    • Investigation of Apparent Oil/Water Partition Coefficients of Berberine and Its Absorption Characteristics in in situ Intestinal Perfusion Model

      Abstract:Objective: To determine apparent oil-water partition coefficient of berberine monomer and berberine in coptis total alkaloids,investigate absorption kinetics of berberine at jejunum and ileum of rats from berberine monomer and coptis total alkaloids and Qingre Tongfu tablets,then compare correlation between apparent oil-water partition coefficient and absorption kinetics. Method: Apparent octanol-water partition coefficient of berberine was determined by shake-flask method,in situ intestinal recirculating perfusion model of berberine was performed to study its absorption at different intestinal regions.The concentrations of berberine were analyzed by UPLC.Calculated berberine intestinal absorption rate constant(Ka) and apparent permeability coefficient(Papp) in different intestinal regions of rats. Result: Oil-water partition coefficient of berberine and coptis total alkaloids were 0.213 5 and 0.175 7,respectively.Ka of berberine were 0.046 6 and 0.034 9 in jejunum and ileum,respectively;While Papp were 4.88×10-6,4.13×10-6cm·s-1,respectively.Ka of berberine in coptis total alkaloids were 0.035 1 and 0.026 6 in jejunum and ileum,respectively;While Papp were 3.92×10-6,3.21×10-6cm·s-1,respectively.Ka of berberine in Qingre Tongfu tablets were 0.028 3 and 0.018 1 in jejunum and ileum,respectively;While Papp were 3.11×10-6and 2.23×10-6cm·s-1,respectively(n=6). Conclusion: Berberine had absorption in jejunum and ileum with slow absorption rate,but the former was greated than the latter with a significant difference;Absorption rate of berberine was in order of berberine reference standard>coptis total alkaloids>Qingre Tongfu tablets,the predicted results of oil-water partition coefficient of berberine were basically the same with intestinal absorption results.

    • Preliminary Exploration of Industrialization for Purification of Hydroxysafflor Yellow A from Carthamus tinctorius by Macroporous Resin

      Abstract:Objective: To optimize industrialization amplification process for purification of hydroxysafflor yellow A from Carthamus tinctorius by macroporous resin. Method: The content of hydroxysafflor yellow A was determined by HPLC,single factor tests were adopted to investigated effects of pH of sample solution,the concentration and amount of eluant,quality ratio of raw materials to resin on purification technology,then verified optimized technology through pilot and industrialization production. Result: Optimum purification conditions were as follows:ratio of crude drug to D101 resin 1: 6,pH of sample solution 4.0,eluted impurity with 4 BV water at flow rate of 3 BV·h-1,then eluted with 6 BV 5% ethanol at 3 3 BV·h-1,collected eluent. Conclusion: This technology was simple and feasible with good separation,it could be suitable for industrialized production of hydroxysafflor yellow A.

    • Optimization of Preparation Technology of Total Glucosides of Buyang Huangwu Decoction Capsules

      Abstract:Objective: To optimize preparation process of total glucosides of Buyang Huanwu decoction(BYHWT) capsules. Method: With the content of astragaloside Ⅳ and extract yield as indexes,water extraction process of this capsules was optimized by orthogonal design.Isolation and purification of total glucosides from BYHWT was achieved by using 732 cation exchange resin firstly,then extracting with chloroform,and finally using DA-201 macroporous resin.Molding technology of this capsules was investigated by single factor tests. Result: Optimum water extraction technology was:extracted 2 times with 10,8 times the volume of water,1 h per time.Lactose as hygroscopic agent,80% ethanol as wetting agent,relative humidity should be less than 50% for production environment,1.02 g particles should be dispensed into 6 No. 2 capsules. Conclusion: Optimized extraction and molding technology was reasonable and feasible,which provided a reference for industrial production of total glucosides of BYHWT capsules.

      Abstract:Objective: To screen dominant bacteria and explore effects of pure fermentation on quality of processed products of Liushenqu. Method: Microbial strains separation method was adopted,bacteria in Liushenqu from Tongrentang and laboratory-made,amylase and protease activity was determined after Liushenqu pure fermented by some mildew,amylase activity was determined based on iodine-starch colorimentric method,protease activity was measured with Folin reagent. Result: Twelve mildews was screened,aflatoxin could improve vitality of amylase and protease in Liushenqu,protease activity was the highest which produced by aspergillus versicolor isolating from homemade Liushenqu,followed aspergillus carneus and absidia corymbifera isolating from Tongrentang,produced protease activity of the rest groups was significantly lower than traditional natural fermentation groups. Conclusion: There was no amylase and protease in raw materials of Liushenqu,pure fermentation could stabilize improve quality of processed products of Liushenqu.

    • Preparation and Characterization of Gentiopicroside Nano-lipid Carriers

      Abstract:Objective: To prepare gentiopicroside nano-lipid carriers(GEN-NLC) and investigate its stability,in vitro release and properties. Method: Solvent dispersion method was used to prepare GEN-NLC.With particle size,Zeta potential and encapsulation efficiency as indexes,effects of proportion of solid-liquid lipid,drug-lipid ratio and the type and concentration of surfactant on preparation technology were investigated,morphology of prepared GEN-NLC was observed by transmission electron microscopy,its characteristics and stability was investigated. Result: Optimal prescription was as following:0.1% poloxamer 188 as surfactant,drug-lipid ratio 1: 10,liquid lipid ratio 10%.Encapsulation efficiency of prepared GEN-NLC was (38.19±1.61)%,drug loading was(3.47±0.08)%,particle size was(129.9±3.07) nm,PDI was (0.264±0.01),Zeta potential was (-22.5±0.42) mV. GEN-NLC was spherical particles and showed monodisperse distribution,rounded appearance and uniform size.After stored at 4℃ for 30 d,indicators were not changed significantly,release of gentiopicroside 39.65% in 4 h,cumulative release amounted to 79.86% at 36 h. Conclusion: It showed that there were good physicochemical properties and stability of GEN-NLC prepared by solvent dispersion method,and had a certain degree of sustained-release long-lasting effect.

      Abstract:Objective: To Establish comprehensive evaluation model of production process of Qingre Jiedu oral liquid,and apply to production monitoring of this preparation. Method: Qingre Jiedu oral liquid was selected as study object,with shared peaks as multivariate variables,principal component analysis,factor analysis and other methods were adopted to achieve data compression and interpretation,and the overall quality changes of preparations was revealed by a small amount of principle component analysis. Result: Optimum extraction technology of Qingre Jiedu oral liquid was:extracted 2 times with 10 times the amount of water,100 min per time. Conclusion: Dimensionality reduction analysis could simplify analysis process and reveal inherent laws of raw data penetratingly,it was suitable for control of the overall quality of traditional Chinese medicine.

    • Investigation of Effects of Moisture Content on Quality Guarantee Period of Decocted Extract

      Abstract:Objective: To investigate effects of moisture content on quality of decocted extract and determine its quality guarantee period. Method: Decocted extract was prepared by traditional concentrated process,moisture content was determined,according to microbial limit test method from the appendix XIII C in 2010 edition of 'China Pharmacopoeia',examination for effects of moisture content on microbiology of pastes was adopted. Result: Moisture content had significant effects on microbiology of decocted extract,with increasing of moisture content,bacterial colonies would appear,stability of decocted extract decreased. Conclusion: By controlling moisture content strictly,we could avoid decocted extract becoming mildew,harden and other phenomenon,then quality of decocted extract was ensured and its quality guarantee period was extended.

      Abstract:Objective: To do a comparative study of compressed pieces and traditional pieces of Lonicera japonica. Method: Appearance and TLC identification had been used to compare compressed pieces and traditional pieces of L. japonica.With the content of chlorogenic acid and dry extract rate as indexes to compare decoction quality between compressed pieces and traditional pieces of L. japonica in honeysuckle decoction and Yinqiao powders.Then dissolution curve of chlorogenic acid between compressed pieces and traditional pieces of L. japonica was compared,and f2similarity factor evaluation was adopted. Result: Appearance of compressed pieces was good,TLC identification of compressed pieces and traditional pieces of L. japonica was distinct and highly specific.The content of chlorogenic acid and dry extract rate in compressed pieces were a little higher than traditional pieces in honeysuckle decoction and Yinqiao powders.Dissolution behavior was similar between compressed pieces and traditional pieces through contrast of f2 similarity factor. Conclusion: Quality of L. japonica did not change after compressed,which didn't affect dissolution of chlorgenic acid,and could ensure boiling quality of decoction.

    • Optimization of Alcohol Precipitation Technology of Polysaccharides from Sophora alopecuroides by Response Surface Methodology

      Abstract:Objective: To optimizate alcohol precipitation technology of polysaccharides from Sophora alopecuroides. Method: Based on single-factor test,with yield of polysaccharides as index,precipitation time,the concentration and adding multiples of ethanol was optimized according to mathematic model built by central composite design in Design Expert software on principle of response surface methodology.The content of polysaccharides from S. alopecuroides was determined by phenol-concentrated sulfuric acid method. Result: Optimum alcohol precipitation technology was as following:precipitation time of 26 h,the concentration of ethanol 97%,adding multiples of ethanol 4.3 times.Under these conditions,yield polysaccharides from S. alopecuroides was 6.98% while the predicted value was 7.03%,their relative error was 0.71%. Conclusion: Response surface methodology was effective to optimize alcohol precipitation technology of polysaccharides from S. alopecuroides.

    • In vitro Transdermal Test of Yinhu Ganmao Spongia

      Abstract:Objective: To investigate in vitro transdermal absorption properties of Yinhu Ganmao spongia. Method: The content of chlorogenic acid was determined by HPLC and GC determination of linalool content,release was detected by Franz diffusion cell method with 10% ethanol as receiving liquid,transdermal receiving liquid at different time points was collected,and calculated cumulative permeation amount and ratio. Result: Regression equations of cumulative permeation amount of chlorogenic acid and linalool were Q=7.143 8t+8.229 4(r=0.996 1),Q=125.375t-143.705(r=0.989 8),respectively.J of them were 7.144,125.375 μg·cm-2·h-1,respectively. Conclusion: Chlorogenic acid in Yinhu Ganmao spongia continuous permeated uniformly within 24 h,but linalool almost permeated compeletly in 12 h;This method was simple and rapid,it was a ideal method to comprehensive evaluate in vitro transdermal performances of Yinhu Ganmao spongia.

    • Effects of Grinding Degree of Medicinal Materials and Soaking Time on Ginsenoside Rg1 Content and Total Solid Content in Shiyifang Vinum

      Abstract:Objective: To investigate effects of different grinding degree of medicinal materials and soaking time on ginsenoside Rg1 content and total solid content in Shiyifang vinum. Method: With ginsenoside Rg1 as index,which was determined by HPLC,ginsenoside Rg1 content and total solid content changes in Shiyifang vinum were investigated after soaked different time(4,6,8 months) and crushed into coarse particles(5-10 mm),the most coarse powder(2 mm),coarse powder(0.85 mm). Result: After medicinal materials were crushed into coarse particles,the content of ginsenoside Rg1 was the highest in Shiyifang vinum.Total solid content was similar between crushed into coarse particles,immersed 6 months and crushed into coarse particles,immersed 8 months,there was no significant difference. Conclusion: Quality of Shiyifang vinum was good by crushing medicinal materials into coarse particles and soaking 6 months.

    • Preparation and Quality Conrol of Andrographolide Liposomes

      Abstract:Objective: To prepare andrographolide liposomes and establish its quality control method. Method: Andrographolide liposomes was prepared by film dispersion-ultrasound method,with encapsulation efficiency as index,orthogonal test was adopted to investigate effects of ratio of soybean lecithin-cholesterol,ratio of soybean lecithin-andrographolide,the amount of vitamin E and ultrasonic time on preparation technology of andrographolide liposomes.Particle size and morphology of these prepared liposomes was assessed.The content of andrographolide was determined by HPLC. Result: Optimum preparation technology was as following:ratio of soybean lecithin-cholesterin 6: 1,ratio of soybean lecithin-andrographolide 4: 1,the amount of vitamin E 20 mg,ultrasonic time 5 min.Morphology of prepared liposome showed sphere structure with uniform diameter.The average particle size was 625.8 nm and encapsulation efficiency was 81.35%,rate of leakage was 1.54%.The linear range of andrographolide was 8.16-48.96 mg·L-1(r=0.999 8). Conclusion: Prepared andrographolide liposomes had high entrapment efficiency and low leakage rate,this established method for quality control was reliable,which lay foundation for preparing andrographolide liposomes aerosol.

    • Optimization of Purification Technology for Total Flavonoids and Cyanogenetic Glycosides in Leaves of Sorbus tianschanica by Macroporous Resin

      Abstract:Objective: To optimize purification technology of total flavonaoids and cyanogentic glycosides in leaves of Sorbus tianschanica by macroporous resin. Method: With absorption capacity,absorption rate and desorption rate as indexes,5 different macroporous resins were screened by static adsorption test,effects of the concentration of sample solution,ratio of diameter to height,adsorption velocity,the concentration of ethanol,elution velocity and other process parameters on purification technology of total flavonoids and cyanogenetic glycosides in leaves of S. tianschanica were investigated by single factor tests. Result: HPD-100 resin was adopted,its optimum purification technology was as following:absorption capacities of crude drugs to resin 2.2 g·g-1,the concentration of sample solution 0.1 g·mL-1 with ratio of diameter to height at 1: 6,adsorption velocity at 2 BV·h-1,standing for 30 min,eluted with 4 BV 50% ethanol,elution velocity 2 BV·h-1,collected eluent. Conclusion: HPD-100 resin could be used successfully to purify total flavonaoids and cyanogentic glycosides in leaves of S. tianschanica with the mass fraction of effective part in ethanol eluate was more than 50%.

