Shigan decoction was gived for 5 days, influences of Maxing Shigan decoction in the asthmatic latency and duration were analyzed based on the above standard. Result: The average amplitude variation between the first or the last convulsive wave and the normal breath wave was 2 465% or 159%, Maxing Shigan decoction could delay the asthmatic latency and reduce the asthmatic duration, that showed that it had obvious anti-asthmatic effects. Conclusion: The anti-asthmatic effects of drugs can be observed objectively and standardly by the biological signal acquisition and processing system.

Abstract:Objective:To investigate the molecular mechanism of the anti-influenza A virus effects of epigallocatechin gallate(EGCG) derived from green tea in vitro. Method: The cell viability of EGCG was determined by using MTT assay. And virus inhibitory rate was evaluated based on the observation of cytopathic effect (CPE), hemagglutination assay and Real-time Quantitative PCR (QPCR). The mechanisms of action for the observed sensitivity of influenza A to EGCG at the molecular level was assessd by confocal microscope and flow cytometry. Result: Exposing Madin-Darby canine kidney (MDCK) cells with 20 mg·L-1 EGCG could inhibit influenza A replication in a time-dependent manner, probably by interrupting postbinding step. EGCG treatment could mitigate oxidative stress in influenza-infected cells and reduce the apoptosis induced by influenza infection. Conclusion: EGCG inhibits oxidative stress and apoptosis induced by influenza virus infection.

Abstract:Objective:To observe the effect on the flavoniod of Huanglian Jiedu decoction, and discuss its material base in reversing multiple-drug resistance(MDR) of cancer so as to provide the theory basis for Huanglian Jiedu decoction in clinical practices. Method: MTT assay was adopted to test the sensitivity of baicalin and geniposide to adriamycin(ADM) of K562/ADM, the inhibitory rate was calculated, the value of 50% inhibition concentration(IC50) and the multiple of the reversal of drug resistance as well as the change of the concentration of the intracellular ADM were assayed by using ADM K562/ADM of human chronic myelogenous leukemia(CML) erythroleukemia K562 as cell line. Result: Both baicalin and geniposide could partly reverse K562/ADM cell. The value of IC50 of baicalin and geniposide was 5.06 mg·L-1and 6.74 mg·L-1 respectively.The multiple of the reversal of drug resistance of baicalin and geniposide was 1.95 times and 1.46 times.The fluorescence intensity of the intracellular K562/ADM was higher than the control group, and the fluorescence intensity in the group of baicalin rised to 3.6%, The fluorescence intensity of the group of geniposide rise to 1.7%. Conclusion: The material base of the Huanglian Jiedu decoction in reversing MDR of cancer is probably related to its components baicalin and geniposide.

Abstract:Objective:To investigate the effect and mechanism of Qingying decoction on endotoxin fever in rabbits. Method: First, rabbit fever model was established by Escherichia coli endotoxin. Then rabbits were treated with Qing Ying decoction high dose, low dose to observe the animals symptoms and changes of body temperature; and tumor necrosis factor (TNF-α) content in serum, thromboxane B2 (TXB2) and 6-ketone-prostaglandin F1 alpha (6-keto-PGF1α) levels were measured in plasma by radioimmunoassay, and, and plasma tissue plasminogen activator (t-PA), and tissue plasminogen activator inhibitor (PAI) activity were determined by chromogenic substrate method. Result: Qingying decoction of both high dose group and low dose group had a significant antipyretic effect on endotoxin fever in rabbits (P<0.01), could markedly inhibit TNF-α, TXB2 levels(P<0.05 or P<0.01) and lower PAI activity (P<0.01), increase 6-keto-PGF1α content and t-PA activity (P<0.01)in fever rabbits. Conclusion: Qingying decoction has antipyretic circulation effect on endotoxin fever in rabbits. It may play the role of antipyretic circulation by inhibiting the release of pyrogenic cytokines and regulating the secretory function of vascular endothelial cells in febrile rabbits.

Abstract:Objective:In this study two substances 1-methyl-4-phenyl pyridineion(MPP+) and glutamate(Glu) were used to induce neurotoxicity and degenerative change in dopaminergic neurons in primary cell culture. By additional applying strychnine in such cellular toxic models we tested the pharmacological and toxicological influence of strychnine on neurodegenerative processes. Method: the pregnant OF1/SPF mice were choose, the midbrains were collected, the primary cells of dopaminergic neurons were cultured at 5%CO2,37 ℃,100% humidity conditions in incubator. On the day in vitro(DIV) 10, strychnine at concentration of 0.1, 1, 5, 10 μmol·L-1, MPP+ 10 μmol·L-1, Glu 500 μmol·L-1 was added for 48 h and performed staining on DIV 12.numbers of dopaminergic neurons, numbers of processes and length of processese were measured and analyzed. Result: The results demonstrated that strychnine at all range of concentrations (1-10 μmol·L-1) had no significant toxic effect on normal dopaminergic neuron cells in 48 hours without MPP+ or glutamate. However, a significant stimulating effect on neuron occured, namely, in which under treatment of strychnine at dose 10 μmol·L-1 cell number was enhanced compared with control (P<0.05); the value of length of neuronal processes also increased by 10 μmol·L-1 strychnine (P<0.05); as well as number of neuronal processes elevated in the range of 5-10 μmol·L-1 (P<0.05). When neurons were treated by MPP+ (10 μmol·L-1), strychnine played anti-neurodegenerative effect. Under concentration 10 μmol·L-1 of strychnine, the number of neurons was higher than that in MPP+ treatment (P<0.05); the value of length of neuronal processes was increased by strychnine at 10 μmmol·L-1 (P<0.05); the number of neuron processes under 5 μmol·L-1strychnine was also higher than that in MPP+ treatment group (P<0.05). In Glu(500 μmol·L-1) induced neurotoxicity, strychnine showed significant neuroprotective action, which indicated by recovery of morphology or the number of dopaminergic neurons 10 μmol·L-1 (P<0.05). Conclusion: The present results demonstrate that strychnine stimulated in vitro the growth of dopaminergic neurons from substantia negra par compacta, and played the neuronal protective effect against neurodegenerative damage in vitro.

