    • Comparison of in vitro Dissolution between Different Dosage Forms of Danqi Prescription

      Abstract:Objective: With total panax notoginsenosides from different formulations of Danqi prescription as a model,to explore new ideas and methods of in vitro dissolution study for multi-components from traditional Chinese medicine(TCM). Method: The small cup method was used with distilled water as dissolution medium at a rotate speed of 50 r·min-1,in vitro dissolution rates in the molar concentration of notoginsenoside Rl,ginsenoside Rb1 and ginsenoside Rg1 were determined by HPLC,and in vitro accumulative dissolution rate of total panax notoginsenosides in the molar concentration was calculated by taking percentage of each component in total panax notoginsenosides as self-defined weighting coefficient. Result: There were significant differences between dripping pills and tables in dissolution rate of notoginsenoside Rl,ginsenoside Rb1,ginsenoside Rg1 (96.87% vs 16.10%,97.15% vs 15.48% and 96.10% vs 16.03% at 30 min);In vitro molar dissolution rate of tables could up to 90% in 120 minutes.It showed that Td and T50 of total panax notoginsenosides and three components in dripping pills were more than tables,which were fitting by Weibull equation. Conclusion: These in vitro dissolution results was in line with in vitro analysis of similar ingredients in TCM,these obtained parameters could characterized in vitro overall release rules of similar substances in TCM.

    • Application of Refractometry in Purification Technology of Active Ingredients from Citrus aurantium by Macroporous Resin

      Abstract:Objective: To explore application feasibility of refractometry in purification technology of macroporous resin. Method: Taking extract of Citrus aurantium as research object,active ingredients in it was refined by AB-8 macroporous resin,washed with water and 50% ethanol,interval sampled,refractive ratio of eluent was monitored on line and analyzed its regularity.At the same time,HPLC and UV spectrophotometric method was used to determine hesperidin as references,and to judge initiation point and terminal point during elution. Result: In purification process of active ingredients from C. aurantium,water elution terminal point 4 BV(refractive index 0) of which was the initiation point by 50% ethanol,9 BV(refractive index 16) of 50% ethanol elution was the terminal point. Conclusion: Refractometry was accurate,reliable,convenient,quickly in purification technology of active ingredients of TCM,it had a strong practical significance.

    • Optimization of Processing Technology for Atractylodes macrocephala Frying with Bran

      Abstract:Objective: To optimize processing technology of Atractylodes macrocephala frying with bran. Method: With the content of atractylenolide Ⅰ,atractylenolide Ⅱ,atractylenolide Ⅲ and atractylone as indexes,which were determined by HPLC,effects of frying temperature,frying time and the amount of bran on processing technology was investigated by L9(34) orthogonal test,in order to determine the best technology. Result: Optimum processing technology was as following:frying temperature 170 ℃,frying time 2 min,the amount of bran 10%. Conclusion: This optimized processing technology was stable and practicable.

    • Optimization of Processing Technology for Lamiophlomis rotate Frying with Vinegar by Orthogonal Test

      Abstract:Objective: To optimize processing technology of Lamiophlomis rotate frying with vinegar. Method: With the content of total flavonoids as index,processing method was selected by single factor test,orthogonal test was adopted to optimize processing technology with moistening time,frying temperature,frying time and the amount of vinegar as factors. Result: Stir-baking with vinegar was adopted,the best processing technology was as following:moistened 5 min with 30% vinegar,adjusted temperature to 200 ℃,processing time 15 min. Conclusion: The content of total flavonoids from L. rotate processing with vinegar increased 21% by comparing with unprocessed.It provided experimental basis for effective utilization of resources of L. rotate.

    • HPLC Simultaneous Determination of Five Nucleoside Compounds in Extracts of Buthus martensii by Different Extraction Technologies

      Abstract:Objective: To compare effects of 3 extraction technologies (water boiling,alcohol extraction and wet ultrafine grinding) on the content of uracil,hypoxanthine,xanthine,guanosine and adenosine from Buthus martensii. Method: Samples of B. martensii were extracted with different technologies,the content of five nucleosides was determined by HPLC.Conditions were as followings:Aglient Zorbax C18 column (4.6 mm×250 mm,5 μm),gradient eluted with 0.07% ice acetic acid-methanol as mobile phase,flow rate 1.0 mL·min-1,detection wavelength 254 nm and column temperature 35 ℃. Result: Uracil, hypoxanthine, xanthine, guanosine, adenosine had a good linear relationship in a certain range of concentration,r>0.999 5,the average recoveries of five nucleoside compounds in all samples were between 98.9%-103.9% with RSD<3%. Conclusion: This established HPLC was convenient and accurate with good repeatability,it could be used to determine simultaneously the content of five nucleosides(uracil, hypoxanthine, xanthine, guanosine, adenosine) and compare difference among extracts of B. martensii by different technologies.