    • Investigation of Factors Influencing Stability of Five-dimensional Lysine Oral Solution

      Abstract:Objective: To investigate effects of factors on stability of five-dimensional lysine oral solution. Method: With content changes of vitamin C as index,single factor tests were adopted to investigate influence of antioxidants,water and pH to stability of five-dimensional lysine oral liquid. Result: The content of vitamin C in five-dimensional lysine oral solution plus antioxidants was higher than the blank heating group without antioxidants,adding single antioxidant was higher than adding composite antioxidant,content changes was large before and after heating.The boiling water group was stable by comparing with the often water group.Stability of prepared oral liquid weakened by increasing of pH. Conclusion: Stability of five-dimensional lysine oral solution was fine by adding antioxidants with boil water and pH 3.6.

    • Analysis of Cleansing Mechanism of Aconitum kusnezoffii

      Abstract:Objective: To analyze cleansing mechanism of Aconitum kusnezoffii. Method: HPLC was adopted to determine the content of mesaconine,aconitine,hypaconitine,benzoylmesaconine,benzoylaconitine,benzoylhypaconitine in A. kusnezoffii with stems,A. kusnezoffii without stems and stems of A. kusnezoffii. Result: The total content of three kind of diester alkaloids in A. kusnezoffii with stems,A. kusnezoffii without stems and stems of A. kusnezoffii was 0.59%-0.66%,0.61%-0.69%,0.39%-0.42%,respectively;The total content of three kind of monoester alkaloids was 0.09%-0.10%,0.09%-0.12%,0.06%-0.08%,respectively.Except six alkaloids chromatographic peaks,there was another common peak(peak 4),area of peak 4 in stems of A. kusnezoffii was 55%-80% to that in A. kusnezoffii without stems. Conclusion: Content of six alkaloids was found in A. kusnezoffii without stems and stems of A. kusnezoffii was different.Stems of A. kusnezoffii was the non-medicinal parts whose content of six alkaloids was low,it could be removed.

    • Effects of Different Pretreatment Methods on Vacuum Reflux Extraction Technology of Epimedium brevicornum

      Abstract:Objective: To investigate effects of different pretreatment methods on vacuum reflux extraction efficiency of Epimedium brevicornum. Method: Taking yield of icariin as index,vacuum reflux extraction technology of E. brevicornum was optimized by orthogonal test with vacuum degree,extraction time,temperature and times as factors,single factor tests were adopted to investigate effects of soaking time and crushing degree on vacuum extraction efficiency of E. brevicornum. Result: Optimum vacuum extraction technology was as following:extracted 3 times with 10 times the amount of 60% ethanol,20 min each time,vacuum degree 0.07 MPa.Yield of icariin increased by 9.5% after E. brevicornum crushed into fine powder,it was 7.9% after crushed into coarse powder,and extract yield increased by 2.6%,1.7%,respectively.After soaked different time,yield of icariin increased by 6.4% and dry extract yield increased by 6.9% at most,soaked 20 min had optimal extraction efficiency,but coarse powder and fine powder had no significant differences. Conclusion: Yield of active ingredients and dry extract rate of E. brevicornum would increase under different pretreatment methods.

    • On-line Determination of Free Radicals Scavenging Effect of SMS and YQFM by HPLC-ABTS-DAD

      Abstract:Objective: To compare the vitro antioxidant components of Shengmaisan (SMS) and Yiqi Fumai sterile powder (YQFM). Method: The scavenging activities of SMS and YQFM for ABTS·+ radicals was determined by HPLC-ABTS-DAD. Result: SMS had a good effect of scavenging ABTS·+ radicals with the saccharides, amino acids and lignans as potential active components responsible for its scavenging activities. The scavenging activities of YQFM were relatively weak. Conclusion: Although SMS and YQFM are the different preparations for the same complex prescription, they have large differences in directly scavenging radicals.

    • Content Determination of Total Flavonoids in Resina Draconis by Colorimetric Method

      Abstract:Objective: To study the colorimetric determination of total flavonoids in Resina Draconis. Method: Comparison on the difference of absorption curve was done using the characteristics that total flavonoids of Resina Draconis can produce color with various chromogenic reagents. Result: Resina Draconis colored by HCl-Mg power produce a large absorption peak at 475 nm where the interference is small. It had good stability within 6 h and showed a good linear relationship in the range of 0.012-0.12 g·L-1 (r=0.999 3). And its absorption peak was not much change after colored by other chromogenic agent. Conclusion: AlCl3, NaNO2-Al(NO3)3-NaOH colorimetries usually used for determination of total flavonoids were not suitable for Resina Draconis flavones assay. HCl-Mg powder can be used as chromogenic agent for colorimetric determination of the total flavonoids in Resina draconis. But it still needs to find a suitable reference substance.

    • Determination of Eight Nucleosides in Rhizoma Arisaema, Rhizoma Pinellie and Rhizoma Typhonii

      Abstract:Objective: To establish an HPLC method for simultaneous determination of eight nucleosides in Rhizoma Arisaema, Rhizoma Pinellie and Rhizoma Typhonii. Method: The chromatographic separation was achieved on a Phenomenex Luna PFP (2) (4.6 mm×250 mm, 5 μm) column with methanol-water solution as mobile phase (gradient elution). The flow rate was 0.8 mL·min-1, the column temperature was maintained at 35℃ and the detective wavelength was set at 260 nm. Result: The linear ranges of uracil, hypoxanthine, xanthine, uridine, inosine, guanosine, thymidine and adenosine were 0.004 34-0.434 μg (r=0.999 9), 0.002 34-0.234 μg (r=0.999 9), 0.004 07-0.407 μg (r=0.999 7), 0.010 90-1.090 μg (r=0.999 9), 0.002 01-0.201 μg (r=0.999 9), 0.007 32-0.732 μg (r=0.999 9), 0.001 22-0.122 μg (r=0.999 7), 0.002 07-0.207 μg (r=0.999 9) respectively. The average recoveries of the eight nucleosides were 104.2%, 98.7%, 103.7%, 99.4%, 101.8%, 102.9%, 98.3%, 102.3% and RSDs were 2.1%, 2.4%, 3.0%, 1.4%, 2.0%, 1.9%,2.8%, 2.7% respectively. Conclusion: The method is simple, accurate and rapid. It can be used for determination of nucleosides in Rhizoma Arisaema, Rhizoma Pinellie and Rhizoma Typhonii.

    • Determination of Total Polysaccharides in Different Growth Phase of Puerariae Lobatae Radix

      Abstract:Objective: To determine the total polysaccharides in different growth phase and different years of Puerariae lobatae Radix. Method: Total polysaccharides content was also determined by pheno-sulfuric acid method using dextrose as standard sample. Result: Glucose was linear in the range of 0.020-0.142 mg (r=0.999 9). The average recovery rate was 99.34% (RSD 1.82%). The content of total polysaccharides appeared regular changes and the content of total polysaccharides was highest in August and September of perennial Puerariae Lobatae Radix. Conclusion: The result might provide scientific basis for havesting of Puerariae Lobatae Radix.

      Abstract:Objective: To establish a fingerprint analysis method for identification of Akebiae Fructus by HPLC-ELSD, get the control chromatogram and compare the fingerprints of Akebiae Fructus collected from different producing areas so as to establish a sensitive and specific method for controlling the quality of Akebiae Fructus. Method: The HPLC-ELSD fingerprints of Akebiae Fructus from 10 different places were obtained. The HPLC separation was performed on a Cosmosil-C18 column (4.6 mm×250 mm, 5 μm) in gradient elute mode with a mixture consisting of acetonitrile and 0.2% formic acid water at the flow rate of 1.0 mL·min-1. The ELSD detector is used and the drift tube temperature of ELSD was set at 110℃ with the flow rate of 3.0 L·min-1. The data were analyzed by Fingerprint Similarity Evaluation Software to compare the similarity of the samples from different habitats. Result: Fingerprint spectrum of Akebiae Fructus was established. 10 samples were detected and 14 peaks in the chromatogram were common. There was a high similarity and each chromatographic peak was obtained with good separation. And the correlation could meet the technical requirements of fingerprint of Chinese traditional medicine. And four peaks in HPLC-ELSD chromatogram were firstly assigned as 3-O-β-D-glucopyranosyl (1→3)-α-L-rhamnopyranosyl-(1→2)-α-L-arabinopyranosyl hederagenin (saponins X), 3-O-β-D-xylopyranosyl-(1→2)-α-L-arabinopyranosyl hederagenin (saponins B), 3-O-α-L-rhamnopyranosyl-(1→2)-α-L-arabinopyranosyl hederagenin (saponins PD), 3-O-α-L-arabinopyranoside hederagenin (saponins A). Conclusion: This method shows high precision and reliability and the fingerprint spectrum provides a scientific basis for controlling the quality of Akebiae Fructus.

      Abstract:Objective: To establish the quality standard of Peitu Qingxin granules. Method: TLC was used for the qualitative identification of Imperatae Rhizoma, Forsythiae Fructus, Dictamni Cortex and Glycyrrhizae Radix et Rhizoma. HPLC was used to determine the content of phillyrin, which was obtained by a Yilite Hypersil ODS2(4.6 mm×250 mm, 5 μm) column with the mobile phase composed of acetonitrile-water(25: 75), and the column temperature was maintained at 25℃. The flow rate was 1.0 mL·min-1. The detection wavelength was set at 277 nm. Result: TLC spots were clear and well-separated without negative interference. The linear range of phillyrin was 0.179-4.29 μg (r=0.999 97) with an average recovery of 100.67% (RSD 1.03%, n=9). Conclusion: The method is simple, accurate and reproducible, which can be used for the quality control for Peitu Qingxin granules.

      Abstract:Objective: To establish the fingerprint of Shixiao San, enhance the quality control of the method. Method: The RP-HPLC was performed on an C18 chromatography column, with acetonitrile-0.1% of TFA as mobile phase, the flow rate was 1 mL·min-1; the wavelength was set at 360 nm. The temperature of column was maintained at 30℃. Result: The common modes for the HPLC fingerprints were set up by ensuring 15 common peaks in the chromatograms. The similarity degrees in the ten batches were in the range of 0.962 to 0.996. Conclusion: The method is sensitive, accurate and simple, thereby it can be used specifically for the quality control of Shixiao San.

      Abstract:Objective: According to the regulations about quality requirements and the experience of traditional classification of Chinese medicine Yinpian, combined with the modern testing technology, an innovative concept of Yinpian classification was put forward with Fupian (Heishunpian) selected for model practice. Method: Twenty batches of Heishunpian of different origins were graded according to the principle of 'three-step' hierarchical concept. The first step:the indicators of pharmacopoeia, such as traits, inspection, content determination and so on were adopted to determine the general level; the second step:size (length, width) and color (outer color E*ab, inner color E*ab) were employed to determine the first level; the third step:the geoherbalism and the more advanced inspection items were adopted as indicators to determine the best level. Result: Use of the research ideas and methods of classification, 3 batches of samples were removed from 20 batches because of its disqualification. Three levels were divided:2 batches were the best level; 7 batches were the first level; 8 batches were the general level. Conclusion: In this paper, the thought of Yinpian's classification and methods could divide the level of Heishunpian, and provide a basis for the classification of other varieties of Heishunpian and a reference for the thought of classification and technique of traditional Chinese Yinpian.

    • Study on Dissolution Test of Breviscapine Pill

      Abstract:Objective: To establish a method for determination of the dissolution of Breviscapine Pill. Method: Agilent TC-C18 column (4.6 mm×250 mm, 5 μm) was used to separate the target components. The mobile phase was composed of methanol-tetrahydrofuran-0.1% phosphate solution (14:14:72). The flow rate was 0.8 mL·min-1 and the detection wavelength was set at 335 nm. Phosphate BS (pH 6.8) was used as solvent. Paddle method was used with rotation rate at 50 r·min-1. Scutellarin content was assayed by HPLC and the release percentage was then calculated. Result: The linear range of scutellarin was 20.8-416 μg·L-1 (r=0.999 9) with an average recovery of 99.92% (RSD 0.43%). The accumulation release percentage in 20 minutes was not lower than 70% of the content of scutellarin. Conclusion: The differences of the accumulation release percentage were shown in identical batch and the different batches samples. The method is repeatable for the quality control of Breviscapine Pill.

      Abstract:Objective: To set up the method for simultaneous determination of four active compounds in Chimonanthus nitens by HPLC. Method: The chromatography separation was performed on the Elite hypersil ODS2 (4.6 mm×250 mm, 5 μm) with gradient elution. The mixture of acetonitrile and water as the mobile phases at a flow rate of 1.0 mL·min-1 was used, the detective wavelength was set at 344 nm and the column temperature was kept at 40℃. Result: The calibration curve was linear in the range of 5.665-56.65 mg·L-1 for scopoletin (r=0.999 9), the average recovery was 97.06% and RSD was 1.62%. The calibration curve was linear in the range of 2.431 2-24.312 mg·L-1 for isofraxidin (r=0.999 9), the average recovery was 102.86% and RSD was 0.93%. The standard curve presented a linear range from 2.444-24.44 mg·L-1 for scoparone (r=0.999 9), the average recovery was 102.18% and RSD was 1.81%. The standard curve presented a linear range from 9.012-90.12 mg·L-1 for rutin (r=0.999 2), the average recovery was 104.44% and RSD was 4.2%. Conclusion: The method is available with a good reproducibility, sensitive and simple and could be used to control the quality of C. nitens.