Abstract:Objective:To prepare acylated derivatives of mangiferin, and then to evaluate their anti-inflammatory activity. Method: Mangiferin was respectively reacted with acetic anhydride, propionic anhydride and butyric anhydride to produce acylated-derivatives. Swelling of mice ear and permeability of mice celiac capillary were used to evaluate the anti-inflammatory activity. Result: 3 compounds were confirmed: (a) 7,2',3',4',6'-penta-acetyl-mangiferin(PAM), (b) 3,6,7,2',3',4',6'-hepta-propionyl-mangiferin(HPM), (c) 3,6,7,2',3',4'-hexa-butyryl-mangiferin(HBM). Mangiferin's high and medium dose group (1.0, 0.5 mmol·kg-1), PAM's high and medium dose group (0.25, 0.125 mmol·kg-1), HPM's high, medium and low dose group (0.25, 0.125, 0.063 mmol·kg-1) and HBM's high, medium and low dose group (0.25, 0.125, 0.063 mmol·kg-1) could significantly inhibit the ear swelling and permeability of celiac capillary in mice, showing obvious anti-inflammatory effect. Conclusion: PAM, HPM and HBM were novel compounds and were reported for the first time.Acylated derivatives have stronger anti-inflammatory activity than their parent compound mangiferin.

Abstract:Objective:To investigate radiosensitization and mechanism of Chinese medicine Jiawei Shengmai San on nasopharyngeal carcinoma. Method: The SD rats swimed in Glass tank for 10 minute in 30 days, at the same time,subcutaneous injection of dinitrosopiperazine(DNP) was performed,the dose was 0.5 mg every time,every 3 days the injection was carried out once for 90 days. Then,nasopharyngeal carcinoma models with Qi deficient status was produced.The samples were divided into control group,drug group,radiotherapy group,drug+radiotherapy group.The radiosensitization effect was measured by measuring tumor volume and weight,and multidrug resistance 1 gene(MDR1) expression was detected by quantitative PCR and western-blot. Result: In the control group tumor weight was (1 230.0±106.8) mg,in the radiotherapy group being (447.2±121.9) mg. Compared with the control group,the ratio of tumor inhibition was 53.25%, there was a significant difference (P<0.05). In the drug+radiotherapy group tumor weight was (259.9±64.7) mg,the ratio of tumor inhibition was 72.81%, there also was a significant difference (P<0.05). The Relative expression ratio, in the drug group was 1.21,the radiotherapy group was 1.85,drug+radiotherapy group was 1.34. Conclusion: Jiawei Shengmai San can enhance the sensitivity of radiotherapy to the nasopharyngeal carcinoma,and its mechanisms may be related to regulating MDR1 gene expression.

Abstract:Objective:To discuss protective effect of different extractions of two origin Bupleurum longicaule var.giraldii on acute liver injury in mice caused by intraperitoneal injection(ip)D-galactosamine, and to screen the optimum process and the active site of extraction. Method: The mice were divided into 9 groups: control group, model group, galactosamine group,bifendate group (150 mg·kg-1), water extraction group,65% ethanol extract group,95% ethanol extract group of B. longicaule var.giraldii sample No.1 and No.2, delivery 20 mL·kg-1 by intragastric administration(ig).Model group and control group were given normal saline by ig once a day with the same dose. After continuous administration of 7 days, model of mice were made by intraperitoneal injection of D-galactosamine hydrochloride solution, and after 16 hours, ocular venous blood were collected to determine the serum alanine aminotransferase(ALT)and aspartate aminotransferase(AST)activities and the malondialdehyde(MDA)content and superoxide dismutase(SOD)activity in liver homogenate. Result: Two origin of B. longicaule var.giraldii on aqueous extraction group, 65% ethanol group and 95% ethanol group showed that they all could decrease the serum ALT and AST activity in model, reduce the content of liver MDA and increase the SOD activity, and the 65% ethanol extract of B. longicaule var.giraldii gathered from East Taibai Mountain had the strongest effect (P<0.01, P<0.05). Conclusion: B. longicaule var.giraldii has obvious protective effect on D-galactosamine hydrochloride induced acute liver injury in mice. There is a dose-effect relationship between its hepatoprotective effect and saikosaponin a, d content.

Abstract:Objective:To observe influence of matrine on cell action potential in guinea pig papillary muscles thus to clarify its antiarrhythmic mechanism. Method: Guinea pigs were divided into control group, matrine group(10,50,100 μmol·L-1),aconitine group 1 μmol·L-1, aconitine 1 μmol·L-1+matrine 50 μmol·L-1group, aconitine 1 μmol·L-1+matrine 100 μmol·L-1 group. Conventional microelectrode technique was used to record action potential duration(APD), action potential amplitude (APA), 50 % and 90 % of APD repolarization(APD50 and APD90). Result: Matrine could prolong moderately APD50 and APD90 in a dose-dependent manner(10,50,100 μmol·L-1)and reduce APA. Aconitine could excessively prolong APD (vs control group, P<0.05), APD50 and APD90. Matrine(100 μmol·L-1)could remedy for excessive elonggation of aconitine on APD,APD50 and APD90 (vs aconitine group, P<0.05 or P<0.01). Matrine(100 μmol·L-1)also could reduce APA(vs aconitine group, P<0.05). Conclusion: Matrine could remedy for excessive elonggation of aconitine on APD,APD50 and APD90 and reduce APA of aconitine-induced arrhythmia, which could maitain action potential in a normal range, and was probably one of matrine antiarrhythmic mechanisms.

Abstract:Objective:To observe the anti-inflammatory effects of Viola yedoensis, and analyze the mechanism. Method: The models of ear edema induced by xylene and paw edema induced by carrageenan in mice were used to observe the anti-inflammation effects, the spectrophotometric method and the enzyme-linked immunosorbent assay (ELISA) were used separately to measure the content of prostaglandin E2 (PGE2) in inflammatory tissue and the content of tumor necrosis factor-alpha (TNF-α), interleukin-1β (IL-1β) in serum. Result: Compared with the model control group, the aqueous extraction and alcohol extraction of V. yedoensis (9.0, 3.0 g·kg-1) could significantly inhibit the inflammation caused by xylene or carrageenan (P<0.05-P<0.01), and decreased the content of PGE2 in inflammatory tissue and the content of TNF-α, IL-1β in serum of the mice. induced by carrageenan. There were significant difference between the high dose group (9.0 g·kg-1) and the model control group (P<0.01). Conclusion: The aqueous extraction and alcohol extraction of V. yedoensis have significant anti-inflammation effects, which may be related to the inhibition of the generation of TNF-α, IL-1β and PGE2.

Abstract:Objective:To explore the effects of anti-radiation pneumonia decoction (ARPD) on immune function in the rats with radiation-induced pulmonary injury. Method: SD rats were randomly divided into four groups: normal control group (group A), model group (group B), dexamethasone treatment group (group C) and ARPD treatment group (group D). Rats in group B, C and D were exposed to 6 MV X-ray at a dose of 20 Gy thoracically. After X-ray exposure group C and D were treated with dexamethasone (1.5 ×10-3 g·kg-1·d-1) and ARPD (35 g·kg-1·d-1) respectively by gastrogavage one time a day. The rats in group A and B were gavaged with equal volume 0.9% sodium chloride. The rats were sacrificed at days 7, 15, 30, 50 after X-ray exposure. Pathological changes in spleen tissues were observed and the index numbers of thymus and spleen, T-cell subgroups of peripheral blood, serum hemolysin level, phagocytotic rates of macrophage in abdominal cavity were detected respectively. Result: The pathological changes in group D were slighter and recovered sooner than those in other groups exposed to X-ray. After 50 days, the thymus and spleen indexes, CD3+, CD4+, CD8+, hemolysin levels, phagocytotic rates of macrophage in group D were higher than that in group B and C (P<0.05). Conclusion: ARPD can improve the immune function in the rats with radiation-induced pulmonary injury.

