  • 1  Study on a Seropharmacological Method for an Effective Antifibrotic Formula

    Abstract:
    The serum was collected from rats fed on Fuzheng Huayu (FZHY) decoction, and FZHY decoction was lyophilized into powder. And both were incubated with hepatic stellate cells (HSC) and hepatocytes. Then cell morphology and functions were observed to study the seropharmacological methods for antifibrotic traditional Chinese herbs. The results showed that the FZHY powder inhibited cell proliferation and viability. The deactivated rat's sera drawn 1h after FZHY-treatment twice had no cytotoxity, promoted hepatocyte proliferation, and inhibited HSC proliferation. Therefore, this seropharmacological method could be applied for antifibrotic herbal formulae.

    Abstract:
    Using a serum pharmacological method that the sera,derived from the rats orally taking FZHY,were incubated with culturd ito cells(Fat-storing cells,passage 1) of the rats,it wasinvestigated that FZHY affected ito cell functions such a cell proliferation and collagen synthesi.The results showed that the FZHY sera had no cytotoxicity towards the ito cells inhibited cell proliferation and collagen synthesis.Furthermore,this inhibition had a positive relation with administration and dosage of the FZHYdecoction bood-taken tine,and the concentration of the sera in the culture,was concluded that the inhibition of FZHY on ito cells may be one of mechanisms through which liver fibrosis was prevented with it.

    Abstract:
    We studied the effects of salvianolic acid B(Sa1.B)、Rb、Re and Rg fractions of Danshen or Sanqi on platelet aggregation and adhesion,TXA2 and PGI2 in normal rats. The results demonstrated that Re fraction (68mg/kg)and Rg fraction (108mg/kg),given orally,could markedly counteract ADP induced platelet aggregation.Rg fraction (108mg/kg) and Sal.B fraction(48mg/kg) could markedly inhibit platelet adhesion.Rb fraction(112mg/kg) have no effects on platelet aggregation and on adhesion.Rg fraction(108mg/kg) and Sal.B fraction (48mg/kg) could markedly decrease the level of TXA2,but had little effect on 6-Keto-PGF. Moreover, Rg fraction 108mg/kg could decrease the ration of TXB2/6-Keto-PGF,too. The results suggested that the inhibition effects on platelet aggregation and adhesion of Rg fraction(108mg/kg) and Sal.B fraction(48mg/kg) of Danshen or Sanqi might be closely related to its inhibition effect on the release of TXA2 in normal rats.

    Abstract:
    The factors effecting the physical property of cataplasm were investigated by means of the homogeneity,layer residue,flexibility and adhesive ability of the plaster as comprehensive consideration,and adhesive force(180°) while exfoliated as the quantitative marker.The main factors,stirring time,refining temperature,and adding order of matrixs were studied by orthogonal design tests.The optimized procedures were stirring for 20~40minute,plaster at 50℃,and after mixing the excipients and filling material,then adding the bonds.

    The technical parameters of PNS's purification with macroporous resin were studied by monitoring the eluted ratio and purity of PNS.Over 80% of PNS was eluted from macroporous resin.The purity of PNS was up to 71.1% in the dried part of 50% ethanolic elution.This technique was a good method to purify PNS.

    Abstract:
    Effect of Yixin oral liquid on Tnyocardial infarct size and hemorheology were studied in rats with acute myocardial ischemia,The results showed that the liquid at a dosage of 8 or 16g/kg,ig×5d could significantly reduce the size of myocardial infarcts and decline serum CK and LDH activities at 24h after ligation of LAD. It could also decrease ηBH、ηBL、ηBHR、ηBLR、ηP、ERI、RCET、whereas HCT、Fib、ESR and EEC were not changed markedly.Yixin at a dosage of 4g/kg had no significant effects on the above parameters.

    Abstract:
    Cognition-enhancing effect of Liuwei Dihuang decoction(LW)on SAM was observed by using passive avoidance performance,shuttle box and water maze tests in the present paper.It was showed that memory registration and retention ability,spatial memory as well as conditioned avoid response were all greatly declined in SAMP8.However,chronic administration of LW to SAMP8 enhanced the memory registration and retention ability in passive avoidance performance, promoted the spatial memory ability in water maze test and partially improved the leaning behavior in conditioned avoidance performance.The present results suggested that chronic administration of LW significantly ameliorated agerelated decline of learning and memory ability in SAM.

    Abstract:
    It was introduced for new progression of chemical component and pgarmacological studies on main chemical fractions of Radix Bupleuri in recent ten years.Saikosaponin k,l,m,n,q,t,were discovered lately.Through the pharmacological studies on main chemical fractions,saikosaponin could inhibite the activity of trypsin,regulate immunity,and treat nephrosis and insomnia.The mechanisms of these actions were studied.Through farther separated studies of polysaccharide of Radix Bupleuri,the possible active fractions were brought forward.It provides the background for the new drug study of gastric mucosa protection and immunoloregulation.New prospect was showed for the application of Radix bupleuri because of double action to adenylate cyclase (AC) of the extracts of Radix Bupleuri.