    • Determination of Alkaloids in Phellodendri Amurensis Cortex by HPLC

      Abstract:Objective: HPLC was used to measure the content of berberine, jatrorrhizine and palmatine in Phellodendri Amurensis Cortex from ten different areas of Heilongjiang Province simultaneously. Method: A Kromasil C18 column (4.6 mm×250 mm, 5 μm) was used with the mobile phase consisting of acetonitril-0.1% phosphonic acid (25: 75, 0.10 g sodium lauryl sulfate was added per 100 mL). The flow rate was 1 mL·min-1; the detecting wavelength was set at 345 nm, and the column temperature was maintained at 30℃. Result: The calibration curves of berberine hydrochloride, jatrorrhizine hydrochloride and palmatine hydrochloride were linear within the ranges of 12.08-144.96 mg·L-1 (r=0.999 3), 1.60-25.60 mg·L-1 (r=0.999 3), 7.26-87.12 mg·L-1 (r=0.999 3) respectively. The average recoveries (n=9) were 102.51%, 103.47%, 102.89%, respectively. Conclusion: This method is proved to be convenient, reliable and accurate, it can be used in content determination of berberine, jatrorrhizine, palmatine in Phellodendri Amurensis Cortex. There are significant differences in the Alkaloid content in Phellodendri Amurensis Cortex from different areas.

      Abstract:Objective: To study quality standard of Coptis chinensis in the Qingshen granule. Method: Thin-layer chromatography was used to identidy C. chinensis.The content of berberine hydroc hloride was determined by HPLC.The HPLC separation was performed on kromasil-C18 column (4.6 mm×250 mm,5 μm) with acetonitrile-0.033 mol·L-1KH2PO4(25: 75) as mobile phase; the detection wavelength was set at 345 nm; the flow rate was 1.0 mL·min-1, and the column temperature was set at 30℃. Result: The spot in the TLC was clear,with no interference from negative control. In the HPLC experiments of precision, reproducibility and stability, the samples could be separated entirely. The linear range of berberinehydroc hloride was 0.028-0.892 μg(r=0.999 9), the average recovery was 100.16% with RSD of 1.87%(n=6). Conclusion: The methods are accurate and quick in qualitative identification and quantitative assay of this preparation,which can be used for the quality control of Qingshen granule.

      Abstract:Objective: To evaluate the quality of Rabdosia rubescens from different varieties collected in Jiyuan of Henan Province. Method: According to the Chinese Pharmacopoeia (2010 edition),HPLC was used to investigate the content of oridonin in 34 different varieties with oridonin as control. The analytical column was a Agilent TC-C18(4.6 mm×250 mm,5 μm),mobile phase:methanol-water(55: 45), UV detection wavelength 239 nm, the flow rate was 1 mL·min-1, and the temperature of column was 25℃. The regression equation was Y=1 006X+9.046(r=0.999 9),which showed the good linear relation in the range of 0.488-0.88 μg, and the average recovery was 98.80%, the repetitive RSD was 2.17%. Result: There were significant differences in the content of oridonin of R. rubescens from different varieties,the minimum content was 0.63%,and the highest content was 1.98%.The content of oridonin in all samples accorded with the pharmacopeia standards. Conclusion: The oridonin was a main active ingredient in this plant,it could be used as an important indicator for the quality evaluation.This method of quality evaluation was simple, reproducible and which had a high degree of separation.It provided scientific basis for the assessment of the quality of R. rubescens.

Method: Catalpol and acteoside were reflux extraction with methanol and determined by HPLC. The chromatographic column was Dikma Diamonsil C18(4.6 mm×250 mm,5 μm); the flow rate was 1.0 mL·min-1;the column temperature was kept at 30℃;the mobile phase consisted of acetonitrile-0.1% phosphoric acid solution(1:99) and the detection wavelength was set at 210 nm for catalpol;the mobiles phase consisted of acetonitrile-0.1% acetic acid solution(16:84) and the detection wavelength was set at 334 nm for acteoside. Result: The content of catalpol and acteoside was different in different varieties from genuine producing area and the differences also exist in the fresh and dried R. glutinosa. Conclusion: The quality of varieties of R. glutinosa was different and it was necessary to screen the optimal variety.

      Abstract:Objective: To establish the quality standards of Pingweishu capsules. Method: Coptis chinensis and Corydalis Rhizoma were identified by TLC. The content of berberine hydrochloride was determined by RP-HPLC. Result: The spots in the TLC were fairly clear, and no interference was shown in the blank samples. Berberine hydrochloride showed a good linear relationship within a range of 0.368-3.68 μg (r=0.999 8). The average rate of recovery was 99.86% (RSD 1.46%). Conclusion: The method is a simple, rapid and accurate analysis assay, which could be used for quality control of Pingweishu capsules.

      Abstract:Objective: To establish the method to determine the concentration of hyperin in Compound Hypericum Perforatum Dropping Pills. Method: HPLC was used for the determination of hyperin in Compound Hypericum Perforatum. A COSMOSIL packed column 5 C18-MS-Ⅱ (4.6 mm×250 mm, 5 μm) was used. The mobile phase was acetonitrile-water (0.5% glacial acetic acid and 2% tetrahydrofuran) (13:87). The detection wavelength was set at 360 nm. Result: The linear range for hyperin was 10.8-54.0 mg·L-1 (r=0.999 1). The average recovery was 98.31% (RSD 1.23%). Conclusion: The method can control the quality of Compound Hypericum Perforatum Dropping Pills effectively. It was sensitive, accurate and feasible.

Method: The column temperature was controlled by a program with capillary column HP-5MS and the MS analysis was performed with EI and quadruple mass analyzer. The chemical composition was identified by NIST08 and Wiley275 library, and their relative contents were determined by using normalization method of chromatographic peak areas. Result: There were 19 common compounds in A. kravanh from various habitats which were eucalyptol, α-caryophyllene,β-pinene, α-pinene,α-cubebene, o-cymene. Conclusion: The main constituents were different in various habitats.

Method: High performance liquid chromatography and Dikma Diamonsil C18 column (4.6 mm×250 mm, 5 μm) was adopted with methanol-water (containing 0.1% phosphoric acid) as the mobile phase in gradient elution form. The column temperature was set at 35℃, and the flow rate was 0.8 mL·min-1. The detection wavelength was set at 270 nm. The sample size was 20 μL. Chromatographic peaks were identified by standard substance comparison. Result: Good precision, stability and repeatability were proved. Five standard substances were adopted for qualitative determination of chromatographic peaks during the 34 total common peaks, and their similarities were above 0.968. Conclusion: The study lays a foundation for the overall quality evaluation of Guilubushen Pills.

      Abstract:Objective: To study the chemical constituents of Petasites japonicus. Method: Mutiple kinds of column chromatographies were used in the isolation procedure. The structures of the isolated compounds were elucidated by spectral methods. Result: Nine compounds were isolated from P. japonicus, including mucusoside (1), dihydrodehydrodiconiferyl alcohol (2), n-butyl acetylglutamate (3), uracil (4), 3α,6α-dihydroxyeremophilenolide (5), petasiphenol (6), caffeic acid (7), β-sitosterol (8),β-daucosterol (9). Conclusion: Compounds 1-4 were isolated from genus Petasites and P. japonicus for the first time.

      Abstract:Objective: To study the volatiles from Elsholtzia splendens and Molsa chinese. Method: The volatile chemical constituents from E.splendens and M. chinensis. were analysed by HS-SPME-GC-MS. Result: Thirty-nine and forty-six components from E.splendens and M. chinese, which accounted for 70.55% and 92.44% of the peak areas, were separated and identified. Conclusion: The volatile chemical constituents have high application value in medicine and chemical industry and mainly consisted of alkane, olefin, alcohol, ketone, phenol, organic acid, etc. The present experiments provide a scientific basis for the further exploitation and utilization of this herb.

      Abstract:Objective: To clarify the interaction mechanism of astragaloside IV and cycloastragenol with bovine serum albumin(BSA) by fluorescence spectroscopy. Method: Scanning the fluorescence quenching and synchronous spectrum through adding astragaloside IV and cycloastragenol to the solution of BSA. Result: The fluorescence quenching mechanism of astragaloside IV and cycloastragenol was static quenching. Under 293 K, the binding constants (KA) of astragaloside IV and cycloastragenol were 1.35×104 and 8.51×104L·mol-1, respectively. And the numbers of binding sites were 0.726 4 and 0.731 2, respectively. Under 310 K, the binding constants of astragaloside IV and cycloastragenol were both decreased. The thermodynamic parameters of astragaloside IV and cycloastragenol with BSA were ΔH<0, ΔS>0, and ΔG<0, the interaction of astragaloside IV and cycloastragenol with BSA were driven mainly by electrostatic interaction. The results of synchronous spectrum showed that both the astragaloside IV and cycloastragenol affected the space conformation of tryptophane. But the space conformation of tyrosine was not affected by astragaloside IV and cycloastragenol. Conclusion: The interaction mechanism of astragaloside IV and cycloastragenol with BSA was clarified by fluorescence spectroscopy. It can supply theoretical supports for the rational administration of astragaloside IV and cycloastragenol.

      Abstract:Objective: To compare the differences of the chemical constitutions of Frutus Aurantii Immaturus of different origin Places. Method: By one-dimensional FT-IR spectroscopy, second derivative spectra and two-dimensional related spectral analysis of IR spectra, the features of these spectrum peaks of Frutus Aurantii Immaturus were identified. Result: In one-dimensional map, the raw material of Frutus Aurantii Immaturus from 5 different origins had the similar peak shape among the rang of 400-4 000 cm-1,and 7 common peaks. Frutus Aurantii Immaturus from each origin place had the same chemical content. In the second derivative spectra the characteristic peaks of Frutus Aurantii Immaturus from Yunnan were less and blunter, the more peaks of Frutus Aurantii Immaturus from Jiangsu and Jiangxi were found, and the shapes of peaks were sharp. The two-dimensional related spectral map showed there were obvious differences between the peaks of Frutus Aurantii Immaturus from 5 different origin places. Conclusion: This IR spectra technique can identify Frutus Aurantii Immaturusof different origin places.

      Abstract:Objective: To study the chemical constituents and bioactivity of essential oil from Dasymaschalon rostratum. Method: The essential oil was extracted by hydrodistillation and separated by capillary GC. The chemical constituents were determined by normalization and were identified by MS. Result: Ninty-one chromatographic peaks were detected and seventy-three compounds were identified, which was 90.25% of the total essential oil. The essential oil from D. rostratum showed inhibitory activity against Staphyloccocus aureus Rosenbach,Escherichia coli,Bacillus subtilis, exhibited higher activity to restrain Leukemia cells (K-562) and Liver cancer cells(BEL-7402), with IC50 values of 10.47,47.2 mg·L-1 respectively. Conclusion: It is first time to extract volatile oil from Dasymaschalon and identify the compunds, most of which were alkene compunds. Many active ingredients were contained in the essential oil from D. rostratum, the experiment basis were provided for the further research of D. rostratum.

      Abstract:Objective: To establish a FTIR-Raman analytic method of the ginkgolide B. Method: The ginkgolide B was extracted by fat extractor, and was purified by column chromatography, and was tested by the IR and Raman spectrometry after drying, and the spectrograms were compared with the FTIR spectrogram and Raman spectrogram of ginkgolide B reference substance which was treated with processing method of the sample. Result: The peak shape and peak position were consistent with the sample spectrogram and reference spectrogram, the IR spectrogram and Raman spectrogram can complement each other, so data are complete. Conclusion: The method is accurate and reliable, it can be used for the identification of substances.

      Abstract:Objective: To analyze the change rules of carbohydrate content in Ginseng Radix et Rhizoma root, stem and leaf from different growth period. Method: Acquisition Ginseng Radix et Rhizoma root, stem and leaf samples from the spreading stage, the flowering stage, the fruit growth stage, the root growing after fruit stage and the withering stage, ultraviolet spectrophotometry was adopted to detect the content of total sugar, reducing sugar and soluble polysaccharide in Ginseng Radix et Rhizoma. Result: The content of total sugar and reducing sugar in Ginseng Radix et Rhizoma root reached the maximum value at the fruit growth initial stage, the soluble polysaccharide content reached the maximum value at the fruit growth later stage; the total sugar, reducing sugar and soluble polysaccharide content in Ginseng Radix et Rhizoma stem and leaf had no significant change at the throughout growth stages. Conclusion: The content of total sugar, reducing sugar and soluble polysaccharide in Ginseng Radix et Rhizoma have their own variation at the different growth period, and may be closely related to the plant morphogenesis and material metabolism.