      Abstract:Objective: To optimize extraction conditions of Runfu Zhiyang liposome gels. Method: With the content of hydroxy safflower yellow A and matrine,extract yield as comprehensive evaluation index,based on single factor test,effects of soaking time,extraction time and the amount of water on technology was investigated by orthogonal test. Result: Optimum extraction technology was as following:extracted 2,1.5 h with 12,10 times the amount of water after soaking 0.5 h,respectively. Conclusion: This optimized technology was stable and feasible,it offered expermental basis for further development of Runfu Zhiyang liposome gels.

    • Optimization of Extraction Technology of Effective Parts in Artemisia annua and Evaluation of Its Repellent Effect

      Abstract:Objective: To optimize extraction technology of effective parts in Artemisia annua and evaluate its repellent effect. Method: Extraction method of repellent effective parts in A. annua was screened by single factor test;With yield of volatile oil as index,effects of time,temperature and pressure on extraction technology was investigated by orthogonal test.Repellent effect of the extract was evaluated with effective protection rate as index. Result: Supercritical CO2 extraction technology was adopted,optimum technology was as following:temperature 60 ℃,pressure 30 MPa,extraction time 2 h,yield of volatile oil approximately 0.35%.Effective protection rate of these prepared extract of A annua (concentration>0.05 g·mL-1) reached 99% in 2 hours. Conclusion: This optimized technology was reasonable and stable and its extract had well repellent effect.

    • Optimization of Separation and Purification Technology for Chlorogenic Acids in Leaves of Lonicera japonica with Polyamide Resin

      Abstract:Objective: To optimize separation and purification technology of chlorogenic acid in leaves of Lonicera japonica. Method: With the content of chlorogenic acid as index,static and dynamic absorption,desorption properties of polyamide resin were observed by single factor test,and sample amount,the amount of washing water,type and dosage of eluent were investigated;With purity and retention rate as index,effects of elution flow rate,the concentration and volume of eluent on separation and purification technology was investigated by orthogonal test. Result: Optimized separation and purification technology was as following:sample amount 10 mg·g-1 resin,washed impurity with 2 BV water,and eluated with 3 BV the amount of 65% ethanol,flow rate of 0.4 BV·h-1,collected eluent. Conclusion: This optimized technology was stable and practicable,it could provide reference for industrial separation and purification of chlorogenic acid.

      Abstract:Objective: To optimize extraction technology of total flavonoids from Camellia oleifera seeds. Method: With extraction temperature,extraction time,ethanol concentration and solid-liquid ratio as independent variables,Box-Behnken central composite design was used to establish mathematical model,response surface and contour were made with yield of total flavonoids as response value,response surface methodology was adopted to optimize extraction technology of total flavonoids from C. oleifera seeds. Result: Optimum extraction technology conditions were as followings:extraction temperature 66 ℃,ethanol concentration 70%,extracting time 1.3 h,solid-liquid ratio of 1:22.Under these conditions,yield of total flavonoids from C. oleifera seeds was 1.87%,which was close to the theoretical value of 1.89%. Conclusion: This optimized technology was stable and reliable,and it could provide reference for rational utilization of resources of C. oleifera.

    • Optimization of Microwave-assisted Extraction Technology for Alantolactone and Isoalantolactone from Inula helenium Roots

Method: With the content of alantolactone and isoalantolactone as indexes,which were determined by HPLC,effects of the concentration of ethanol,liquid-solid ratio,extraction time and temperature on technology was investigated by single factor test and orthogonal test. Result: Optimal technology conditions described as followings:microwave extracted 180 s with 15 times the amount of 90% ethanol at 45 ℃. Conclusion: This optimized technology was stable and feasible with low energy consumption and time saving.

Method: With the content of W2 as index,which was determined by HPLC,effects of extraction time,extraction solvent,liquid-solid ratio and extraction times on extraction technology were investigated by single factor test and orthogonal test,then to determine optimum technology of W2.Result: Optimum extraction process of W2 was as following:reflux extracted 2 times with 12 times the amount of 30% ethanol,2.5 hours per time.Conclusion: Optimized technology was reasonable,stable and feasible,which could provide a experimental basis for the follow-up research of C. manshurica Rupr.