Abstract:Objective:To study the anti-inflammation effect of volatile oil from Periploca forrestii and the mechanism of action, for providing the basis of the clinical application. Method: The emulsion of volatile oil was prepared. The model of adjuvant arthritis in mice was established with complete freund's adjuvant (CFA) and the changes of nitrogen oxide(NO), malondialdehyde(MDA) content in the serum were determined to estimate the pharmacodynamics of anti-inflammatory activities the in vivo and mouse capillary permeability caused by acetic acid capillary permeability experiment. Rats were randomly divided into control group, model group, positive drug group (tripterygium wilfordii, 6.0 mg·kg-1), the emulsion of volatile oil high dosage group, low dosage group (the dosage 0.375, 0.094 mL·kg-1) by gavage administration.Mouses were randomly divided into control group, positive drug group (dexamethasone, 4 mg·kg-1), the emulsion of volatile oil high dosage group, group,middle dosage group, low dosage group (the dosage 0.500,0.250,0.125 mL·kg-1). According to a set of drug doses to weight 20 mL·kg-1 drug gavage. Result: The results indicate that volatile oil from P. forrestii can obviously inhibit the increase of the mouse's abdominal cavity capillary permeability induced by acetic acid, protect against the paws swelling in adjuvant-induced arthritis mice, decrease NO and MDA in the serums of model mice. The volatile oil can alleviate the pathological responses. Conclusion: The volatile oil from P. forrestii had a marked anti-inflammatory effect.

Abstract:Objective:To observe the effects of the separate application and time-dependent combination of Ziyin prescription(Liuwei Dihuang Pill, LWDHP) and Yiyang prescription (Jingui Shenqi Pill, JGSQP) on hypothalamus-pituitary-adrenal-thymus(HPAT) axis of kidney deficient aging model rats. Method: The rats were randomly divided into six groups. In addition to the normal group, other groups were injected 5%D-galactose solution 100 mg·kg-1 combined with glucocorticoid dexamethasone solution 0.5 mg·kg-1 to establish the model for six weeks, after establishing the model two weeks, each medication group was given LWDHP, JGSQP, LWDHP(morning use)combined with JGSQP (night use), JGSQP (morning use) combined with LWDHP(night use),1.16 g·kg-1 for four weeks, then the content of serum cortisol(COR),plasma adrenocorticotropic hormone(ACTH), corticotropin-releasing hormone(CRH),and CD4+,CD8+were detected. Result: Compared with natural group, serum COR of kidney deficient aging model group significantly decreased(P<0.05), plasma ACTH, CRH significantly increased(P<0.05),the level of CD8+ significantly decreased(P<0.01), CD4+/CD8+ significantly increased(P<0.01). Compared with kidney deficient aging model group, COR of Ziyin combined with Yiyang group(ZY group) significantly increased(P<0.01), ACTH of Yiyang group, Yiyang combined with Ziyin group(YZ group) significantly decreased(P<0.05), CRH of Ziyin group , ZY group, YZ group significantly decreased(P<0.05 or P<0.01),the level of CD8+, CD4+/CD8+ have no significant differences, the level of CD4+ have no significant differences between each group. Conclusion: The effects of separate application and time-dependent combination of Ziyin prescription and Yiyang prescription on HPA axis of kidney deficient aging model rats are different, ZY prescription have the best effect, but these prescriptions have no effect on immunity.

Abstract:Objective:To observe the effect of Langqing Ata (LQAT) on the metabolism of collagen in rat hepatic fibrosis model. Method: Composite factors (subcutaneous injection of 40% CCl4/olive oil solution+high-fat and low-protein compound feed+give 5% ethanol drinking water) were used to establish rat liver fibrosis model. Except the normal control group, the model rats were divided into five groups as model group, Fufang Biejia Ruangan table group(0.55 g·kg-1) and LQAT groups at three doses(11.4, 5.7, 2.85 g·kg-1 respectively). The rats were orally treated with corresponding decoctions once a day for 7 weeks. At the end of the dosing cycle, rats were sacrificed and livers were collected and then were dyed by hematoxylin-eosin staining to observe the pathological changes. The degree of fibrosis in liver tissue was observed by Masson staining. The expression levels of tissue inhibitor of metal protease 1(TIMP-I) in liver tissue were detected by immunohistochemistry method. The expression levels of type 1 collagen(CoL-I) mRNA in livers were measured by Real-time fluorescence quantitative PCR (RTFQ PCR). Result: The fiber tissue proliferation and liver inflammation activity on the hepatic fibrosis could be ameliorated by the high and middle dose groups (P<0.05-P<0.01). The immunohistochemical results showed that LQAT significantly reduced the expression levels of TIMP-I in livers (P<0.01). Results also showed the dose-dependent inhibition of LQAT on the expression levels of CoL-I mRNA in livers, and the high dose group showed significant difference (P<0.01) compared with the model group. Conclusion: LQAT played the role of anti-liver fibrosis by significantly improving hepatic fibrous tissue proliferation, and reducing the expression of TIMP-I and CoL-I mRNA.

Abstract:Objective:To investigate anti-inflammatory and antioxidant function of total flavones of Bidens bipinnata L (TFB) on lipoplysaccharide(LPS)-induced acute lung injury(ALI) and the mechanism. Method: One hundred and twenty SD rats were randomly divided into six groups:control group(physiological saline,ip),model group, dexamethasone group(5 mg·kg-1,ip), TFB treatment group(100,150,200 mg·kg-1,ip), twenty rats in each group. The rat ALI model was induced by LPS,5 mg·kg-1,ip. Six hours after modeling, all rats were sacrificed, protein content, white blood cell in bronchoalveolar lavage fluid(BALF), expression of NF-κB p65, and level of tumor necrosis factor(TNF-α), interleukin-6(IL-6),interleukin-10(IL-10)superoxide dismutase(SOD),malondialdehyde(MDA) and the activity of myeloperoxidase(MPO) in lung were measured. Result: Compared with control group, content of WBC and the protein in BALF was higher(P<0.05-P<0.01), compared with model group, NF-κB p65 expression level was(38.46±4.56)%,(31.39±4.18)% (in TFB high and medium dose group, respectively), the content of MDA was(1.73±0.19),(1.46±0.15)nmol·mg-1,and the content of TNF-α was(259.33±37.48),(211.46±32.69)μg·L-1,and the content of IL-6 was(287.46±100.18),(223.55±93.49)ng·L-1,and the activitiy of MPO was (2.69±0.57),(2.43±0.38)U·g-1, respectevly.NF-κB p65 expression level and the content of MDA, TNF-α,IL-6 and the activities of MPO in lung were significantly lower(P<0.05-P<0.01).On the other hand, the level of IL-10 was(16.73±3.87),(17.28±3.58)μg·L-1, the content of SOD was (69.46±9.84),(73.24±8.39)U·mg-1,the content of IL-10 and SOD was increased significantly(P<0.05-P<0.01). Conclusion: TFB can reduce pulmonary vascular permeability of LPS induced acute lung injury, reduce the inflammatory exudation and oxidative stress damage.