      Abstract:Objective: To establish X-ray diffraction characteristic fingerprint of Natrii Sulfas Exsiccatus. Method: X-ray diffraction method was applied to do qualitative analysis of 10 batches of Natrii Sulfas Exsiccatus,correlation coefficients and angle cosine were applied to calculate similarity of shared peak in each sample. Result: Similarity of shared peak in X-ray diffraction of Natrii Sulfas Exsiccatus was>95%. Conclusion: This X-ray diffraction characteristic fingerprint of Natrii Sulfas Exsiccatus had strong specificity,it could provide a reference for identification and quality evaluation of Natrii Sulfas Exsiccatus.

      Abstract:Objective: To investigate relativity between the content of rutin and traditional specification grade of Farfarae Flos. Method: The content of rutin from Farfarae Flos was performed by HPLC on Dikma Diamonsil C18 column (4.6 mm×250 mm,5 μm) with acetonitrile-1%acetic acid (20:80) as mobile phase and detection wavelength at 254 nm.HPLC fingerprint and similarity analysis of different specifications of Farfarae Flos was performed on Dikma Diamonsil C18 column (4.6 mm×250 mm,5 μm) with acetonitrile-0.1%H3PO4 gradient elution and detection wavelength of 240 nm. Result: Average content of rutin in superior grade was slightly higher than gradeless samples with little difference;Similarity of fingerprint of Farfarae Flos from different grades,habitats and batches was more than 0.85.Average values of twelve relative peak area of superior samples were larger than that of gradeless samples. Conclusion: Quantification method of rutin and fingerprint analysis was feasible and reproductive.Traditional classification of Farfarae Flos had a certain rationality,but the content of rutin had little correlation with traditional classification,so to classificate Farfarae Flos with the content of rutin as main index was unreasonable.

      Abstract:Objective: To establish a method for determining the content of Pb,Cd,As,Hg,Cu and Cr in Cervi Cornus Colla. Method: Microwave digestion and inductively coupled plasma-atomic wmission spectrometry (ICP-AES) were adopted to determine the content of Pb,Cd,As,Hg,Cu and Cr in 11 batches of Cervi Cornus Colla.Microwave digestion conditions were as followings:0-3 min,1 600 W,110℃;3-6 min,1 600 W,150℃;6-16 min,1 600 W,185℃.Determination conditions of ICP-AES were:incident power of 1.4 kW,argon as cooling gas with flow rate of 12 L·min-1,flow rate of carrier gas and auxiliary with 1,0.8 L·min-1,volume of sample enhance 1 mL·min-1. Result: Detection limit of Pb,Cd,As,Hg,Cu and Cr were 0.15,0.01,0.27,0.08,0.03,0.03 mg·kg-1,respectively.recoveries was between 90%-112%,precision and repeatability were in line with detection requirments.Cd,As and Hg were not detected in each sample,apart from sample from F was not detected Pb,Pb,Cu and Cr were detected in other samples,the content of Cr was high in some samples. Conclusion: This method was accurate,fast and suitable for determination of harmful elements in glue herbs.

    • Determination of Macroporous Resin Residues in Extract of Swertia pseudochinensis by Headspace Gas Chromatography

      Abstract:Objective: To Establish a method for detecting residues of macroporous resin in extract of Swertia pseudochinensis. Method: Headspace-GC was used to determine the content of macroporous resin residues of n-hexane,benzene,methylbenzene,dimethylbenzene,phenylethylene,diethylenzene,divinylbenzene in extract of S. pseudochinensis. Result: All of seven residues were separated and good linear responses were obtained in the mass concentration with r between 0.994 0 to 0.999 7.The average recoveries of these 7 residues were in the range of 93.4%-98.4% with RSD<10%,these 7 residues were not detected in three batches of samples. Conclusion: This analytical method was convenient and accurate with good repeatability,it could be used to determine organic solvents residues in macroporous resin.

      Abstract:Objective: To develop an ultra performance liquid chromatography tandem mass (UPLC-MS-MS) method for determination of notoginsenoside R1, gensenoside Rg1, and gensenoside Rb1 in Chinese formula KLW. Method: The Chinese formula KLW is extracted by ultrasonic and centrifugal, then filtered before test. The ultra performance liquid chromatographic separation was performed on a Waters ACQUITY BEH C18 column (2.1 mm×50 mm, 1.7 μm) by gradient elution with methanol-water as the mobile phase at a flow rate of 0.4 mL·min-1. The analysis of 3 compounds was performed under ESI multiple reaction monitoring (MRM) mode, and the quantification was conducted by external calibration method. The monitoring ions for notoginsenoside R1 were m/z 1 007.2/423.3 m/z; gensenoside Rg1 were 875.2/423.5 m/z; gensenoside Rb1 were 1 183.3/487.3 m/z. Result: The method showed a good linearity in the range of 0.05-2.0 mg·L-1, with the correlation coefficient above 0.999.The average recoveries (n=6) of notoginsenoside R1, ginsenoside Rg1 and ginsenoside Rb1 were 97.7%, 96.3%, 97.1% respectively. Conclusion: This method is simple, rapid, accurate and sensitive, can also provide a reliable method for clinical research and can be used for the quality control of Chinese formula KLW.

    • Simultaneous Determination of Retusine and Pogostone in Pogostemon cablin by HPLC

      Abstract:Objective: To establish a high performance liquid chromatography (HPLC) method for simultaneous determination of retusine and pogostone in Pogostemon cablin, and analyze the quality of 11 batches of retusine and pogostone in P. cablin. Method: The RP-HPLC system consisted of a Phenomenex Luna C18 column (4.6 mm×250 mm, 5 μm) and the mobile phase of acetonitrile-formic acid by isocratic elution. The flow rate was set at 0.8 mL·min-1 and detected at 310 nm; the column temperature was kept at 30℃. Result: The linear regression for retusine and pogostone was obtained in the range of 0.020-0.36 μg (r=1.000 0) and 0.025-0.40 μg (r=1.000 0) respectively; the average recovery was 98.57% (RSD 2.06%) and 100.04% (RSD 2.66%) respectively. The content of retusine and pogostone in 11 batches of P. cablin were in the range of 1.13-8.72 mg·g-1 and 1.19-4.38 mg·g-1 respectively. Conclusion: The method is quick, simple and reproducible for the determination of retusine and pogostone in P. cablin simultaneously.

      Abstract:Objective: To establish a method to examine the limited content of aristolochic acid A in Qingxueneixiao Pills. Method: 1% sodium carbonate solution is repeatedly heated to dissolve the sample, extracted with chloroform for several times. Discard the chloroform layer, adjusted the pH of the alkaline layer with dilute hydrochloric acid, and extracted with chloroform again for several times. Collect and evaporate the chloroform layer. The residue was dissolved in methanol, 1 mL of which was diluted to 20 mL, eluted with a gradient composed by acetonitrile-water (40: 60), water (0.1% formic acid+5 mmol·L-1 ammonium formate )/( %) and acetonitrile (containing 5% water, 0.1% formic acid+5 mmol·L-1 ammonium formate )/( %), assayed in high performance liquid chromatography/electrospray ionization mass spectrometry multiple reaction monitoring (MRM) mode. Result: Aristolochic acid A was linear int the range of 11.21-299.0 pg (r=0.999 5).The average recovery was 119.2% (n=9) and the RSD was 4.0%. Conclusion: This method is accurate, rapid, simple, reproducible, less interference and specific, appropriate to the trace detection for the Qingxueneixiao Pills.

      Abstract:Objective: To establish an effective HPLC method for determining the content of artemitin and chrysosptertin B in Laggera pterodonta. Method: By HPLC, a Diamonsil C18 column (4.6 mm×250 mm, 5 μm) was adopted with methanol-water (60: 40) as the mobile phase for elution at the flow rate of 1.0 mL·min-1. The detection wavelength was set at 257 nm and the temperature was kept at 32.2℃. Result: Good linearity was obtained in the range of 0.448-2.24 and 0.352-1.76 μg for artemitin and chrysosptertin B respectively. The method recoveries of artemitin and chrysosptertin B were 103.48% and 103.42%, RSDs were 0.87% and 0.91% respectively. Conclusion: The method is simple, accurate and suitable for the determination of artemitin and chrysosptertin B, which can be used for quality control of Laggera pterodonta.

      Abstract:Objective: A gas chromatographic (GC) method for ethylene glycol and diethylene glycol determination in glycerol samples was established. Method: 6% cyanopropyl-phenyl and 94% dimethyl polysiloxane capillary column (DB-624) was used. The internal standard method with temperature-programmed mode was used for quantitative analysis. Result: The sample peak area and the internal standard peak area ratio to the concentration of samples for both the ethylene glycol and the diethylene glycol had a good linear relation in the range of 20.00-160.00 mg·L-1. The linear equation of ethylene glycol was YEG=0.004X-0.025 (r=0.999 8). The average recovery was 101.80% and RSD was 1.6% (n=9).The linear equation of diethylene glycol was YDEG=0.004 2X-0.045 7 (r=0.999 8). The average recovery was 98.95% and RSD was 1.9% (n=9). Conclusion: The method has a short peak retention time, which can be used for quality control of the glycerol.

      Abstract:Objective: To establish a method for determining the content of naringin and naringenin in rat liver microsomes by UPLC,and preliminary investigation in vitro metabolism of two ingredients at different times points. Method: Determination was performed on UPLC chromatographic system,ACQUITY BEH C18 column (3.0 mm×100 mm,1.7 μm) with a mobile phase of methanol-0.1% acetic acid and gradient elution at a flow rate of 0.5 mL·min-1,detection wavelength was 283,289 nm,column temperature was 30℃,with quercetin as an internal standard,injection volume was 3 μm. Result: Naringin,naringenin and internal standard quercetin had good separation degree and no endogenous interference.The linear range of naringin and naringenin were 3.814-38.143(r=0.999 2),3.586-35.857 mg·L-1(r=0.999 6),respectively.Method recoveries of naringin ranged from 95.43% to 97.95% and absolute recoveries ranged from 97.33% to 98.18%.Method recoveries of naringenin ranged from 95.27% to 99.31% and absolute recoveries ranged from 97.71% to 99.73%.I phase metabolism of naringenin in in vitro liver microsomes was more significant than naringin. Conclusion: This method was rapid,sensitive and accurate,it could be used for determination of naringin and naringenin in rat liver microsomes.

      Abstract:Objective: To prepare nanostructured lipid carriers modified with mPEG2000-DSPE(mPEG2000-DSPE-NMD-NLC) and investigate its properties and in vivo pharmacokinetics in rats. Method: mPEG2000-DSPE-NMD-NLC were prepared by melt ultrasonication method,morphology was observed by TEM,particle size,entrapment efficiency,drug loading and Zeta potential were determined,in vitro release was examined.Plasma concentrations of nimodipine at different time point in rats were determined by HPLC,with nimodipine injection as control,pharmacokinetics parameters of mPEG2000-DSPE-NMD-NLC in rats were calculated. Result: Prepared mPEG2000-DSPE-NMD-NLC showed spherical with uniform size and rounded surface;Particle size was about 85 nm,Zeta potential was (-12.77±0.15) mV,entrapment efficiency and drug loading were (97.66±0.45)% and (3.36±1.53)%,respectively;Cumulative release of mPEG2000-DSPE-NMD-NLC was 20.03% within 0-6 h,and 43.06% at 24 h.Pharmacokinetic parameters were as follows:t1/2=21.65 h,MRT=21.09 h,CL=557.30 mL·h-1·kg-1,AUC(0-t)=6 411.96 μg·h2·L-1. Conclusion: Particle size distribution of mPEG2000-DSPE-NMD-NLC was uniform,its showed obvious in vitro and in vivo sustained-release characteristic.This study could lay foundation for improving clinical efficacy of nimodipine.

    • Effect of Glycyrrhetinic Acid on the Plasma Concentration of Kaempferol in Rats

      Abstract:Objective: To study the effect of glycyrrhetinic acid on the plasma concentration of kaempferol in rats. Method: Seventy-eight SD rats,weighing 150-210 g,half male and half female, were distributed into 4 groups,including control group of high concentration and low concentration,experimental group of high concentration and low concentration. Then each group was divided into three sub-groups, namely group of distribution phase (1 h), equilibrium phase (2 h) and elimination phase (3 h). Experimental group of high concentration and low concentration was administered by gavage at a dose of 50 mg·kg-1 kaempferol mixed with 50 mg·kg-1 grycyrrhetinic acid and 25 mg·kg-1 mixed with 50 mg·kg-1 grycyrrhetinic acid,respectively. The control group was administered by gavage at a same dose like the experimental group except grycyrrhetinic acid. Plasma samples were collected by picking eyeballs 1, 2, 3 h after administration. Plasma was hydrolyzed at 90℃ for 120 min with hydrochloric acid. Then they were extracted with acetonitrile and concentration of kaempferol was determined by HPLC. Result: No free kaempferol was detected in rat's plasma after administration of kaempferol. The results showed that at two concentration levels there was no significant difference between two sets data of experimental group and control group, but there was significant difference (P<0.05) between two group at high concentration level in plasma(P<0.05). Conclusion: Grycyrrhetinic acid can affect the pharmacokinetics of kaempferol in plasma at high concentration level.