    • Preparation and Transdermal Absorption of dl-tetrahydropalmatine Flexible Liposomes

      Abstract:Objective: To prepare dl-tetrahydropalmatine flexible liposomes and study its transdermal absorption effect. Method: The flexible liposomes was prepared by film dispersion method,modified Franz diffusion cell method was used to study its transdermal absorption effect. Result: Particle size of the flexible liposomes was (112.3±59.2) nm with a normal distribution basically,entrapment efficiency was 82.7%.Cumulative permeation amount of dl-tetrahydropalmatine flexible liposomes was 2 121 μg within 12 h,which was significantly higher than that of tincture (1 153 μg).The retention rate in skin was 53.9%,which was obviously higher than that of tincture(11.2%). Conclusion: Transdermal absorption effect of these prepared dl-tetrahydropalmatine flexible liposomes was superior than that of tincture.

    • Preparation and Anti-tumor Activity Investigation of Combretastatin A-4P Nanoparticles Encapsulated with PLGA

      Abstract:Objective: To prepare combretastatin A-4P(CA4P) sustained release nanoparticles with polylactic acid-polyglycolic acid(PLGA) as carrier material,and investigate its in vitro and in vivo anti-tumor activity. Method: CA4P-PLGA-NPs were made by W/O/W double emulsion solvent evaporation method,morphology of CA4P-PLGA-NPs was observed by transmission electron microscopy,particle size distribution was measured by dynamic laser scattering instrument,drug loading,encapsulation efficiency and in vitro cumulative release rate were determined by HPLC.In vitro cytotoxic activity of CA4P-PLGA-NPs on Hela cell was measured by MTT assay,in vivo antitumor activity was evaluated by tumor-bearing nude mice test. Result: Particle size distribution of CA4P-PLGA-NPs was narrow with average particle size was 113.7 nm,average drug loading was (12.5±0.8)%,average encapsulation efficiency was (58.9±1.2)%,these data showed that CA4P-PLGA-NPs had a certain sustained-release effect.CA4P-PLGA-NPs had good in vitro and in vivo anti-tumor activity through MTT and tumor-bearing nude mice test. Conclusion: CA4P-PLGA-NPs prepared by W/O/W solvent evaporation method had sustained-release effect,its anti-tumor activity was better than CA4P.

    • Establishment of Rapid Detection Method for Sulfur Dioxide Residues in Chinese Herbal Medicines

      Abstract:Objective: To establish a fast detection method of sulfur dioxide residues in stoved traditional Chinese medicine(TCM). Method: Colour chromaticity was detected by spectrocolorimeter,taking advantage of the principle of purple substance creation by sodium sulfite reacting with OPA and ammonium acetate under certain conditions,20 batches of TCM were detected,and their sulfur dioxide residues were semi-quantitative. Result: A linear relationship was obtained between color chromaticity and the addition of sodium sulfite in the range of 0-25.23 μg,the range of sulfur dioxide residues in samples could be determined by comparing with the standard addition of sodium sulfite. Conclusion: This method was simple,rapid and sensitive,it could provide a reference for fast detection and half quantification of sulfur dioxide residues in TCM.

      Abstract:Objective: To optimize formulation of ursolic acid self-microemulsion drug delivery system using central composite design/response surface methodology. Method: With ratio of surfactants and cosurfactants and oil content as independent variables,particle size and saturated drug doading were taken as dependent variables,optimum formulation was screened by SAS9.1.3 software,and verified. Result: Optimized formulation of ursolic acid self-microemulsion was oil phase(MCT)-emulsifier(HS15)-cosurfactants(alcohol) 12.5:62.5:25. Conclusion: This optimized formulation of ursolic acid self-microemulsion drug delivery system could be obtained exactly and conveniently by central composite design response surface methodology,and established model had a reliable predictability.

      Abstract:Objective: To optimize extraction technology of Qixue Shuangbu oral liquid. Method: Alcohol and water dual extraction method was adopted,with extraction amount of astragaloside IV as index,effects of extraction time,the concentration of ethanol,the amount of ethanol and extraction times on alcohol extraction technology was investigated by orthogonal test;With extraction ratio of peoniflorin and icariin as indicators,extraction time,extraction times and the amount water as factors,water extraction technology was optimized by orthogonal test. Result: Optimum extraction technology conditions were as follows:Astragalus membranaceus and Angelica sinensis were extracted twice with 8 times the amount of 70% ethanol by 1.5 h per time;Then the gruffs and else prescription drugs were reflux extracted 3 times with 9 the amount of water by two hours every time. Conclusion: This optimized extraction technology was scientific,reasonable,stable and feasible.

    • Prescription Optimization and Quality Evaluation of Andrographolide Self-microemulsion Preparation

      Abstract:Objective: To optimize formulation of andrographolide self-microemulsifying preparation and evaluate its quality. Method: Type and dosage range of excipients was selected by solubility test,prescription compatibility comparison test and drawing ternary phase diagram,with appearance,particle size,zeta potential,self-emulsifying time and preparation stability of the formed emulsion as indexes,central composite design-response surface methodology was used to optimize self-microemulsion prescription and evaluate its quality. Result: Optimum prescription of this preparation was as following:mtween-80:methyl oleate:mglycerol=50:26:26,particle size 39.5 nm,zeta potential -15.18 mV,self-emulsifying time 40.53 s. Conclusion: These acquired androgra-pholide self-microemulsion preparation was clear and transparent with good physical stability.