Abstract:Objective:To explore the effect of mulberry leaves polysaccharide on the immune function of mice with long-term heavy training. Method: One hundred and twenty mice were divided into six groups at random: quiet control group, training group, saline movement group, high dosage mulberry leaves polysaccharide movement group, medium dosage mulberry leaves polysaccharide movement group(250,200,150 mg·kg-1·d-1), low dosage mulberry leaves polysaccharide movement group, with 20 mice in every group. In addition to quiet control group, other groups of mice do swimming training with 5% weight for 50 minutes, with 6 times a week. The mice in mulberry leaves polysaccharide movement group were orally given with the dosage of 150,200, 250 mg·kg-1·d-1 while the mice in saline movement group were given with normal saline for 4 weeks.The hemogram, body weight, phagocytic function of macrophages, index of spleen and thymus. serum agglutination titer, the quantity of plaque-forming cell were tested. Result: Compared with the saline movement group,there was a significant increase in WBC, Lym absolute value, absolute value of neutrophilic granulocyte, mulberry leaves polysaccharid could increase phagocytic percentage and phagocytic index of mice peritoneal macrophage,which was significantly higher in phagocytic percentage of mulberry leaves polysaccharid medium high dose group(35.7±5.17)%,(36.7±4.83)% than that in the saline movement group (29.2±4.27)% and was significantly higher in phagocytic index of mulberry leaves polysaccharid medium high dose group(3.19±1.23,2.49±1.29) than that in the saline movement group(3.91±1.29)(P<0.05). polysaccharid could increase the weight of thymus and spleen significantly(P<0.05),and could increase index of spleen and thymus. Mulberry Leaves polysaccharid could increase serum agglutination titer and the quantity of plaque-forming cell significantly(P<0.05-P<0.01). Concllusion: Mulberry leaves polysaccharides can enhance long-term heavy load exercise mice immune function, regulation of immune cells and immune molecules. The effect in medium and high dose mulberry leaves polysaccharides is more significant.

Abstract:To explore the effect of total flavonoid on cerebral ischemia reperfusion injury in rats. Method: The middle cerebral artery occlusion (MCAO) was used to induce focal cerebral ischemia reperfusion injury model in rats, and the normal Wistar rats were used as control group. After Caragana microphylla Lam total flavonoid (CMF) at dose of 25,50,100 g·kg-1 and aspirin 12.5 mg·kg-1 was orally given for 4 weeks, the hemorrheologic parameters including blood viscosity(η), platelet aggregation rate (PAgT), erythrocyte aggregation index(EAI), rigidity index (IR), erythrocyte deformed index (DI), were observed. The contents of superoxide dismutase (SOD) and malonyldialdehyde (MDA) were determined. Result: In terms of the change of hemorheology [η(mPa·s), PAgT(%), EAI, IR, DI], positive control group (1.71±0.08) mPa·s,(56.49±3.74)%,(8.83±0.59),(3.94±0.22),(0.69±0.08), CMF medium group (1.72±0.11) mPa·s,(57.67±4.31)%,(8.30±0.92),(3.99±0.22),(0.73±0.06) and CMF high dose group (1.57±0.10) mPa·s,(55.59±4.58)%,(7.83±0.58),(3.65±0.17),(0.70±0.07) were obviously reduced in η, PAgT, EAI, IR. And they were decreased in DI. Through Comparation of different groups in contents of SOD and MDA, CMF low (55.45±11.67)U·mL-1,(1.38±0.96)nmol·mL-1, medium (60.23±13.59)U·mL-1,(1.17±0.44)nmol·mL-1, high dose group (64.58±9.36)U·mL-1,(1.08±0.42)nmol·mL-1 increased the bioactivity of SOD and decreased the content of MDA. Conclusion: The CMF has a protective effect on cerebral ischemia reperfusion injury in rats; it might be relevant to the role of anti-oxidative stress and decreased blood viscosity.

Abstract:Objective:To study the molecular mechanism of cinnamon polyphenols on improvement of insulin resistance in HepG2 cells in vitro. Method: The HepG2 cells were used as cell model. Cells were incubated with cinnamon polyphenols of three different concentrations (5, 10,15 mg·L-1).Control cells were incubated without cinnamon polyphenols. After 24 h,the mRNA expression of porcine glucose transporter 2 (GLUT2) phosphoenolpyruvate carboxykinase(PEPCK) and glucose-6-phosphatase (G-6-Pase) related to insulin resistance was detected by Real-time PCR(RT-PCR). Result: Compared with the control group, the RT-PCR showed the mRNA expression of GLUT2 decreased after being treated with the cinnamon polyphenols(P<0.05);the cinnamon polyphenols significantly decreased the expression of PEPCK and G-6-Pase mRNA(P<0.05,P<0.01),moreover, the cinnamon polyphenols inhibited the expression of PEPCK and G-6-Pase mRNA in a dose-dependent manner. Conclusion: Cinnamon polyphenols can significantly improve insulin-resistance of HepG2 cells and its mechanism is related to the inhibition of GLUT2,PEPCK, G-6-Pase gene expression.

Abstract:Objective:To compare inhibition effect of four neoflavonoids on respiratory burst of rat neutrophils. Method: Vc as reference, chemi-luminescence was used to compare strengths of the four neoflavonoids compounds in antineutrophill rspiratory burst. Result: Four neoflavonoids have different effect on neutrophils respiratory burst, and the marked logarithmic relationship existed between bioactivity and chemical structure. The inhibition abilities of four neoflavonoids on neutrophils respiratory burst were as follows: 2,4-dihydroxy-5-methoxy-benzophenone, R(+)-dalbergiphenol,R(-)-latifolin, 2-O-methyllatifolin. Conclusion: In chemical structure of neoflavonoids,the Cross-Shaped conjugation of A ring was favorable for the inhibition effect of four neoflavonoids on neutrophils respiratory burst, and the phenolic hydroxyl group,in A ring, was the key bioactivity group. In addition, the hydroxymethylation of hydroxyl in B ring was not favorable for improving bioactivity.