    • Effects of Gualou Xiebai Decoction on Myocardial Ischemia-reperfusion Injury

      Abstract:Objective: To investigate the curative effect of Gualou Xiebai decoction on physiology, serology, histology target of myocardial ischemia-reperfusion injury. Method: The ischemic model was made by occluding the left anterior descending artery (LAD) in rats. Not only the change of ST segment was observed, but also the activity of creatine kinase-MB (CK-MB),lactate dehydrogenase (LDH) and superoxide dismutase (SOD) and maleic dialdehyde (MDA) in serum were measured. The histological changes were observed. Result: Compared with the model group, the T amplitude of wave was lowered apparently during the period of 10 to 30 min in Gualou Xiebai decoction group. Compared with the model group, MAD level in Gualou Xiebai decoction was reduced(P<0.05), and the level of SOD was increased(P<0.05). Histopathological results showed that the high dose group was superior to the low dose group. Conclusion: Gualou Xiebai decoction has a protective effect on myocardial ischemia-reperfusion injury.

    • Effects of Wuzi Jiangtang Fang on the GLUT4 Expression in Skeleton Muscle of Type 2 Diabetic Rats

      Abstract:Objective: To observe the effects of Wuzi Jiangtang Fang on blood glucose and the glucose transporter 4(GLUT4) expression in skeleton muscle of type 2 diabetic model rats to explore the molecular mechanism of improving glucose metabolism. Method: The rat model of type 2 diabetes was established by feeding with high-fat diet for 6 weeks and intraperitoneal injection of small dose of streptozotocin(STZ). The diabetic rats were randomly divided into model group administrated with purified water, metformin group with metformin suspension, Wuzi Jiangtang Fang groups with water extracts of different doses respectively. Meanwhile, normal group was treated with purified water. The dose was 7.5 mL·kg-1 of body weight every day. The interventions were administrated by ig for 6 weeks. The fasting blood glucose and 2 h postprandial blood glucose were detected at the beginning of the treatment, and at the ends of the second, the fourth and the sixth week during the 6 weeks' treatment period respectively. At the end of the sixth week, the skeleton muscle was taken for the measurement of GLUT4 protein expression. Result: Compared with the normal group, the blood glucose levels of type 2 diabetes mellitus(T2DM) rats were elevated and the GLUT4 expression of skeleton muscle was decreased in T2DM model rats, with significant differences in all of the items above (P<0.05); Wuzi Jiangtang Fang reduced the fasting blood glucose and 2 h postprandial blood glucose levels of T2DM model rats, and improved the GLUT4 expression in skeleton muscle of T2DM model rats. The differences regarding the item examined above all had statistical significance (P<0.05). Conclusion: Wuzi Jiangtang Fang can decrease the fasting blood glucose and 2 h postprandial blood glucose of T2DM model rats, which may be related with the improvement of GLUT4 expression in skeleton muscle of T2DM model rats.

    • Preventive Effect of Haobie Yangyin Ruajian Prescription on Hepatic Fibrosis Induced by CCl4

      Abstract:Objective: To study the preventive effects of Haobie Yangyin Ruajian prescription (HBYYRJ) on rat hepatic fibrosis induced in rats by CCl4 and explore the effective mechanisms. Method: Rats were randomly divided into 7 groups:normal control group, model group, high,middle and low dose of HBYYRJ groups(8.20,2.59,0.82 g·kg-1), compound Biejia Ruangan table group(0.54 g·kg-1), colchicin group(1×10-4 g·kg-1). First CCl4 5 mL·kg-1 was given by sc, 40% CCl4-peanut oil 3 mL·kg-1 was given by sc every three days from the second time to the twelfth time; 30% alcohol (1 mL) was ig given to model group and therapeutic groups every other day, drugs were given to the rats in therapeutic groups for six consecutive weeks, distilled water was given to the rats of normal and model groups. Histological observation of the liver tissue and collagen content were carried out. Blood serum was used for measuring the contents of alanine transaminase(ALT), aspartate aminotransferase(AST),hyaluronic acid(HA),laminin(LN), type III procollagen(PCIII),type IV collagen(IVC), albumin(ALB). The content of malondialdehyde (MDA) was detected by ultraviolet visible spectrophotometry. The content of hydroxyproline,collagen and albumin were detected in liver tissue. Result: The degeneration and necrosis,inflammatory cell infiltration,false lobular formation,and a large number of collagen fiber depositions were observed in liver tissue of the model group.The level of chronic hepatic damage was improved greatly and none of the false lobular was formed in the high dose of HBYYRJ. The contents of ALT,AST,HA and PCIII of the model group were significantly higher than that in normal group (P<0.01,P<0.05). The content of ALT,AST,HA and PCIII of HBYYRJ groups was significantly lower than that in model group (P<0.05). The content of MDA and collagen in liver tissue of HBYYRJ groups was lower than that in model group (P<0.01). The content of ALB of the high dose of HBYYRJ was significantly higher than that in model group (P<0.01). Conclusion: HBYYRJ has preventive effects on hepatic fibrosis in rats induced by CCl4.The mechanism might be associated with anti-lipid peroxidation, antiinflammation and restraining the compose of collagen protein.

    • Effect of Beta-asarone on Adriamycin-induced Cardiomyocyte Injury in Suckling Mouse

      Abstract:Objective: To observe the effect of beta-asarone on adriamycin-induced cardiomyocyte injury in suckling mouse. Method: Cardiomyocytes of neonate rat were cultivated for 72 hours and randomly divided into normal control group, adriamycin(ADR,2 mg·L-1) model group,and beta-asarone(10,20,40 mg·L-1) dose groups. MTT colorimetric method was deployed to detect cardiocyte survival rate,activities of medium lactate dehydrogenase(LDH), superoxide dismutase(SOD) and malondialdehyde(MDA) were detected,and protein expression of B cell lymphoma/lewkmia-2(Bcl-2) and Bcl-2 associated X protein(Bax) were detected by immune histochemical method. Result: Compared with normal control group, model group ADR cardiomyocyte shrinkage deformation,irregular,beat rhythm irregularities,frequency obviously slow down, numbers of survival cells were decreased, the leakage of LDH was increased; SOD activity was decreased; the contents of MDA were increased; protein expressions of Bcl-2 were reduce, protein expressions of Bax were increased (P <0.01 or P <0.05). And compared with ADR model groupand ADR model group, each dose group of beta-asarone had no changes in cardiomyocytes cell pulsation frequency, present centrality synchronization pulsing; cardiomyocytes survival rate was increased; the leakages of LDH was decreased;and the activity of SOD was increased; contents of LDH and MDA were decreased; protein expression of Bcl-2 was increased, protein expression of Bax were reduced (P<0.01 or P <0.05). Conclusion: Beta-asarone protects mice from adriamycin-induced cardio-toxicity by reduction of oxygen free radicals and apoptosis in mouse cardiac tissues.

      Abstract:Objective: To study the effect of Xinfukang(XFK)oral liquid on MM isoenzyme of creatine kinase(CK-MM) mRNA and CK-MM protein expression level in rat of heart failure model after myocardial infarction. Method: The rat heart failure mode after myocardial infarction was created by ligating the left anterior descending coronary arteries. All the rats were randomly divided into 4 groups:sham group, model group, XFK 5.4 g·kg-1 group and captopril 0.1 g·kg-1 group. After 24 hours of modeling, rats were ig given XFK and captopril for 4 weeks, Cardiac function, myocardial ultrastructure and key enzymes of myocardial energy shuttle, CK-MM mRNA and protein expression level were separately measured in all groups. Result: ①Compared with model group, in captopril group and XFK group CK-MM mRNA and protein expression increased (P<0.01). ②Compared with captopril group, in XFK group CK-MM mRNA expression level decreased (P<0.05). In XFK group CK-MM protein expression has no significant change. Conclusion: XFK oral liquid can effectively increase the expression level of CK-MM mRNA, as well as the protein content of CK-MM to improve myocardial energy shuttle and remodel the process of myocardial energy metabolism function.

      Abstract:Objective: To study the effect and mechanism of Tianshu capsule on the migraine's mice, which was composed of Gastrodiae Rhizoma and Ligustici Chuanxiong Rhizoma. Method: Model was building on ICR mice after subcutaneous injection of reserpine 0.1 mg·kg-1·d-1×12 d, control group mice were injected 0.9% sodium chloride injection, treatment were started on the seventh day of modeling for 6 consecutive days, dose were 1.55, 4.65 g·kg-1. The pain threshold was tested on the twelve day, the clotting time was tested next day and concentration of serotonin (5-HT), noradrenaline(NA) in the brain.was detected. Zolmitriptan tablets were used as a positive durg. Result: Compared with model group, the tianshu capsule could increase the pain threshold(P<0.01), prolong the clotting time(P<0.01), increase the concentration of 5-HT and NA in brain(P<0.05,P<0.01). Compared with positive drug, the capsule could increase the concentration of 5-HT(P<0.05). Conclusion: The capsule can treat the migraine's mice by balancing release of 5-HT, inhibiting vascular dilatation of migraine attacks.

    • Effect of Platelet Rich Plasma Combined with Bone Marrow Mesenchymal Stem Cells in Treatment of Rabbit Femoral Head Necrosis

      Abstract:Objective: To study effect of platelet rich plasma(PRP) combined with bone marrow mesenchymal stem cells(BMSCs) in treatment of rabbit femoral head necrosis. Method: Fifty rabbits were randomly divided into normal group, model group, PRP group (3%), BMSCs group (1×106/mL) and combination group. The model of rabbit femoral head necrosis was made by operation. After 4 weeks of operation, the PRP group was implanted the PRP, the BMSCs group was implanted the BMSCs, the combination group implanted the PRP and BMSCs. Normal group was not treated. The rabbit femoral head necrosis were observed by HE trichrome coloration after 12 weeks of operation. The change of the trabeculae of bone, bone cells in the bone,fat cells were observed by light microscopy. Then PDGF protein in the rabbit femoral head was tested by Western blot. Result: Compared with the normal group, the trabecular of bone became thinner, decreased in bone cells,fat cells increased, empty osteocyte lacuna in the trabeculae were increased in model group (P<0.05). Compared with the model group, in all treatment groups trabecular bone became thicker, bone cells increased, fat cells decrease, empty osteocyte lacuna in the trabeculae were decreased and improved the expression of PDGF in the rabbit femoral head (P<0.05). Conclusion: PRP combined with BMSCs can effectively induce bone regeneration, improved the expression of PDGF and improve bone tissue recovery.

      Abstract:Objective: To explore the effect of vinegar processing Cyperus rotundus on the expression and function of P-glycoprotein(P-gp)in Caco-2 cells. Method: The function of P-gp in Caco-2 cells was measured by FCM and the expression was by immunohistochemistry after the interaction between the cell(2×105/mL) and the medicine of concentration(5%,10%,20%). Result: The low, medium, high concentration of crude C. rotundus could increased 8.25%,9.57%,16.84% of the accumulation of digaoxi,vinegar processing C. rotundus could increase 22.44%,29.12%,33.09% of the accumulation of digaoxi.Serum containing crude and vinegar processing C. rotundus could remarkedly increase the accumulation of digaoxi and decrease the positive rate of P-gp's expression. Conclusion: C. rotundus after vinegar processed can promote the function of inhibiting the expression and function of P-gp in Caco-2 cells.

      Abstract:Objective: To explore the enhancing effect on killing hepatocarcinoma cells of CBRH7919 by herpes simplex virus thymidine kinase and ganciclovir (HSV-tk/GCV) system together with constituents absorbed into blood after oral administration of Liuwei Dihuang Wan. Method: MTT assay was used to detect growth inhibition of CBRH7919 after being treated with tk/GCV system and constituents including loganin,morroniside,5-(methoxymethyl)-2-furoic acid,sweroside,paeonol and mixed drugs of these five compounds; also to detect growth inhibition of CBRH7919 after being treated with tk/GCV system and lymphocytic supernatant fluid activated by constituents. Result: There was no synergy killing effect on CBRH7919 between tk/GCV system and constituents; while there was synergy killing effect on CBRH7919 treated with tk/GCV system lymphocytic supernatant fluid activated by constituents together, witch showed significantly dose-effect relationship. Conclusion: The active substrate activated by constituents of Liuwei Dihuang Wan has synergy effect on HSV-tk/GCV system,and the endogenous active substrate may be the material basis of synergy effect of Liuwei Dihuang Wan on HSV-tk/GCV system.

      Abstract:Objective: To observe influence of of Dihuang Yizi (DHYZ) on serotonin 1 A receptor (5-HT1AR) and serotonin 2 A receptor (5-HT2AR) mRNA express in poststroke depression rats, and further discuss mechanism. Method: The sixty healthy adult SD rats were randomly divided into 6 groups including blank control group, model group, the optimal solutions group, DHYZ high dose group, medium group, low group. Afterpoststroke depression model was established the corresponding drugs were given (hydrochloric acid prozac,DHYZ 450,150,75 mg·kg-1. The real-time fluorescent quantitative RT-PCR method was used to assay 5-HT1AR and 5-HT2AR mRNA expression. Result: Compared with model group; DHYZ medium (0.76± 0.13), (1.21±0.21) and high dose groups (0.75±0.1), (11.25±0.26) increased the 5-HT1AR mRNA expression, and decreased the 5-HT2AR mRNA expression (P<0.05). Conclusion: DHYZ may raise 5-HT1AR mRNA, and decrease 5-HT2AR mRNA, which can explain mechanism of DHYZ in treatment of poststroke depression.