    • Investigation of UPLC Fingerprint for Kangfuxin Liquid

      Abstract:Objective: To establish an UPLC characteristic chromatographic profile analysis method for Kangfuxin liquid,and provide basis for rapid evaluating quality of it. Method: Taking ZORBAX RRHD Eclipse plus C18 column (2.1 mm×100 mm,1.8 μm),with methanol-0.05% acetic acid solution as mobile phase,detection wavelength 256 nm. Fifteen batches of Kangfuxin liquid from different manufacturers were investigated,fingerprint was analysed by 2004A edition of chromatographic fingerprint similarity evaluation system and cluster analysis software. Result: There were 10 common peaks in 11 batches of Kangfuxin liquid from Kelun pharmaceutical and 3 batches from Haoyisheng Panxi pharmaceutical,the peaks similarity was more than 0.990;However,only 8 common peaks in these productions from Inner Mongolia Jingxin pharmaceutical, the similarity was lower than 0.900 by comparing with the control fingerprint.Five common peaks were identified in the fingerprint. Conclusion: This established method was rapid and effective,it could be useful for quality evaluation of Kangfuxin liquid.

    • Quality Standard of Shentao Ruangan Capsules

      Abstract:Objective: To establish the standard for quality control of Shentao Ruangan capsules. Method: Panax ginseng,Angelica sinensis,Salvia miltiorrhiza,Rheum officinale were identified qualitatively by TCL.The content of ginsenoside Rb1was determined by HPLC,chromatographic conditions were as follows:with acetonitrile-water as mobile phase in gradient elution,the detection wavelength of 203 nm,the flow rate of 1.0 mL·min-1. Result: TCL were reflecting a good specificity and could detect target substance with spots were clear and negative control was without interference.Ginsenoside Rb1 showed a good linearity at the range of 0.406-4.059 7 μg(r=0.999 7),the average recovery was 100.96% with RSD 1.16%. Conclusion: This established method was simple,accurate and reproducible,which could effectively control the quality of Shentao Ruangan capsules.

    • Optimization of Water Extraction and Alcohol Precipitation Technology of Kubai Fuyan Suppositories

      Abstract:Objective: To optimize water extraction and alcohol precipitation technology of Kubai Fuyan suppositories. Method: Water extraction and alcohol precipitation method was adopted,with the quality of matrine,berberine and osthole as indexes,effects of liquid-solid ratio,extraction times and time on water extraction technology was investigated by orthogonal test;With composite score of the quality of matrine,berberine and osthole as index,the relatively density of concentrated liquid,the concentration of alcohol precipitation and storage time were chosen as factors,alcohol precipitation technology was optimized by orthogonal test. Result: Optimum extraction process was as following:extracted 3 times with 12 times the amount of water,2 hours each time;Optimized alcohol precipitation technology was:the relative density of concentrated liquid 1.18 (25 ℃),the concentration of alcohol precipitation 60%,placed 12 hours. Conclusion: These optimized technologies was stable,reasonable and feasible,them could provide experimental basis for clinical application of Kubai Fuyan suppositories.

    • Content Determination of Sodium Benzoate and Acesulfame in Ibuprofen Oral Solution by HPLC

      Abstract:Objective: To establish a HPLC method for simultaneously determining the content of sodium benzoate and acesulfame in ibuprofen oral solution. Method: The content of sodium benzoate and acesulfame was determined by HPLC,chromatographic conditions were as follows:J'sphere ODS-H80 column(4.6 mm×250 mm,4.0 μm),the mobile phase of methanol-0.02 mol·L-1 ammonium acetate (15:85),column temperature 30 ℃,the flow rate of 1.0 mL·min-1,detection wavelength 230 nm,the injection volume 10 μL. Result: Acesulfame showed a good linear relationship at the range of 15.75-110.25 μg, the average recovery of it was 100.51% with RSD 0.79%.Sodium benzoate showed a good linear relationship at the range of 7.8-47.8 μg,the average recovery was 100.42% with RSD 1.56%. Conclusion: This method was sensitive,simple and reproducible,it could provide a reference for determining the content of sodium benzoate and acesulfame in ibuprofen oral solution.