Abstract:Objective:To study the effects of triptolide on proliferation and apoptosis of human hepatocytes(L-02 cells), providing experimental basis for the clinical application of triptolide in treatment of liver diseases and the prevention of its liver toxicity. Method: Culture L-02 cells in vitro and treat the L-02 cells with 10, 30, 50 nmol·L-1 triptolide respectively for 48 h. Detect the cellular growth and proliferation using trypan blue staining and Brdu incorporation, detect cell cycle distribution using PI staining,detect the percentage of cellular apoptosis using AV/PI double staining,detect the expression levels of p21 and p53 using western blot analysis. Result: Compared with the control, after treated with 10, 30, 50 nmol·L-1 triptolide, the cell number was reduced from (2.72±1.14)×105 cells/mL to (1.95±0.38)×105,(1.44±0.44)×105,(6.97±0.47)×104 cells/mL respectively;the percentage of Brdu positive cells was reduced from (7.67±0.51)% to (5.67±0.29)%, (4.05±0.14)%, (2.51±0.15)% respectively;the percentage of cells arrested in G1 stage was increased from 65.39% to 68.47%, 71.19%,83.98% respectively;the percentage of apoptotic cells was increased from (20.54±1.44)% to (28.02±0.93)%,(36.79±0.49)%,(43.35±1.12)% respectively; the expression level of p21 and p53 protein were both up-regulated, and showed a dose-effect relationship. Conclusion: Triptolide could inhibit L-02 cells proliferation through up-regulation of p21 expression thus arresting cells in G1 stage, and induce cellular apoptosis through up-regulation of p53 expression in L-02 cells.

Abstract:Objective:To study the influence of Bushen Huazhuo Fang (BHF) on improving leptin and insulin resistance in rats with nonalcoholic fatty liver, and to explore its mechanism. Method: Forty-five Wistar rats were randomly divided into blank group, model group, silybin capsules(19 g·kg-1) group and BHF group. The blank group was fed with normal diet, model group, silybin capsules and BHF group were fed with the high fat diet. Silybin capsules and the BHF group were fed with the corresponding drug. After 12 weeks, rats were sacrificed, the rat liver index, Lee’s index, insulin resistance index(IRI), insulin sensitivity index(ISI) were calculated. Liver chemical parameters, blood lipids, blood glucose, insulin(INS), leptin(LEP) and tumor necrosis factor and liver Pathological investigation were carried out and evaluated. Result: BHF and silybin could significantly improve the alanine transaminase(ALT), aspartate transaminase(AST), LEP (P<0.01); BHF was better than silybin for improving INS, IRI, ISI (P<0.05 or P<0.01), Silybin had no significant effect on improving INS and IRI. Silybin and BHF groups were better than the model group in fat staining, glycogen staining (P<0.05 or P<0.01). Conclusion: The therapeutic effect of BHF may come from protecting liver, regulating blood lipids, improving LEP and IRI.

Abstract:Objective:To study ampelopsin on apoptosis in human gastric cancer cells (SGC-7901). Method: Cell proliferation inhibition rate was measured by MTT,The situation in different apoptosis phase was evaluated by flow cytometry (Annexin-V/PI double staining). Result: Ampelopsin could significantly promote early apoptosis in SGC-7901 after 48 h. Ampelopsin of 50 and 75 mg·L-1 induced apoptosis of (29.03±6.21)%, and (43.43±1.00)% with a dose-dependent relationship, significantly higher than that in the control group (11.83±4.67)% (P<0.05,P<0.01,P<0.01). Cells in G1 phase was (52.07±4.41)%, (57.07±2.70)%, (58.43±2.66)%, significantly higher than (42.00±3.04)% in control group (P<0.05, P<0.01,P<0.01). Conclusion: Ampelopsin could induce the SGC-7901cell apoptosis and block the cells in G1 phase.

Abstract:Objective:To study the synergistic effect between Gynostemma pentaphyllum (GP) and Scutellaria baicalensis Georgi (SBG) in control of diabetic cardiopathy(DC), providing the scientific basis for the clinical application. Method: The in vitro effects of GP (0.5, 5, 50 mg·L-1), SBG (0.5, 5, 50 mg·L-1) and the mixture of GP extract and SBG extract (GPSBG, 0.5, 5, 50 mg·L-1) on vascular endothelial cells, myocardial cells and mesangial cells were investigated. Rat model of DC was induced by high fat feeding and injection of streptozotocin(STZ)of 25 mg·kg-1. The DC rats were randomly divided into GP 300 mg·kg-1,SBG 300 mg·kg-1,GPSBG 600 mg·kg-1 and DC model group. The investigation groups were respectively ig given corresponding extracts once daily for 8 weeks, while the controls and the models were given the same volume of sodium carboxy methyl cellulose (0.5%). The cardiac hemodynamic indexes and serum lipids were measured at 24 hours after the last administration. Result: SBG 5 mg·L-1and GPSBG 5 mg·L-1could compete the suppression of ECV-304 growth in 30 mmol·L-1 glucose, increasing NO generation, reduce GSH levels, compete the inhibition of mesangial cell proliferation in 30 mmol·L-1 glucose medium. SBG 5 mg·L-1, GP 5 mg·L-1 and GPSBG 5 mg·L-1could compete the suppression of ECV-304 growth, increase NO generation, reduce GSH levels by H2O2 on myocardial cell. GP 300 mg·kg-1and SBG 300 mg·kg-1 could decrease the cardiac index in typeⅡdiabetic rats, GPSBG 600 mg·kg-1 could significantly reduce the serum levels of TG and TC, index of heart in typeⅡdiabetic rats. GP 300 mg·kg-1 could significantly increase left ventricular systolic blood pressure and heart rate, left ventricular end-diastolic pressure, the rate index of left ventricular contraction and relaxation time, and lower left ventricular end-diastolic pressure. SBG 300 mg·kg-1could significantly reduce left ventricular end-diastolic pressure, GPSBG 600 mg·kg-1 could significantly increase heart rate, left ventricular systolic blood pressure, left ventricular contraction and relaxation time rate, reduce left ventricular end-diastolic pressure in typeⅡdiabetic rats. Conclusion: GP and SBG show synergistic effect on improving cardiac function in typeⅡdiabetic rats. This may be related to the effects of GP and SBG on reducing serum TG, TC in typeⅡ diabetic rats, and their effects on endothelial cells, myocardial cells, mesangium cells and aldose reductase.