      Abstract:Objective: To study the protection effect of Polygonatium sibiricum polysaccharide(PSP) on cisplatin-induced hepatotoxicity. Method: Forty Sprague-Dawley rats were randomly divided into normal control group,model group,the low dosage group,the middle dosage group and the high dosage group of PSP, with eight rats in each group. The model rats with hepatotoxicity were duplicated with injection of cisplatin(5 mg·kg-1). The rats were injected with cisplatin(5 mg·kg-1) after fifth days rats in the low, middle and high dosage group of PSP were treated by PSP 10,20,40 g·kg-1 once a day. After 10 days of administration with PSP, the changes of liver coefficient were examined and the blood alanine aminotrans ferase(ALT), aspartate amino transferasen(AST), the activity of superoxide dismutase(SOD), glutathione peroxidase(GSH-Px) and the content of malondialdehyde(MDA) was measured. Result: Compared with the normal control group, contents of ALT, AST in serum were significantly increased in each treatment group(P<0.01), the content of MDA was significantly increased(P<0.01) and the activity of SOD and GSH-Px was significantly decreased(P<0.01). Compared with the model group, contents of ALT, AST in serum were significantly decreased in each treatment group(P<0.05 or P<0.01), the content of MDA was significantly decreased(P<0.05 or P<0.01) and the activity of SOD and GSH-Px was significantly increased(P<0.05 or P<0.01). Conclusion: PSP may improve the function in liver injury rats induced by cisplatin and inhibite the oxidative damage of liver.

    • Protective Effects of Alcohol Extract of Kadsura heteroclita on Acute Hepatic Injury Induced by Carbon Tetrachloride in Rats

      Abstract:Objective: To observe the protective effects of alcohol extract of Kadsura heteroclita (Roxb) Craib on acute hepatic injury induced by carbon tetrachloride in rats. Method: SD rats were randomly divided into the normal group, model group, bifendate group, and alcohol extract of K. heteroclita low, middle, high dosage groups. In sixth day, other rats were injected subcutaneously with CCl4 except the normal group with the same volume of saline after they were performed by gavage once a day and lasted for 7 days. After injected 24 hours, the contents of alanine aminotransferase(ALT) and aspartate aminotransferase(AST) in serum, the liver index and contents of reduced glutathione(GSH) in liver tissue were measured and the liver pathological changs were observed. Result: The alcohol extract of K. heteroclita decreased significantly the levels of serum ALT and AST and liver index, and increased the level of GSH (P<0.05 or P<0.01). The pathological changes were relieved in the treatment groups. Conclusion: The alcohol extract of K. heteroclita has protective effects on acute hepatic injury induced by carbon tetrachloride in rats.

    • Preliminary Study on Soothing Liver and Choleretic Effects of Bupleurum chinense and its Processed Products

      Abstract:Objective: To explore the impact of different processing methods on soothing liver and choleretic effects of Bupleurum chinense. Method: Wistar rats were randomly divided into normal, model, health products, vinegar products, wine products, honey products and positive control group. Chronic hepatic injury model of rat was induced by 40%CC14.Then rats were treated with B. chinense and its processed products (both for 5 g·kg-1) for 8 weeks. Alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), total bilirubin (TBiL), Albumin (Alb) and total protein (TP) levels were measured, and of the albumin and globulin ratio (A/G) was calculated. The common bile duct was incubated, and the flow of bile was detected. Result: ALT, AST, ALP and TBiL contents in B. chinense vinegar product group was reduced significantly (P<0.01); honey product ranked the second place (P<0.05); ALT, AST and ALP contents of health product and wine product groups had a lowered effect (P<0.05). TP, Alb and A/G contents in vinegar product group was increased significantly (P<0.01 or P<0.05); Honey product increased TP and Alb contents (P<0.01 or P<0.05); health product and wine product increased TP content (P<0.05). The bile flow of vinegar product group increased significantly (P<0.05). Conclusion: The soothing liver and choleretic effects of B. chinense vinegar product are the strongest, honey product is the second, and wine product and health product effect is fair.

    • Pharmacodynamics and Nasul Ciliotoxicity of ZhiBing Nasal Cavity In-situ Gels

      Abstract:Objective: To study the pharmacodynamics and ciliotoxicity of Zhibing nasal cavity in-situ gels (ZBISG). Method: Fifty mice were randomly divided into five groups, normal saline (0.8 g·kg-1) were given for blank group,aspirin (0.04 g·kg-1) was given orally for the pasitive control group and 0.4,0.8,1.2 g·kg-1 ZBISG given orally for ZBISG groups. Five minutes after administration, 0.6% acetate solution (0.1 mL·10 g-1) was injected intraperitoneally in control group and ZBISG groups. Then the time of writhing response appeared and the number of mice writhing response were observed and recorded. Analgesic rate was also calculated. The pain threshold increase rate was measured at before and 15,30,60,90 min after ZBISG administration using hot plate test. The nasal ciliotoxicity of ZBISG was evaluated with in vivo toad palate, normal saline of 0.5 mL was used as the control. Result: For the mice torsion reaction, there was significant differences between treatment group and blank group(P<0.01). Between ZBISG treatment group and aspirin group, the high dose group and medium dose group were significantly different compared with that of aspirin group(P<0.01), however low dose group did not show difference to aspirin group. And as the dosage increased, the effect of analgesic action increased. In the time when the writhing response appeared, there was significant differences between administration group and blank group(P<0.01). And as the dosage increased, the effect of analgesic action increased. There was obvious difference in the pain threshold increase rate at each time point between ZBISG group and blank group(P<0.01) or between ZBISG treatment group and aspirin group(P<0.01) or between the high, medium dose group and low dose group(P<0.01).ZBISG had no obvious effect on the ciliary movement. Conclusion: ZBISG is a kind of effective and safe preparation.

    • Root of Actinidia chinensis Planch Induces Anoikis in Colon Carcinoma RKO Cells

      Abstract:Objective: To observe the effects of Tengligen (root of Actinidia chinensis Planch) on cell growth in suspension and anoikis in human colon carcinoma RKO cells. Method: RKO cells were treated with Tengligen (200-800 mg·L-1) or same volume of RPMI1640.Cell growth in suspension was detected with CCK-8 assay. Anoikis was visualized by EthD-1 staining. Caspase-3 activities were detected by colorimetric assay. Intracellular reactive oxygen species (ROS) production was observed by 2', 7'-dichlorofluorescin diacetate staining. Result: 200-800 mg·L-1 of Tengligen, significantly inhibited RKO cell growth in suspension in a dose dependent manner (P<0.01). After 200-800 mg·L-1 of treatment, EthD-1 was absorbed by suspension-cultured RKO cells to yield a red-fluorescent nuclear staining, and nuclear fragmentation was observed in partial cells. The EthD-1 fluorescence in 200-800 mg·L-1 of Tengligen treated RKO cells were compared with controls (P<0.01). 200-800 mg·L-1 of Tengligen also activated caspase-3 and upregulated intracellular ROS generation (P<0.01). Conclusion: Tengligen could inhibit RKO cell growth in suspension, induce anoikis in RKO cells, and may associate with caspase-3 activation and ROS production.

      Abstract:Objective: To observe the therapeutic effect of Qingre Jiedu decoction (QJD) and find the bacteriological mechanism on septic model in rats. Method: The septic model was induced by cecal ligation and puncture. The experimental rats were divided into three groups:control group, septic model group and QJD (24 g·kg-1) treated group. The survival and the bacterial populations of ascites and blood in experimental sepsis treated with QJD were determined by bacterial culture and real time PCR. Result: The populations of conditioned pathogen such as Escherichia coli and Staphylococcus aureus in QJD treated group were decreased significantly compared with that of model group(P<0.01). Conclusion: Conditioned pathogen may be primary pathogen of septic rats in our study. QJD are contribute to improve survival in septic model by suppressing proliferation of conditioned pathogen.

    • Pharmacological Comparison between Traditional Decoction and Machine Decoction of Sijunzi Tang

      Abstract:Objective: To compare the pharmacological effect of three Sijunzi Tang adopted different processing technology. Method: The pharmacodynamic differences for three decoctions of Sijunzi Tang processed by different technics were compared, using two kinds of spleen-deficiency model. One model was induced by Pendulifory Elshortzia. The rats were randomly divided into the model group, the blank control group, Sijunzi Tang groups with high, medium and low doses (10, 5, 2.5 g·kg-1), Sijunzi Tang adopted the high pressure processing with high, medium and low doses (10,5, 2.5 g·kg-1), Sijunzi Tang adopted the atmospheric pressure processing with high, medium and low doses (10,5, 2.5 g·kg-1), and cisapride as the positive control group. The other model was induced by reserpin. The mice were randomly divided into the model group, the blank control group, Sijunzi Tang adopted the traditional decoction processing with high, medium and low doses (13.2, 6.6, 3.3 g·kg-1), Sijunzi Tang adopted the high pressure processing with high, medium and low doses (13.2, 6.6, 3.3 g·kg-1), Sijunzi Tang adopted the atmospheric pressure processing with high, medium and low doses (13.2, 6.6, 3.3 g·kg-1).Xiangsha Yangwei pills were used as positive control. The criteria included general physical signs, D-xylose excretion rate, amylase activity, serum gastrin content and indexes of organs determined before and after treatment. Result: After treatment with Sijunzi Tang, the spleen symptoms of spleen-deficiency animals were improved, and the final data analysis showed that Sijunzi Tang could significantly improve spleen-deficiency symptoms. The indicators showed that Sijunzi Tang adopted the traditional processing was slightly better than that adopted the machine processing. However, there was no significant difference among the three Sijunzi Tang adopted different processing technics for the therapeutic effect on the spleen-deficiency animals. Conclusion: The three processing technics of Sijunzi Tang show similar therapeutic effect for the spleen-deficiency animals.

    • Effect of Compound Phenytoin Sodium Gel on Restoration of the Wound’s Surface and Bacterial Infection

      Abstract:Objective: To observe the influence of compound phenytoin sodium gel (CPSG) for experimental animals on restoration of the wound's surface and bacterial infection. Method: The incised wound model of mice was established, CPSG (4.25,4.55 g·kg-1)was external applied for 12 days and the wound contraction rate were observed after treatment for 3, 6,8,10, 12 days. Rats incised wound model accompanied by staphylococcus aureus were replicated, CPSG (1.70,1.82 g·kg-1)was external applied for 12 d, and the wound recovery rate were detected after treatment for 3, 6,8,10,12 days. The histopathological changes of wound healing tissue were examined. Result: Compared with the wound contraction rate of gel group (16.77±4.79)%, (29.95±5.63)%,(50.69±4.19)%,(67.47±5.13)%,(82.48±4.40)%, those in low and high doses of CPSG were (20.16±5.78)%,(40.02±9.79)%,(71.04±7.27)%,(96.14±2.53)%,(99.44±1.30)%;(28.50±4.77)%,(55.66±8.39)%,(79.67±5.11)%,(96.14±2.53)%,(100.00±0.00)% respectively(P<0.05,P<0.01,P<0.01,P<0.01. P<0.01,P<0.01,P<0.01,P<0.01,P<0.01)after treatment for 3, 6,8,10,12 days in mice. The wound recovery rate of the low and high dose of CPSG were (29.45±5.11)%,(50.17±4.92)%,(72.48±5.05)%, (87.44±4.11)%,(96.66±3.75)%;(33.71±3.43)%,(58.67±5.59)%,(81.86±4.15)%,(95.92±2.39)%,(100.00±0.00)% higher than (20.71±3.58)%,(40.42±3.89)%,(58.53±6.21)%,(74.71±9.23)%,(86.47±7.15)% of gel group(all P<0.01)in the treatment of 3, 6, 8,10, 12 days. Conclusion: CPSG could promote the restoration of the wound's surface in the animals with experimental trauma and bacterial infection.

    • Effects of Compound Cordyceps Militaris on Immunity Function and Lymphocyte Proliferation in Spleen of Mice

      Abstract:Objective: To investigate the influence of compound cordyceps militaris granules on the immunity function of mice. Method: Mice were randomized into the normal group, levamisole hydrochloride group and compound cordyceps militaris granules group with 0.355,0.71,2.13,4.26,8.52,12.78 g·kg-1, and dinitrofluorobenzene induced delayed type hypersensitivity test (DTH) and carbon clearance test in vivo were employed to evaluate the immunologic function of mice. Compound cordyceps militaris granules containing serum on effects of lipopolysaccharide (LPS) and concanavalin A (ConA) induced lymphocyte proliferation was detected by MTT test. Result: The compound cordyceps militaris granules given by ig 0.355 g·kg-1 could significantly potentiate DTH in mice (P<0.01), and 2.13 g·kg-1 of the granules was significantly enhanced the DTH in mice (P<0.05). Carbon clearance test showed that compound cordyceps militaris granules given by ig (0.71, 0.355 g·kg-1) could evidently improve the carbon clearance indexes (P<0.05).Lymphocyte proliferation in 2.13 g·kg-1 compound cordyceps militaris containing serum group was significantly enhanced comparing with control group (P<0.01), and 4.26,8.52,12.78 g·kg-1 compound cordyceps militaris containing serum group was significantly enhanced comparing with control group (P<0.05). Compound cordyceps militaris contaming serum 4.26 g·kg-1 could enhance ConA-induced T cell proliferation (P<0.01). Compound cordyceps militaris contaming serum 12.78 g·kg-1 could improve the proliferation of LPS-induced B lymphocyte obviously(P<0.05). Conclusion: Compound cordyceps militaris granules can enhance immune function obviously.