    • Study on the Four Flavone-C-glycosides by Electrospray Ionization Tandem Mass Spectrometry

      Abstract:Objective: To study on the four flavone-C-glycosides which were vicenin-2, luteolin-6-C-glucopyranosyl-8-C-xylopyranoside, quercetin-6-C-glucopyranoside and dihydroquercetin-6-C-glucopyranoside by electrospray ionization tandem mass spectrometry (ESI-MSn). Method: The samples were analyzed by ESI-MSn under positive and negative ion models. Result: Under negative ion model, flavone-C-glycosides which had pentose substituent take place sugar bond cleavage and generate fragment ions (m/z 90 and 60), these which had hexose substituent generate fragment ions (m/z 120 and 90) and these which had two sugar substituents successively generate fragment ions (m/z 120, 90 and 60). Under positive ion model, flavone-C-glycosides take place dehydration,and these flavones which had pentose substituent take place sugar bond cleavage and generate fragment ions (m/z 60), these which had hexose substituent generate fragment ions (m/z 120). Conclusion: The ESI-MSn methods for the identification of flavone-C-glycosides, especially which had two sugar substituents, were established.

    • HPLC Determination of Quercetin and Kaempferol in Ailanthi Fructus

      Abstract:Objective: To establish an HPLC method for the simultaneous determination of quercetin and kaempferol in Ailanthi Fructus. Method: Quercetin and kaempferol were separated on Inertsil ODS-3 (4.6 mm×250 mm, 5 μm) column and detected at 360 nm. The mobile phase was methanol-0.4% phosphate (50:50). The flow rate was 1.0 mL·min-1. Result: Quercetin and kaempferol were linear within the range of 11.1-333 μg (r=1), 2.3-69 μg (r=1) respectively. The average recovery was 97.82% with RSD of 1.1%, 96.81% with RSD of 1.7% respectively. Conclusion: The method is simple, reproducible, and it can be used for the quality control of Ailanthi Fructus.

      Abstract:Objective: To develop a ultra performance liquid chromatography (UP-LC) method for fingerprints of Qili Qiangxin Capsule. Method: The Waters Acquity UPLC BEH C18 (2.1 mm×100 mm, 1.7 μm) column was used. Acetonitrile and water containing 0.15% phosphoric acid were used as mobile phase of UPLC with gradient elution. The detection wavelength was set at 203 nm. Flow rate was set at 0.5 mL·min-1 and column temperature was maintained at 40 ℃. Result: UPLC fingerprint was established with 36 common peaks, 8 of 36 common peaks were identified, and the similar degrees of the UPLC fingerprints of Qili Qiangxin capsule from 10 batches were between 0.979 and 1.000. Conclusion: The method of fingerprints has acceptable precision, reproducibility and stability, which can be used as a new means for the quality control of Qili Qiangxin capsule.

      Abstract:Objective: To study the effects of different purification ways on the extractive rate of caffeoyl quinic acids in Flos Lonicerae extractive. Method: Several purification processes were used respectively in the preparation of Flos Lonicerae extractive, such as alcohol precipitation method, lime-sulfur method, lime-sulfur ethanol method, macroporous resin method, and extraction method etc. The content of the six caffeoyl quinic acids was determined by HPLC. Result: The content of the caffeoyl quinic acids in Flos Lonicerae extractive by extraction method was higher than the other methods. Total content of caffeoyl quinic acids was up to 83.91%. Conclusion: The extraction method is the best in the preparation of Flos Lonicerae extractive.

MS condition: adopted electrospray ionization (ESI),and multiple reaction monitoring (MRM); monitoring by ion-pairs: repaglinide (453.4→86.0,453.4→230.3), glimepiride (491.4→126.2,491.4→352.3), glibenclamide (494.2→168.5,494.2→368.9), pioglitazone (357.2→119.0, 357.2→134.1), gliclazide (342.2→90.9, 324.2→109.9), glipizide (446.2→92.6, 446.2→102.6), phenethylbiguanide (206.2→59.7, 206.2→104.6), dimethyl biguanide (130.1→60.0, 130.1→71.0). The retention time and relative intensities of the detected qualitative ion pairs using for qualitative, integrating the signals of the detected quantitative ion pairs using for quantitative. Result: The eight kinds of antidiabetic drugs had good linearity with correlation coefficient>0.999, ranging from 4.056-129.792, 13.912-125.184, 3.92-125.44, 3.944-126.208, 4.056-129.792, 4.232-135.424, 4.072-130.304 and 4.264, 136.448 ng·L-1 respectively. The average recoveries were 98.18-100.95%. Repeatability experiments showed that RSD values of content were less than 2.95%. Conclusion: This method is reliable, simple and rapid with qualitative and quantitative accuracy, can be used for determination of antidiabetic drugs illegally added in Wentangan capsule.