Abstract:Objective:To observe the influence of Zexie Danming Drink (ZDD) on serum lipids and adiponectin in hyperlipidemic rats. Method: A high-fat diet was fed 14 d to establish rat model of hyperlipidemia. Rats in different groups were given drugs accordingly for 21 days: ZDD high dose (4.0 g·kg-1), medium dose (2.0 g·kg-1), low dose (1.0 g·kg-1) and Xuezhikang (20 mg·kg-1). Serum triglycerides (TG), cholesterol (TC), high density lipoprotein cholesterol (HDL), low-density lipoprotein cholesterol (LDL), and adiponectin factors (ADP) and liver pathological changes were determined and observed. Result: Compared with model group, TC and TG in ZDD high dose group were significantly lowered (P<0.01), LDL-C decreased significantly (P<0.05), serum ADP level increased significantly l (P<0.01) and with a trend of elevation in HDL-C,. Compared with model group, in the middle dose group serum ADP levelwas significantly increased. Conclusion: ZDD of high dose or medium dose can regulate blood lipids and adiponectin levels, with good improvement for dyslipidemia.

Abstract:Objective:To study the protective effect of total flavonoids from leaves of Lonicera Japonica (TFLLJ) on acute hepatic injury induced by carbon tetrachloride (CCl4 ) in mice. Method: Sixty mice were randomly divided into six groups: the normal group, the model group, high-dose TFLLJ group (400 mg·kg-1), middle-dose TFLLJ group (200 mg·kg-1) and low-dose TFLLJ group (100 mg·kg-1), bifendate group (150 mg·kg-1). Except for normal group and model group, the other mice were orally administered corresponding drugs once per day for ten days. Then, the treated groups and model group were ip given 0.1% CCl4 (0.01 mL·g-1) once to produce acute liver injury on the tenth day. The liver injury was evaluated by the indexes of serum alanine aminotransferase (AST), aspartate aminotransferase (ALT), liver index (LI), liver superoxidedismutase (SOD) and malondialdehyde (MDA), and the morphological changes of liver tissue. Result: FLLJ(400,200,100 mg·kg-1)could remarkably resist the increase of ALT and AST, decrease the MDA level and could improve SOD activity (P<0.01 or P<0.05 versus model group). Furthermore, TFLLJ could ameliorate the hepatic pathological changes. Conclusion: TFLLJ have protective effects on acute hepatic injury induced by CCl4 in mice. It may be related to its anti-oxidative effect for lipids.

Abstract:Objective:To study the mechanism of tripterygium glycosides(TG) in protecting the kidney by observing the effection of TG in abnormal expression of connective tissue growth factor(CTGF) and some factors of kidney function in artificial diabetes rats. Method: Forty-five Male SD rats were randomly divided into normal control group, diabetic nephropathy(DN) and diabetic nephropathy group treated with tripterygium glycosides(TG) group,1.8 g·kg-1·d-1 ig,once a day,using 8 weeks) DN group and TG group rats were intraperitoneal injected with sacrificed. The rats were sacrificed after 4 and 8 weeks. The body weight, blood glucose, blood urea nitrogen(BUN), creatinine(Cr) and 24 h urinary protein were measured in each rat. The pathology changing of the rat kidney was observed by HE staining. The expression of CTGF protein in kidney was measured by immunohistochemistry. Result: Compared with control group, the level of blood glucose, BUN,Cr and 24 h urinary protein were significantly increased in DN group (P<0.01).The expression of CTGF protein was increased in DN group compared with control group(P<0.01); After intervention with tripterygium glycosides, these indexes were all significantly declined except blood glucose (P<0.05,P<0.01). Conclusion: The TG on reducing the urinary protein in DM rats and improving renal injury in diabetic nephropathy may related to the inhibition of renal CTGF expression.

Abstract:Objective:To observe vascular protection effect of Gegen Qinlian decoction(GQD) in rats with arrhythmia. Method: Sixty rats were randomly divided into six groups: sham group, model group, positive drug group, and three dose groups of GQD (2.16, 4.32, 8.64 g·kg-1).Ligating the left anterior descending artery for 30 min ischemia and then reperfused for 15 min, limb II ECG was investigated in the isxhemia/reperfusion model. After the operation, postoperative radioimmunoassay was used to detect the content of plasma endothelin (ET) and angiotensin II(Ang II). The tumor necrosis factor-α (TNF-α) of serum and the nuclear factor-κB (NF-κB) in myocardial tissue were measured by using ELISA. Result: Compared with the sham operation group, increased AngII,TNF-α and NF-κB expressions were found in model group rats after reperfusion(P<0.01);Compared with the model group, GQD could significantly reduce reperfusion arrhythmia score (P<0.05),NF-κB,TNF-α and ET could be significantly reduced in GQD groups(P<0.01). Conclusion: GQD has a certain protective effect against reperfusion induced arrhythmia, which is related to reducing the content of AngII and then suppressing the expression of NF-κB,TNF-α,ET in infarcted reflow.

Abstract:Objective:To explore the anti-fibrosis effects of Cassia seed on the renal fibrosis in diabetic rats and its possible mechanisms. Method: Seventy rats were randomly divided into model group (n=60) and normal control group (n=10). Fasted for 12 h, the model group rats were left intraperitoneal injected 55 mg·kg-1 of streptozotocin to induce diabetes. Control rats were given equal volume of normal saline. The model-established rats were randomly divided into 5 groups: diabetic model group(NS,4 mL·kg-1), captopril group (10 mg·kg-1), low, medium and high dose Cassia seed groups (1, 5, 10 g·kg-1). After eight weeks of treatment, fasting blood glucose, serum creatine, 24 h urine protein (U Pro), clearance rate of creatine were detected. The morphology was observed, and the mRNA expression of transforming growth factor β1(TGF-β1) and connective tissue growth factor(CTGF) were detected by RT-PCR. The protein expression of Smad3 and Smad6 were detected by western blot. Result: Compared with normal control group, fasting blood glucose, serum creatine, 24 h urine protein (U Pro) were significantly increased (P<0.05), as well as the mRNA expression of TGF-β1 and CTGF and the protein expression of Smad3 were significantly increased (P<0.05), but clearance rate of creatine and the protein expression of Smad6 were significantly decreased (P<0.05) in diabetic model group. Compared with diabetic model group, there were obvious improvements after the treatment of Cassia seed (P<0.05), in addition, the renal pathological changes were also improved. Conclusion: Cassia seed has protective effect against the renal fibrosis induced by STZ in rats, the mechanisms may be related to its ability of down-regulating TGF-β1, CTGF and smad3, and up-regulating smad6 in rats.