    • Safety Evaluation of Three Traditional Chinese Medicine Injections

      Abstract:Objective: Evaluate thesafety of Xueshuantong Injection, Kudiezi Injection and Xiyanping Injection. Method: Active systemic anaphylaxis (ASA) test, passive cutaneous anaphylaxis (PCA) test, vascular irritation test and In vitro hemolysis test were used to evaluate the safety of three traditional Chinese medicine injections (TCMI). Result: In the ASA test, positive control group showed strong anaphylaxis effect, three TCMI didn't produce allergic reaction and body weight of all groups showed no significant differences. In the PCA test, all rats of positive control group caused blue spots of which the diameters were all greater than 5 mm and the IgE were significantly increased. However, three TCMI didn't produce blue spots of which the diameters were greater than 5 mm and the IgE levels showed no significant differences compared with negative control group. In the vascular irritation test, local blood vessels by near the administration site of both left and right ears of the animals did not observe irritative reaction symptoms, pathology results showed that after administration and recovery phase, blood vessels and surrounding tissues of auresunitas of the animals didn't appear apparent abnormality. In vitro hemolysis test showed that no haemolysis or agglutination phenomenon were induced by any of the three TCMI. Conclusion: Xueshuantong Injection, Kudiezi Injection and XiyanpingInjection were safety under these experimental conditions.

    • Study on the Effect of Bamboo Leaves Extract on Free Radical Metabolism of Rats Experiencing Exhaustive Exercise

      Abstract:Objective: To broaden the application field of bamboo leaf extract and enrich the type of anti-fatigue supplements for sports training, the effect of bamboo leaf extract on free radical metabolism of rats experiencing high-intensity endurance training were studied. Method: Twenty-four SD rats were randomly divided into quiet control group, sport control group and dosed sport group, with 8 rats in each. Sport control group experienced 6 week treadmill training. Dosed sport group experienced 6 week treadmill training and were fed 300 mg·kg-1·d-1 bamboo leaf extract suspension. Quiet control group and sport control group were fed with the same volume of distilled water. After six weeks, the content or activity of Superoxide Dismutase(SOD), Total Antioxidant Capacity(T-AOC), catalase(CAT), Malondialdehyde(MDA), Reduced Glutathione(GSH), Glutathione Peroxidase(GSH-Px) in heart, liver and kidney of all groups and sport ability of sport control group and dosed sport group were measured. Result: The content or activity of SOD, T-AOC, CAT, GSH and GSH-Px in heart, liver and kidney significantly decrease and MDA content significantly increase, but taking bamboo leaf extract can inhibit the downward trend of antioxidant enzyme activities, reduce the content of MDA and increase exhaustive exercise time by 28.87%. Conclusion: Taking bamboo leaf extract can promote the metabolism of free radicals in rats experiencing high intensity exercise and improve their athletic ability. So it is suitable as raw material of anti-exercise fatigue supplements.

      Abstract:Objective: To investigate the effects of beta-asarone on the behavior and the expressions of mitogen-activated protein kinase phosphatase-1(MKP-1),extracellular signal-regulated kinase(ERK), mitogen and stress activated protein kinase-1(MSK-1), cAMP response element binding protein(CREB), B cell Lymphomal/Leukemia-2(Bcl-2) in the hippocampus of depression model rats. Method: Sixty adult Sprague-Dawley rats were randomly divided into 4 groups:the normal control group (NC), the model control group(MC), the β-asarone group(A), the fluoxetine control group (FC), 15 in each one. Except those in the NC, the rest rats were singly housed and exposed on an unpredicted sequence of mild stressor. From the second day,A and FC group rats were administered with corresponding medicinal liquid everyday (1.2 mg·kg-1·d-1 to the FC group, 25 mg·kg-1·d-1 to the A group) by gastro-gavage for 21 days. The rats' body weight, sucrose consumption volume in the sucrose preference test,and times of grooming in the Open-field-test were detected on the 1,7,14,21th day, respectively. The protein expressions of MKP-1 and Bcl-2 in the hippocampus were detected by immunohistochemical assay, while Real-time PCR for quantitative analysis of MKP-1, MSK-1, CREB and Bcl-2. Result: Compared with the MC group, the scores of A group and FC group in the open field test increased; the protein expressions of MKP-1 in the hippocampus decreased while MSK-1, CREB and Bcl-2 increased with statistical significance(P<0.05). Conclusion: Beta-asarone could obviously improve the depressive state of the model rats. Its mechanism might be correlated with decreasing the protein expressions of MKP-1, increasing MSK-1, CREB and Bcl-2 in the hippocampus.

      Abstract:Objective: Study on the mechanism of Zhishi Xiaopi Wan treatment on Gastric cancer from cell proliferation, apoptosis and cycle. Method: Complete medium was mixed in negative control group;1×104 cells/wells were seeded at 96 wells plate and divided into negative control, Zhishi Xiaopi Wan in 0.05,0.125,0.5,2,4 g·L-1 concentration groups;the cells were treated with drugs for 24,48,72 h, and cell proliferation was detected by MTT method; 1×105 cells/wells were seeded in 6 wells plate, and the drugs of low(0.125 g·L-1),medium(0.5 g·L-1),high(2 g·L-1) concentration respectively were put into wells for 48,72 h, then the cell cycle and apoptosis of gastric cancer SGC-7901 cells were detected by flow cytometry(FCM). Result: Gastric cancer SGC-7901 cells were treated by the 0.125,0.5,2 g·L-1 of Zhishi Xiaopi Wan, and its inhibition rate is in dose and time dependent. Zhishi Xiaopi Wan could increase SGC-7901 cells in G0-G1 phase percentage and promote apoptosis; The high,medium concentration was stronger than the lows. Conclusion: Zhishi Xiaopi Wan can inhibit the proliferation of SGC-7901 cell and there is in dose and time dependent, which mechanism may be related to drug block of cells to G0/G1 phase and induce cell apoptosis.

      Abstract:Objective: To study the uptake status of Guanfu Base I(GFI)in Caco-2 ceils. Method: To establish Liquid Chromotography with Mass Spectrometry(LC-MS) assay, A LC procedure was carried out using an C18(150 mm×2.0 mm,5 μm)and an eluate of 0.2% acetic acid-acetonitrile (30:70)at the flow rate of 0.2 mL·min-1.The MS portion was set with ESI ionsource in the positive ion polarity mode and the selective ion monitoring(SIM).A protonated molecules[M+H] were detected at m/z 388.10 for GFI.The GFI concentrations in Caco-2 cells at various time,concentration,temperature and pH values were measured. Result: The linearity of GFI concentrationcurve was found in the range of 0.2-50.0 μmol·L-1 (r=0.996 7).The minimal detection limit was 0.2 μmol·L-1.The GFI uptakes in Caco-2 cells obeyed a passive diffusion in a time,concentration and pH-dependent manner. Conclusion: This sensitive and specific LC-MS assay can be applied to determine GFI concentrations in Caco-2 cells.

      Abstract:Objective: To Study effect of ethanol extract from Persicae Seman on spatial learning memory and anti-fatigue in dementia mice induced by AlCl3. Method: The mouse dementia model was induced by AlCl3(200 mg·kg-1,ig,40 d). Morris water maze test and the fatigue instrument were used to observe effect of ethanol extract malondial dehyde(MDA) contents and a cetylcholinesterase(AchE) activity in brain tissue were measured. Result: The ethanol extract from Persicae Semen(2.8 g·kg-1) could significantly increase traversing times in 1 min platform. It can could improveanti-fatigue effect, and decrease MDA contents and AchE activity in brain tissue. Conclusion: The ethanol extract from Persicae Semen has improvement effect and anti-fatigue effect on spatial learning memory and anti-fatigue in dementia mice. Its mechanism maybe related to MDA contents and AchE activity.

      Abstract:Objective: To explore effect of Fuzi Lizhong decoction on the expression of hypothalamic orexin mRNA and hypcthalamopitutary-adrenol(HPA) axis in insomnia rats with spleen Yang deficiency syndrome and therapeutic mechanism. Method: Handred rats were randomly divided into control group, model group,low-dose group, middle-dose group, and high-dose group 5 groups. each group included 20 rats. Model rats were ip p-chlorophenylalanine(PCPA) according to 400 mg·kg-1, once a day for two days to establish insomnia model. low-dose group, middle-dose group, and high-dose group were ig given Fuzi Lizhong decoction respectively with 10,20,40 g·kg-1 based on model group for 7 days. Result: Compared with control group, expression of orexin mRNA of model group increased significantly (P<0.01).Compared with control group, content of 5-hydroxy tryptamine(5-HT) of model group reduced significantly (P<0.01).Compared with control group, content of corticotropin-releasing bormone(CRH) of model group increased (P<0.05). Compared with control group, content of adrenocorticotropic hormone(ACTH) of model group increased significantly(P<0.01). Compared with model group, expression of orexin mRNA of treatment group reduced significantly(P<0.01). Compared with model group,content of 5-HT of treatment group increased significantly (P<0.01)and content of 5-HT in middle-dose group increased (P<0.05). Compared with model group,content of CRH of treatment group reduced (P<0.05). Compared with model group,content of ACTH of treatment group reduced significantly (P<0.01). Conclusion: Fuzi Lizhong decoction can reduce expression of Orexin mRNA and so that inhibit HPA axis to reduce stress reaction, which perhaps is one of mechanisms for insomnia treatment.

      Abstract:Objective: To study the efficacy and safety of Mongolian SA dew de Di+Xieri GA-4 soup in the treatment of early diabetic nephropathy (Rexingxijin disease). Method: According to digital method,80 patients with divided into two groups randomly treatment group and control group,40 cases each. Treatment group on the basis of conventional treatment with western medicine fall blood sugar added with Mongolian SA dew de Di+Xieri GA-4 soup treatment. Control group with western medicine fall blood sugar, a course of 8 weeks. Observed two groups' Mongolian medicine symptoms, 24 h urine trace albumin before and after the treatment. Result: The clinical total effective rate of treatment group was 90%,was better than that of control group 77.5% (P<0.05); After treatment, 24 h urine trace albumin excretion rate(49.45±16.86)μg·min-1 of treatment group was lower than control group(96.52±22.56)μg·min-1(P<0.01). Treatment group in Mongolian medicine single symptom urine was cloudy, frequency quantity more fatigued, lumbar debility improvement better than that in control group (P<0.05). Two groups during treatment without any adverse reaction occurred. Conclusion: Mongolian SA dew de Di+Xieri GA-4 soup in the treatment of early diabetic nephropathy is safe and effective, worthy of clinical use and further study.

      Abstract:Objective: To study the effect of intestinal mucosa permeability,endotoxin blood and ammonia of Severe hepatitis treated with multi-ways administration of Chinese medicine. Method: Eighty-eight severe hepatitis patients were randomLy distributed into therapy group (57 cases)and control group(31 cases). According to clinic sequence,the control group was treated with conventional therapy methods such as protecting liver,supporting treatment and artificial liver support system,in the same time the therapy group was treated with conventional therapy plus multi-ways administration of Chinese medicine such as retention enema,sticking,oral administration dialectically and intravenous drip with traditional Chinese medicine. Result: The total effective rate was 82.46% in the therapy group,in which 16 cases were obviously improved, 18 cases were improved, 7 cases were ineffective and 1 case was sheded; whereas the total effective rate was 61.29% in the control group,in which 3 were cases cured,7 cases were obviously improved, 9 cases were improved,12 cases were ineffective,and 2 cases were sheded. The difference between the two groups was obvious (P<0.05),with the treatment group being better than the control group.A obvious difference(P<0.05) was exhibited in refractory complications, time of hospitalization,number of times of artificial liver treatment, with the treatment group being better than the control group. Conclusion: Multi-ways administration of Chinese medicine could promote the recovery of severe hepatitis patients,reduce the refractory complications,cost,time of hospitalization and the number of times of artificial liver treatment of severe hepatitis patients,thus is a valuable optimization treatment method.

      Abstract:Objective: To observe the Shugan Liqi Zhixie Tang (SLZT) in treatment of 68 patients with diarrhea-predominant irritable bowel syndrome (IBS-D). Method: One hundred and thirty-six cases with IBS-D were randomly divided into treatment group and control group, and each group had 68 cases. The treatment group was given SLZT, one dose/d, and they were used by oral and enema administration; the control group was given trimebutine maleate capsules, 100 mg/time, 3 times/d, and they were used by oral for 8 weeks. The symptoms severity were used by IBS scale table (IBS-BSS), and recorded the main digestive symptom score to evaluate the quality of life of patients, recorded Hamilton Anxiety Scale (HAMA) and Hamilton Depression Scale (HAMD) score and recorded the 3 months recurrent rate of follow-up period. Result: The total effective rate of the treatment group was 89.71% higher than 72.05% of the control group (P<0.05); total effective rate of the treaditional Chinese medicine syndromes in treatment group was 92.64% better than 70.58% of the control group (P<0.01); the IBS-BSS, HAMA, HAMD score of the treatment group were better than the control group (P<0.01), and IBS-QOL score was significantly higher and the control group (P<0.01); abdominal pain, bloating, stool frequency, stool shape and mucus score of the treatment group were better than the control group (P<0.01); the patients were followed up for 3 months, and the cumulative recurrence rate of the control group was 88.23% higher than 58.82% of the treatment group (P<0.01). Conclusion: SLZT in treatment of IBS-D can improve the clinical symptoms and psychological symptoms and improve the quality of life of patients, the clinical effect is significant, and the recurrence rate is low, which is worth to be applied in clinical.