Result: Seven compounds were obtained and identified as diosmetin(1), quercetin (2), tricin-7-O-β-D-glucopyranoside (3), quercetin-3-O-β-D-glucopyranoside(4), rutin(5), loniceroside A (6) and loniceroside B (7), respectively. Conclusion: The seven compounds were obtained from the leaves of L. japonica for the first time.

      Abstract:Objective: To investigate the influence on the adhesive activity and effective component content in American ginseng cataplasm with different radiation doses. Method: A HPLC method was established for the determination of 3 kinds of ginsenoside in American ginseng cataplasm; changes of adhesive activity and main constituents were compared in samples irradiated with different radiation doses. Result: 60Co-γ radiation with different radiation didn't lead the adhesive activity of American ginseng Cataplasm to change obviously, and the content of ginsenoside Rg1, ginsenoside Re and ginsenoside Rb1 in American ginseng cataplasm was about 8, 90,90,22 and 189 μg·cm-2 respectively before and after 60Co-γ radiation. Conclusion: There is no obvious influence on the adhesive activity and the content of main constituents of American ginseng cataplasm before and after sterilization with the method of 60Co-γ radiation.

      Abstract:Objective: To compare the component of protein, vitamin C, vitamin E and volatile oil and superoside dismutase(SOD) activity of fresh and dried American ginseng and provide reference for reasonable application of American ginseng. Method: SDS-PAGE electrophoresis was carried out for protein analysis; UV spectrophotometry was used for determination of vitamin C and vitamin E; GC-MS was used for the analysis of volatile oil; SOD activity assay was detected with the methods of pyrogallol autoxidation and the data were analyzed by t-test. Result: The electrophoresis bands between fresh and dried American ginseng was different and the content of protein in fresh American ginseng was higher than dried one. The content of vitamin C and vitamin E, SOD activity assay in fresh American ginseng were significantly higher than dried one (P<0.01). The content and the kind of components of volatile oil were different and in fresh American ginseng was more than dried one. Conclusion: The component of protein, vitamin C, vitamin E, volatile oil, SOD activity in fresh American ginseng and dried one was different, which may be one of the reasons for the different of properties and efficacy.

      Abstract:Objective: To evaluate the existing form of amygdalin in bitter almond and the influence of temperature and acid-base on isomerization of amygdalin. Method: Use high performance liquid chromatography (HPLC) to determine the peak area of D-amygdalin and L-amygdalin in different decocting temperature, and use this method qualitatively investigate the influence of acid-base on isomerization of amygdalin monomer. Result: Amygdalin in Semen Armeniacae Amarum in different temperatures have different isomerization performance; amygdalin monomer did not occur isomerization in acid environment, isomerization under the effect of ammonia, sodium hydroxide. Conclusion: With the rise of decocting temperature results in the peak area of D-amygdalin and L-amygdalin gradually increased. Amygdalin monomer is stable in the acidic environment, alkali lead one of the factors of isomerization.

      Abstract:Objective: To compare the chemical components of essential oil from different varieties (strains) of Angelica dahurica var. farmosana by GC-MS. Method: Eight varieties (strains) of A. dahurica were treated with ethanol ultrasonic. The essential oil was extracted by water steam distillation. The component of essential oil was analyzed by GC-MS. The relative percentage content was calculated by using peak area normalization method. Result: 78 compounds were identified in eight varieties (strains) A. dahurica, mainly including alcohol and alkenes. Eight varieties (strains) of A. dahurica identified 40, 33, 26, 38, 38, 42, 33, 36 compounds, respectively. 12 compounds contained in all common compositions were accounted for over 50% of the total essential oil except B-6.Pyrazines were contained only in B-7. Conclusion: The compositions and content of essential oil are consistent in general, but there are also some differences. It provides some scientific basis for further development and utilization of A. dahurica.

      Abstract:Objective: To study the effect of different drying methods on butylphthalide in different parts of Ligusticum chuanxiong. Method: The content of butylphthalide in L. chuanxiong from different drying methods and different parts was determined by HPLC. Result: There was a good linear relationship between butylphthalide and peak area when the sample injection was from 0.515 2-5.152 0 μg (r=0.999 9), the quantification limit was 5.515 2 mg·L-1, the average recovery was 100.1%, and RSD was 1.10% (n=9). The butylphthalide content in different parts of L. chuanxiong arranged in descending order was rhizome>stem>leaf. Different drying methods had some influence on the butylphthalide content in different parts of L. chuanxiong. Conclusion: Butylphthalide content in different parts of L. chuanxiong is different. Among the three drying methods, oven-drying after sunshine is the optimal one for L. chuanxiong.