Abstract:Objective:To explore the prevention effect of Xuebijing injection on radiation-induced lung injury in patients with non-small cell lung cancer (NSCLC). Method: Eighty three cases of NSCLC patients underwent radiotherapy and confirmed by pathological examinations were randomly divided into two groups. The treatment group (n=42) received Xuebijing injection from the first day of radiotherapy, the control group (n=41) received radiotherapy alone. The levels of tumor necrosis factor-α (TNF-α), coagulation function, KPS scoring, and incidence of radiation-induced lung injury within 6-month-follow up were observed. Result: The levels of TNF-α in the two groups at the second week and the end of radiotherapy increased obviously, while the increasing degree in the treatment group was obviously smaller than that in the control group. The coagulation function in the two groups had different degree of deterioration, while the deterioration degree in the treatment group was smaller than that in the control group. The KPS scoring in the treatment group at the second week and the end of radiotherapy decreased slightly, and obviously increased 6 months after radiotherapy, and was also higher than that in the control group at the end of radiotherapy and 6 months after the end of radiotherapy. The incidences of early and advanced radiation-induced lung injury in the treatment group were obviously lower than that in the control group. Conclusion: Xuebijing injection has the effect on anti-endotoxin, reducing inflammation factors releasing, adjusting anticoagulation and coagulation function, and increasing the KPS scoring, so as to effectively prevent radiation-induced lung injury and improve the life quality of the patients.

Abstract:Objective:To re-evaluate the clinical efficacy and safety of Longfengbao tablets for the treatment of perimenopausal syndrome. Method: Eighty-four patients were randomly divided into treatment group and control group (n=42 each). The control group was treated with livial 2.5 mg daily; the treatment group was orally given Longfengbao tablets 1.2 g tid. The treatment lasted for 12 weeks. Kupperman score, serum estradiol (E2), follicle stimulating hormone (FSH), luteinizing hormone (LH) l, endometrial thickness were measured and traditional Chinese medicine clinical symptom score were compared between the two groups. Result: After the treatment, the two sets of the Kupperman ratings and deficiency syndrome integral were significantly lowered than before (P<0.01), the deficiency syndrome points of the treatment group was lower than that of the control group(P<0.01). Serum E2 level in two groups was significantly increased after treatment (P<0.01). The effective rate of the treatment group was 88.09% while the control group was 92.85%; the endometrial thickness did not change significantly after the treatment in both groups. The treatment group showed no adverse reactions. Conclusion: Longfengbao tablets can significantly improve the clinical symptoms of perimenopause syndrome, reduce the Kupperman score, elevated serum E2 levels without increasing endometrial thickness, and it is safe and effective.

Abstract:Objective:To investigate the clinical efficacy and impact of Qingnao Jiangya tablets (QJT) on endothelial function in the treatment of liver yang hypertension. Method: One hundred and fifty-six patients with hypertension were randomly divided into treatment group of 79 cases and control group of 77 cases. Grade Ⅰ hypertension was treated with QJT; Grade Ⅱ hypertension was additionally treated by QJT based on the original treatment, The controls were only received placebo. QJT was orally given 4-6 tablets tid for 4 weeks. Blood pressure, liver yang score, nitric oxide (NO) and endothelin (ET-1) were evaluated for the two groups. Result: QJT significantly reduced systolic blood pressure (SBP) and diastolic blood pressure (DBP) for the grade Ⅰ and grade II hypertension patients(P<0.01). QJT significantly improved the patient’s liver yang scores, and the disease efficiency and syndromes efficiencies were 84.81%, 89.87%(P<0.01). QJT increased NO level, reduced ET-1 level and adjusted NO/ET-1 ratio(P<0.01). Conclusion: QJT had obvious antihypertensive effect for grade Ⅰ and grade Ⅱ hypertensive patients. It could improve clinical symptoms, and its mechanism may be achieved by adjusting the ratio of NO/ET-1.

Abstract:Objective:To Investigate the clinical effect on chronic viral hepatitis B(CHB)treated with integrative Chinese and western medicine. Method: Sixty cases of chronic viral hepatitis B were randomized into observation group (39 cases) and control group (37 cases). The control group was treated with pegasys 180 μg/times,1time/week, subcutaneously; the observation group was treated with the Shiwei Xihuangcao granules or Renshen Jianpi wan according to the different traditional Chinese medicine (TCM) symptoms on the basis of the control group. The course of treatment was 48 weeks. Liver function, TCM syndrome score, HBV-DNA quantitative and HBV markers of two groups were observed. Result: The level of aspartate aminotransferase (AST), alanine transaminase (ALT), total bilirubin (TBil) of the observation group was lower than that of the control group after 24, 48 weeks treatment (P<0.05, P<0.01). The TCM syndrome score of the observation group was lower than the control group after 12, 24, 48 weeks treatment(P<0.01). The level of serum HBV-DNA of the observation group was lower than the control group after 24, 48 weeks treatment (P<0.05,P<0.01). The Liver function normalization rate and HBV-DNA negative rate of the observation group were better than the control group after 48 weeks treatment(P<0.05). Conclusion: Integrative treatment of CHB has more obvious clinical advantages than the simple treatment of western medicine and worth to promote to use clinically.

Abstract:Objective:To investigate the clinical curative effect of the heart-spleen deficiency insomnia by split Guipi soup according to thepathogenesis. Method: Sixty patients suffering from heart-spleen deficiency type of insomnia were randomly divided into treatment group and control group. The treatment group was given split soups from Guipi soup, namely"wu fang" and "mei fang", in the morning and before going to bed accordingly. The control group was given Guipi soup. The duration of treatment lasted 2 weeks. Patients of both groups were measured before and after treatment by the clinical curative effects, Pittsburgh sleep quality index (PSQI) and mESS. Result: After treatment, the clinical curative effects of the two groups were similar, the improvement of the sleep quality in the treatment group was superior to that of the control group, with statistical difference (P<0.05). Conclusion: The split prescription according to insomnia pathogenesis could gain better clinical efficacy compared to simple syndrome differentiation.

Abstract:Objective:To observe the effect of herbs of reinforcing the kidney and promoting blood flow combined with low dose of memantine for treatment of cognitive dysfunction after stroke. Method: From January 2009 to June 2011, 80 patients with after stroke cognitive dysfunction were collected as investigative cases. And all patients were given the treatment of herbs of reinforcing the kidney and promoting blood flow combined with small dose of memantine. Then we evaluated the curative effect by applying the simple intelligent state examination scale (MMSE) and Montreal cognitive assessment scale (MoCA, Beijing edition) before treatment, a month and three months after treatment. Result: The MMSE score of all patients was 24.84±3.21 and MoCA score was 23.76±3.00 before treatment, those were all below the normal reference value of 26.Along with the treatment continuance, MMSE and MoCA scores were increased gradually, they were 27.28±4.57 and 26.47±3.43 at one month after treatment, and 29.78±4.44 and 28.89±3.37 at three months after treatment. They were higher than those of before treatment (P<0.05). Along with treatment continuance, significant efficiency gradually raised. The significant efficiency at three months after treatment was 21.25%, which was higher than that at a month after treatment (15.0%) (P<0.05); The total effective rate at three months after treatment was 86.25%, and those at a month after treatment was 83.75%, without significant difference. Conclusion: Treatment by herbs of reinforcing the kidney and promoting blood flow combined with small dose of memantine could improve cognitive function for cognitive dysfunction after stroke. The effect is clear and worthwhile to be applied clinically.