      Abstract:Objective: To explore the effect of traditional Chinese medicine combined with atorvastatin on prognosis, blood lipid and inflammation in borderline lesion coronary heart disease. Method: One hundred and fifty-six cases with borderline lesion coronary heart disease diagnosed by 64-row multi-detector computed tomography were randomly divided into control group and observation group. Two groups received both routine treatment and atorvastatin therapy, meanwhile, observation group was given warming yang promoting blood circulation treatment. Clinical effect, major cardiovascular event, blood lipid and serum inflammatory factors were compared between two groups. Result: The effective rate of treatment in observation group was 92.5%,which was much higher than that in control group (76.3%, P<0.05); major cardiovascular events were 16.3% and 38.2% in observation group and control group respectively with significant difference (P<0.05). After 6 months of treatment, total cholesterol(TC), triglyceride(TG) and low density lipoprotein(LDL-C) in observation group were much lower than those in control group (P<0.05); serum high-sensitivity C-creactive protein(hs-CRP), interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α) after treatment greatly decreased in control group and experience group,which in observation group were superior to control group (P<0.05). Conclusion: Traditional Chinese medicine combined with atorvastatin for borderline lesion coronary heart disease significantly decrease the blood lipid, serum inflammatory factors and major cardiovascular events.

      Abstract:Objective: To investigate the clinical efficacy of ruiyun procedure for hemorrhoids(RPH) Combined with hemorrhoidlysant injection in treating hemorrhoids. Method: One hundred sixty patients with hemorrhoids were divided into two groups according to control trail design by randomized method.Eighty patients in control group were treated with RPH and another has eighty patients in testing group were treated with RPH combined with hemorrhoidlysant injection.The changes of the symptom and physical sign at pre-therapy and post-treatment were compared, and the complications after operation were observed,the cure rate at three and six months after operation and the recurrence rate at 1 and 2 years after operation were observed. Result: The scores of symptom and physical sign were obviously decreased in both groups which after treatment (P<0.05); there was no difference in the scores for each symptom and physical sign between two groups at three months after operation; there was significant difference between the two groups in hemorrhage score,decreasing at six months after treatment (P<0.05) and there was no difference in other indexes. The effects of the treatment between the two groups at three months and six months after operation showing no significant difference, but there is significant difference in recutrence rate at the two years after operation. No serious adverse reactions occurred. Conclusion: RPH combined with hemorrhoidlysant injection are effective and safe in treating hemorrhoids.

      Abstract:Objective: To observe the effect of complex Banmao capsules combined with FOLFOX4 regimen in the treatment of advanced gastrointestinal malignancies patients.Method: One hundren and twenty cases with advanced gastrointestinal malignancies were randomly divided into treatment group and simple group with 60 cases in each group.The two groups were given FOLFOX4 regimen.The treatment group was additionally given complex Banmao capsules(1 g, Tid)for 4 weeks. Result: The karnofsky performance status(KPS) improvement rate in treatment group was 58.3%,while in simple group was 33.3%,two groups had significant difference(P<0.05).All patients with advanced gastrointestinal malignancies showed that the CD4+T cells and natural killer(NK) cells were lower than that of the controlled group(P<0.05), the CD8+T cells were higher than those of the control group(P<0.05). After treatment the CD4+T cells and NK cells in the treatment group[CD4+(42.8±8.2)%,NK(22.1±3.5)%] was increased more significantly than simple group[CD4+(37.8±7.6)%,NK(16.7±5.3)%] (P<0.05). After treatment the CD8+ T cells in the treatment group were decreased more significantly than simple group[CD8+(25.4±3.4)% vs (27.6±6.2)%] (P<0.05). The incidence of toxicity after chemotherapy in the treatment group was lower than simple group (P<0.05). Conclusion: The cellular immunological functions were inhibited in patients with advanced gastrointestinal malignancies.FOLFOX4 regimen could reverse the immune suppression state in patients with advanced gastrointestinal malignancies. The combination of FOLFOX4 regimen and Fufang Banmao Capsules could remarkably improve immunological functions,quality of life and reduce toxicity after FOLFOX4 chemotherapyin advanced gastrointestinal malignancies patients.

      Abstract:Objective: To study the effect of different doses and intervals time on induction outcome on the middle pregnancy by misoprostol in vagina. Method: One hundred and fifty pregnant women who were to be executed misoprostol induction were divided into observation group and control group, two group were given misoprostol induction, in observation group the interval time was 4 h,at the doae of 200 μg, during 24 hour the maximum dose reaching 800 μg. In the control group the interval time was 6 h,at the dose of 400 μg and 1 200 μg;the pregnant women labor outcome was observed and compared. Result: The success rate, amount of vaginal bleeding in two groups were not statistically significant; in the observation group the time of uterine contraction, the total production process and the dosage were lower than that in the control group (P<0.05), the incidence of adverse drug reactions was significantly lowe than that in the control group(P<0.01). Conclusion: Misoprostol at dose of 200 μg and 4 h of interval time is safe effective and suitable for clinical application.

      Abstract:Objective: To observe the therapeutic effects of replenishing Qi, invigorating spleen and expelling phlegm on pediatric pulmonary pneumonia in convalescence. Method: Sixty children with pulmonary pneumonia in convalescence were chosen as lung and spleen Qi deficiency pattern by Chinese medicine dialectics. They were divided randomly into therapeutic group and control group.30 cases in the therapeutic group took Jiao Weir Yu Ping Fen Oral remedy ( produced by the Pharmaceutical Factory affiliated to Capital Institute of Pediatrics Beijing China);30 cases in the control group took Pidotimol granules with Ambroxol hydrochloride d Clenbuterol hydrochloride oral solution. The results were compared after seven days treatment. Result: In the improvement integration of the major and minor symptoms therapeutic group was better than the control group significantly. The therapeutic effect of the former was higher than the latter significantly(P<0.01). The marketly effectivetive rate of the therapeutic group was higher than that of the control group significantly (P<0.05). The Total effective rate of the therapeutic group had no significant difference as compared with the control group. Conclusion: The Chinese herbal remedy by replenishing Qi, invigorating spleen and expelling phlegm has better therapeutic effects than the western remedy alone in the treatment of pediatric pulmonary pneumonia in convalescence of lung and spleen deficiency pattern. In effective rates, expelling phlegm,relieving cough and promoting healthy condition,the therapeutic group is better. Therefore this Chinese herbal remoedy is worth recommending.

      Abstract:Objective: To observe the effect of Xuezhikang capsule on the inflammatory factors,endothelial function and carotid atherosclerotic plaques(CAP)in patients with unstable angina pectoris(UAP). Method: Eighty-two cases of UAP and CAP patients were randomly divided into two groups:the control group received conventional therapy+rosuvastatin,Xuezhikang group received Xuezhikang capsule, 2 capsules once,three times daily,based on the control group.Both groups were given continuous medication for 12 months. Lipids, inflammation, endothelial function indicators and CAP were detected before and after treatment,clinical efficacy in patients of two groups were observed after 12 months of treatment,cardiovascular events were followed up for 12 months. Result: After 12 months of treatment,the total efficiency of Xuezhikang group was significantly higher than that of the control group (P<0.05);there were significant differences between the two groups and the health group in blood lipids,inflammation, endothelial function and intima-media thickness(IMT) values and the area of the CAP(all P<0.01);the improvement of blood lipids,inflammation,endothelial function and IMT values and the area of the CAP in Xuezhikang group was significantly better than that of the control group(all P<0.01); the incidence of cardiovascular events of Xuezhikang group was significantly lower than that of the control group(P<0.05);there was no significant difference between two groups in the incidence of adverse drug reactions. Conclusion: Xuezhikang reduce cardiovascular events by improve endothelial function,reducing the inflammatory response, reversal and stable carotid atherosclerotic plaque,and no increase in adverse effects.

Result: The total effectiveness in control group and treatment group was 80.0% and 50.0%,respectively,the short term curative effect in treatment group was better than the control group, and there was significant difference(P<0.05);the improvement rate of life quality was 70.0% and 40.0%, respectively,the treatment group was better than the control group, and there was significant difference(P<0.05);the improvement rate of body weight was 16.0% and 20.0%, respectively, and there was no significant difference; the peripheral blood abnormality rates of treatment group was obviously lower than the control group, and there was significant difference(P<0.05);the treatment and control group were compared before and after treatment, the immune indexes (CD4+,CD8+,CD4+/CD8+and the activity of NK cell) all presented significant difference of different degree(P<0.05 and P<0.01),and the immunity of treatment group was obviously improved; compared with control group, the occurrence rate of adverse reaction in treatment group was obviously lower than control group, and there was significant difference(P<0.05). The survival rates of patients after 1, 3 and 5 years' treatment was better than the control group, and there was significant difference(P<0.05). Conclusion: The tanyuzufei type lung cancer was cured bylung cancer prescription combined with chemotherapy, and it could effectively relieve the clinical symptoms, improve the immunity, reduce the occurrence of adverse reaction, and improve the life quality, and the traditional Chinese medicine combined with chemotherapy has the efficacy and low toxicity.

    • Observation of Curative Effect of Rosuvastatin Combined with Qizhi Tongluo Capsule in Acute Cerebral Infarction Patients with Normal Cholesterol

      Abstract:Objective: To investigate the effect of rosuvastatin combined with Qizhi Tongluo capsule to high-sensitivity C-reactive protein (hs-CRP)in acute cerebral infarction patients with normal cholesterol, as well as its clinical efficacy. Method: Ninty-eight patients were selected and divided randomly into a control group(n=49) and a treatment group(n=49). Patients in the control group were received 10 mg rosuvastatin calcium tablet treatment every day and others in the treatment group were added 0.2 g Qizhi Tongluo capsule 2 times a day based on foundation treatment. Result: hs-CRP levels and neural function defect scales of two groups on the 8th day and the 15th day after treatments were different. The differences were considered statistically significant, respectively, for P<0.05 and P<0.01.The total effective rates of two groups on the 15th day after treatments were respectively 81.6% and 95.9%(P<0.05).Briefly,compared with the control group, hs-CRP levels and neural function defect scales in patients in the treatment group were lower,but the total effective rate was higher. Conclusion: Combination of rosuvastatin and Qizhi Tongluo capsule in acute cerebral infarction patients with normal cholesterol can promote neural function recovery,which is better than that of alone rosuvastatin.

    • Meta Analysis of Buyang Huanwu Decoction Intervening Prodromal Symptoms of Stroke

      Abstract:Objective: To evaluate the effect of Buyang Huanwu decoction intervening prodromal symptoms of stroke. Method: We searched CNKI, VIP, WanFang (1990 to 2012) for randomized control trials about Buyang Huanwu decoction intervening prodromal symptoms of stroke. Result: According to the inclusion and exclusion criteria and assessment the quality of included trials, the extraction of effective data, 5 randomized clinical trials and their efficiency were evaluated, statistical analysis was conducted by using RevMan 4.2 software. ①Meta analysis showed that the efficiency of Buyang Huanwu decoction on prodromal symptoms of stroke was better than the control group[OR=4.44,95%CI(2.52,7.83)]; ② The indexes of high and low shear on hemorheology were evaluated, plasma viscosity was evaluated, Meta analysis results showed that the Buyang Huanwu decoction reduced the whole blood viscosity and plasma viscosity were better than the control group. Conclusion: The Buyang Huanwu decoction intervening prodromal symptoms of stroke was effective.

    • Zhisou San in Treatting Coughi Induced by Infections Based on Randomized Clinical Trials-Meta-analysis

      Abstract:Objective: A systematic review on treatment of infections by Zhisou san was carried out.Method: Meta-analysis of randomized controlled trial (RCT) was used,and RevMan 5.1 software for statistical analysis was adopted.Result: A total of 24 published RCT (n=2 147) was included,which was divided into the experimental group of 1 087 cases and the control group of 1 060 cases. In simple Zhisou san and western medicine groups the combined OR of cure rate of the group fixed effects model was 3.71, 95% CI[2.96,4.66], P<0.000 01; In Zhisou san combined with western medicine and western medicine, OR was 2.76,95% CI[1.98,3.85, P<0.000 01.The combined OR of Zhisou san was 7.90, 95% CI[5.67,11.01], P<0.000 01; OR of Zhisou san combined with Western medicine. Western medicine was 3.62,95% CI[2.22,5.92], P<0.000 01.Conclusion: Zhisou san in treating infections may be effective, but the further confirmation by high-quality research trials was needed.

      Abstract:By consulting related papers published in recent years,the mode was analyzed and summarized.The model of component formula in Chinese medicine included the component formula of prescription, the component formula of single herb,the component formula of standard material.But the study of the component formula of prescription was only limited in a small number of components.Most of the studies of component formula used the single pharmacodynamic index as evaluation method to determine the optimal ratio of compatibility.The inconsistency of component levels and other problems needed to be resolved.The model of component formula in Chinese medicine has made positive progress, but it also need more basic research support.