    Abstract:
    The inhibitory effect of Fuzheng Huayu 319 Recipe(319 Recipe) on hepatic fibrogenesis was observed. 319 Recipe was found to reverse hepatic fibrosis. The results indicated that 319 Recipe was more potent than colchicine and Dahuang Zhechong pill not only in inhibiting hepatic fibrogenesis and promoting reversion of hepatic fibrosis, but also in protecting and recovering hepatic functions.

    To study the effects of Siwu Tang on the mice of blood deficiency induced by cyclophosphamide,we employed the methods of circulating blood cells detection by microcellcounter,flow cytometric enumeration of CD34+ himatopoietic stem and progenitor cells by double fluorescent labeling.The results showed Siwu Tang promoted the recovery of stem and progenitor cells and circulating WBC on the blood deficient mice,and enhanced immune function.proliferative effect of Siwu Tang on bone marrow may play a key role in improvement of hematopoiesis with Siwu Tang treating blood deficient mice.

    Danggui Buxue Decoction did not directly stimulate reproduction of CFU-E in vitro,but the decoction-contained serum could has the effect with a obvious distinction compared with normal control serum.The quantity of CFU-E was promoted with increasing the serum dosage.The serum containing the separated formulae could stimulate reproduction of CFU-E,respectively.The proper sequence of the stimulating effect was as follows:Huang Qi>Danggui and Huang Qi(1:5)>Danggui and Huang Qi(1:1)>Danggui>normal control serum.

    Abstract:
    Effects of compatible application of RHIZOMA CHUANXIONG and RADIX PAEONIAE RUBRA on regulation of lipid metabolism, scavenging of free radicals and protection of vascular endothelium in rat with hyperlipidemia were investigated. It was found that compatible application of RHIZOMA CHUANXIONG and RADIX PAEONIAE RUBRA reduced total cholesterol (TC), triglyceride(TG), low density lipoprotein, which was obtained by single use of RHIZOMA CHUANXIONG and RADIX PAEONIAE RUBRA, respectively.Conceming scavenging of free radicals and protection of vascular endothelium, compatible application of RRHIZOMA CHUANXIONG and RADIX PAEONIAE RUBRA decreased the malondiadehyde (MDA) with increased release of nitric oxide, but single use of RHIZOMA CHUANXIONG and RADIX PAEONIAE RUBRA did not indicate the same actions,which suggested that compatible application of the two drugs can create a synergism.

    Objective:To study the effects of nutrition blood flow of cardiac muscle in mice by different extracts in Radix Ophiopogonis.Methods:The Rb86 mice model was used.Results:The total saponins and total polysaccharides in Radix Ophiopogonis can increase nutrition blood flow of cardiac muscle in mice.Conclusion:The total saponins and polysaccharides in Radix Ophiopogonis could be main effective fractions protecting myocardial ischemia.

    Objective:To investigate the effect of BuYanHanWu (BYHW) Tang upon infarction,vessel damage penumbra (VDP),fibrinolytic system activity and endothelin.Methods:BYHW Tang consists of two parts,invigorationg Qi section (IQs) and promoting blood flow section (PBFs).To utilize the model of cold photochemical initiation of thrombosis to observe the effects of the whole of BYHW Tang,IQs and PBFs,respectively,on infarction size,VDP size,degree of vessel damage,localized cerebral blood flow,plasma tissue plasminogen activator(t-PA),plasminogen activator in hibitor (PAI) and plasma endothelin content.Result:BYHW Tang influences all indexes significantly.IQs and PBFs both have the actions of limiting infarction size and degree of vessel damage,promoting of cerebral blood flow and inhibiting PAI activity.On the other hand,reducing the size of VDP and increasing the activity of t-PA by BYHW Tang come mainly from IQs,and the effect of reducing ET content is based on PBFs.Conclusion:The IQs is emphasized in the priscription of BYHW Tang.It can enhance the activity of fibrinolytic system and reduce the VIP size.The result hints BYHQ Tang is not only formulated for stroke caused by deficiency of Qi,but also effective for the treatment of ischemic stroke.

    Abstract:
    This article reviews the studies of the anti-tumour actions of alkaloids in Radix Sophorae Flavescentis (Kushen). Recently, it has been demonstrated that matrine in Kushen does not only have inhibitory actions on tumour proliferation and transfer, but can also induce apoptosis in tumour cells. In general, many studies suggest that matrine may have unique mechanism in combating tumour growth which may become a useful treatment for tumours.