Abstract:Objective:To investigate the clinical effect of Bushen Huoxue Tongluo capsule(BHTC) combided with western mediciine on patitents with Parkinson's disease (PD). Method: One hundred and twenty PD patients were entorlled in the randomized control tiral (RCT). The patients were randomly divided into two groups (60 each): treatment group (BHTC and western medicine,peros) and control group (wetern medicine only, peros). At the end of the third month, he unified parkinson disease rating scale (UPDRS) and quality of life of Parkinson's disease rating scale (PDQ) were compared respectively for each group by a neurology doctor on the time of pre-treatment and one month, two months and three months post-treatment to record mental, behavioral and emotional state (UPDRS-Ⅰ), daily activities (UPDRS-Ⅱ), motor function check (UPDRS-Ⅳ), motor complications(UPDRS-Ⅳ), improvement of motor function (LSIB) and Parkinson's disease life quality rating scale (PDQ). Result: The study established a comprehensive treatment program for Parkinson's disease. After one month, two months and three months treatment, the effective power was significantly highter in treatment group than in control group (P<0.05). Conclusion: BHTC has an obvious effect on PD patients. BHTC combined with western medicine can improve motor function and daily living activity in PD patients better than western medicine only.

Abstract:Objective:In order to observe the therapeutic efficacy and side effect of acesodyne and lubricating gut condensed decoction(ALGCD) in treatment of constipation after operation of mixed hemorrhoids. Method: Patients with hemorrhoids received operation in hospital were randomized into two groups, treatment group (n=41) take ALGCD orally after operation, control group (n=37) take Maren soft gelatin capsule after operation. The time of first defecation after operation was observed. The stool form scale and pain scale of defecation at first time, 3-4th day, 6-7th day and 9-10th day after operation were also observed. Result: The time of first defecation after operation in treatment group was shorter than that in control group, but there were no significance difference in statistics. The stool form scale and pain scale of defecation at first time, 3-4th day in the treatment group were better than those in the control group (P<0.01), and also better at 6-7th day (P<0.05), until 9-10th day there were no significance difference in the two groups. Conclusion: ALGCD can stimulate bowel movement, intenerate the stool and alleviate the pain, can't cause diarrhea easily. Application of ALGCD is an effective method in prevention and treatment of constipation after operation of mixed hemorrhoids.

Abstract:Objective:Explore combining traditional Chinese and western medicine treatment of prophase diabetic nephropathy therapy effect. Method: Selection in January 2010 to december 2011 in our hospital of 72 cases with prophase diabetic nephropathy, the western medicine that oral enalapril(25 mg,bid) and pancreatic enzyme(240 U,tid)symptomatic treatment of peptides 37 cases as control group, in western medicine treatment on the basis of traditional Chinese medicine to serve with the 34 patients in as a research group. The two groups have been treatmented for 12 weeks. Observe and compare the curative effect of two groups, the determination of the urea nitrogen (BUN) before and after, creatinine (SCr), 24 h urine protein quantitative change. Result: The total effective rate of 82.8% in the control group, the total effective rate of 94.8% in the treatment group treatment of compared with the two groups,the statistically is significant difference (P<0.05); The two groups after treating BUN, SCr, 24 h urine protein quantitative obviously lower than that, there is statistically significant (P<0.05), compared with controls group , the effectiveness of the treatment group's each index is much better than control group, the difference was statistically significant (P<0.05). Conclusion: Combining traditional Chinese and western medicine treatment of diabetic nephropathy therapy effect significantly, which is worthy be popularized.

Abstract:Objective:To investigate the clinical efficacy of traditional Chinese medicine combined with percutaneous laser disc decompression (PLDD) for lumbar disc herniation (LDH). Method: Sixty-four patients of LDH were randomly divided into study group and control group, 32 cases in each group. The study group was treated with use of PLDD and Chinese traditional drug (oral administration for 8 weeks), while the control group patients were treated by PLDD. All patients were followed up and the efficacy was evaluated by Japanese low back pain surgery score (JOA) and visual analog scale (VAS). Result: Compared with the controls, the study group showed decreased VAS in the first and second week after treatment significantly (P<0.01). After PLDD 1 month, 2months and 3 months, the study group still showed superior therapeutic effect than the control group (P<0.05). Conclusion: Traditional Chinese medicine combined with PLDD for treatment of LDH can release pain, elevate therapeutic effect, and is worthy of clinical application.

Abstract:The clinical application of research literature about Chinese medicine Arnebiae Radix was organized and introduced to provide a reference for further study over the past five years. Thirty representative literatures were classificated and summarized in CNKI and VIP database and retrievaled taking Arnebiae Radix as subject words. Arnebiae Radix has the function of cooling blood, detoxify, and laxative effect of intestinal, which clinically can be effective in the treatment of pressure sores, the neonatal red buttocks, skin ulcers, skin itching, dermatitis and other common skin diseases, and commonly used for burns, phlebitis, ectopic pregnancy, cervical erosion, placenta accreta, anorectal postoperative medical abortion, nail bed defect, as well as chronic nephritis. At present, the Chinese medicine Arnebiae Radix has widely used in clinical, but usually used by borage oil or Arnebiae Radix ointment type. The aim of this paper is to provide little clinical reference for the future development of Arnebiae Radix new dosage form.

Abstract:To conduct a literature review and analysis on the influencing factors in the process of the preparation of freeze-dried liposomes. Using freeze-dried liposome as the subject term to collect literature on freeze-dried liposome preparation aspects in the Chinese academic literature library (CNKI), according to its correlation cited 35 articles, and concluded and summarized the influencing factors during the preparation process. The kinds and dosage of protective agent,ways of pre-freezing and speed, the degree of vacuum, drying time, hydration medium and other factors will affect the quality of the freeze-dried liposomes. In the process of preparation of freeze-dried liposomes, a variety of factors must be taken into account to prepare liposomes of a high encapsulating rate, high stability and small particle size changes. Application of freeze-drying technology can prevent drug oxidation, prolong drug liposome guarantee period, and lay a solid foundation for the promotion and application of liposome drug.