      Abstract:Objective: To establish the quality standard of honey bran fried Bombyx Batryticatus. Method: According to the provisions of the Chinese pharmacopoeia methods for microscopic identification, the moisture content, ash content, extract and content were determined. Adopted the chroma difference meter to measure chroma changes before and after processed by using the CIE-LAB chroma value as indicators. The TLC identification was improved based on reference methods and the effects were good. Result: The research results showed that the processed products and raw products had the same microscopic characteristics, and the moisture content was about 6.0%, the total ash content mean was 5.4%, the acid insoluble ash content was 0.12%, water-soluble extract content mean was 21.4%, alcohol-soluble extract content was 13.44%. The thin layer identify showed that the methanol extracts of processed products and raw products were almost not different from each other. But the color difference was quite evident. The processed products' total chrominance mean value was 47.269, and the raw products' total product color value of the mean was 60.595.In L*, a*, b* value as indicators, the honey bran fried products from the numerical value of the L* range of 45.984±3.998, a* value is in the range of 8.315±2.149, b* value was 8.949±3.322.The content of ammonium oxalate in honey bran fried Bombyx Batryticatus should not be less than 4.0%. Conclusion: This quality standards and regulations are both in the range of the product quality standard of Bombyx Batryticatus in China pharmacopoeia except of the color identification. So it can be used as a honey stir frying with bran quality standard of reference.

    • Simultaneous Determination of Six Isoflavonoids in Radix Puerariae Lobatae from Different Habitats by HPLC

      Abstract:Objective: A reversed-phase high performance liquid chromatographic method (RP-HPLC) was developed for the simultaneous analysis of six isoflavonoids of Gegen, the roots of Puerariae lobatae. Method: HPLC determination was performed on a Kromasil C18 column (4.6 mm×250 mm, 5 μm) and detected at 250 nm. The mobile phase was consisted of methanol and 0.1% citric acid solution with gradient elution.The flow rate was 1.0 mL·min-1, and column temperature was kept at 30 ℃. Result: The method was proved to be linear in the ranges of 76.2-1 016, 65.8-1 316, 85.6-1 712, 49.98-499.8, 21.94-219.4, 48.5-485 ng for the six isoflavonoid,3'-hydroxypuerarin, puerarin, daidzin, genistin, ononin, daidzein, respectively. The average recoveries were 99.73% (RSD 0.6%), 100.48%(RSD 1.11%), 100.37%(RSD 0.79%), 100.28%(RSD 1.07%), 99.96%(RSD 1.22%),97.36%(RSD 1.59%),respectively. Conclusion: Results obtained showed that the method is convenient, accurate and reliable, can be applied to the simultaneous determination of the six isoflavonoids in Radix Puerariae lobatae.

    • Studies on Chemical Constituents of Different Extracts from Capsicum chinense by GC-MS

      Abstract:Objective: To study the chemical constituents extracted from Capsicum chinense with different solvents and find out the appropriate solvent which was suitable for the extraction of different types of chemical constituents. Method: C. chinense was extracted with petroleum ether, ethyl acetate and 95%ethanol respectively and then the extracts were analyzed by GC-MS. The varieties and content of the chemical constituents in different extracts were compared. Result: The petroleum ether, ethyl acetate and ethanol extracts from C. chinense mainly include fatty acids, acid amides (capsaicin and dihydrocapsaicin), hydrocarbon, sterols and vitamins. The content of fatty acids in three extracts was 63.40%, 60.66%, 16.16%, respectively, and the acid amides were 16.82%, 27.64%, 42.05%, respectively. Conclusion: The appropriate solvent of extracting the fatty acids and a few of hydrocarbons from C. chinense was petroleum ether, as for the extraction of the acid amides, 95% ethanol was more suitable.

      Abstract:Objective: To establish an HPLC method for a standard fingerprint and determination of saikosaponin b2 for quality control of Radix Bupleurioral liquids. Method: About 10 samples from different batches were analyzed by HPLC. By the Similarity Evaluation System for Chromatographic Fingerprint of Traditional Chinese Medicine (Version 2004A), mean chromatogram was generated as the representative standard fingerprint and the similarity of each chromatogram against the standard chromatogram was also calculated. Result: Eleven main marker peaks were selected in the standard fingerprint and 1 peak of them were identified. The similarity of all samples was over 0.985.The linear range of saikosaponin b2 was 0.039 7-0.793 3 μg (r=0.999 1). The average recovery was 98.27% with RSD of 1.79% (n=6). Conclusion: The developed method was reliable, and can be used for the quality control of Radix Bupleurioral liquids.

    • Determination of 27 Pesticide Residues in Angelica Pubescentis Radix by QuEChERS and Online GPC-GC-MS

      Abstract:Objective: To establish a quick method for the determination of 27 pesticide multi-residues in Angelica Pubescentis Radix by using QuEChERS and online gel permeation chromatography-gas chromatography/mass spectrometry (GPC-GC-MS). Method: Pesticides were extracted from samples with 1% acetic acid in acetonitrile under ultrasonic operation and cleaned up by dispersive solid phase extraction, and then analyzed by using online GPC-GC-MS in selective ion monitoring mode (SIM). Result: Twenty-seven kinds of pesticides showed good linearity in a detected concentration range with good correlation coefficient (r=0.994 3-0.999 9). The limits of detection and limits of quantization ranged from 0.037 5 to 3.040 0 μg·L-1 and 0.125 0 to 10.100 0 μg·L-1. The recoveries were 61.7%-114% and the relative standard deviations were 1.03%-13.9% at the spiked level of 0.1 mg·kg-1. Conclusion: This method is simple, rapid, selective and accurate, meet the requirement of pesticide multi-residues analysis. It can detect 27 pesticide multi-residues in Angelica Pubescentis Radix.

The column temperature was kept at 35 ℃, and the detection wavelength was set at 205 nm. The flow rate was 1.0 mL·min-1. Result: Eight kinds of alkaloids were separated well, and the content was greater than 50%. Conclusion: The method is simple, more convenient and accurate for determination of the content of TASA than the original standard titration method.

      Abstract:Objective: To determine the content of Magnolol in Huoxiang Zhengqi capsules by near-infrared spectroscopy(NIR). Method: Determined the content of magnolol in 109 samples by HPLC. Collected their NIR spectrograms data,calibration model of magnolol was established by the Partial least squares regression analysis. Result: The correlation coefficients (R2), the root-mean-square error of cross-validation and the root-mean-square error of calibration (RMSEC) of the calibration model for magnolol were 0.945 6,0.054 3,0.024 5; and the root-mean-square error of prediction (RMSEP) was 0.045 9, the average rate of recovery was 101.26%. Conclusion: The method is steady, accurate, and can be applied for the fast determination of magnolol within. Huoxiang Zhengqi capsules.

      Abstract:Objective: To develop a LC-MS-MS method for determination of tetrodotoxin (TTX) in Fugu liver. Method: The analyte was vortex-mixed, extracted with acetonitrile by ultrasound, and followed by centrifugation. The supernatant (5 μL) was injected into the LC-MS-MS system for analysis. The separation was performed on an Innovation HP Amide column (100 mm×3.0 mm, 5 μm) at 30 ℃ using acetonitrile-0.3% formic acid (70:30) as mobile phase at a flow rate of 0.5 mL·min-1. Quantification was through tandem-mass spectrometry with positive electro-spray ionization (ESI) and multiple reaction monitoring (MRM) at m/z [M+H]+320.1162.1 for TTX. Result: Calibration curve was linear over the TTX concentration range of 0.0313-2.00 mg·L-1 (r=0.999 9). The lower limit of quantification of TTX was 0.300 μg·L-1. The average recovery of TTX was 96.3%-103.0%. Conclusion: The validated method was shown to be simple, sensitive, rapid, reproducible and suitable for the determination of TTX in Fugu liver.

    • Researches on Formation Mechanism of Genuineness of Glycyrrhiza uralensis Based on β-AS Gene

      Abstract:Objective: To explore the formation mechanism of genuineness of Glycyrrhiza uralensis. Method: Searching for beta-amyrin synthase(β-AS) gene in Genbank and finding out the conserved region by sequence alignment, then primers were designed by primer premier 5.0 software. β-AS genes of G. uralensis from 3 different origins were amplified and sequenced, which were analysed by MegAlign software. In the differential expression experiment, the total RNA of G. uralensis from 3 different origins were extracted, then cDNA were obtained by reverse transcription. Using 18S gene of G. uralensis as internal reference, relative expression of β-AS genes of G. uralensis from 3 different origins were detected and analysed. Result: The gene polymorphism experiment showed that 108 variable sites were expresent in the nine samples, and the quantity of variable sites of intron was twice than that of exon. The differential expression experiment showed that the average relative expression of β-AS gene of G. uralensis from different origins was significantly different. Conclusion: The differences of gene polymorphism and expression of β-AS gene in G. uralensis from different origins may be one of the important reasons to result in the genuineness of G. uralensis.

    • Studies on Pharmacokinetics of Rhubarb Acid Enteric-coated Sustained-release Pellets in Rabbits

      Abstract:Objective: To study the pharmacokinetics and bioequivalence of self-prepared rhubarb acid enteric-coated sustained-release pellets,and compare the pharmacokinetics of rhein between rhubarb acid enteric-coated sustained-release pellets and solution,make clear the pharmacokinetic character of the sustained release preparation in vivo. Method: Ten rabbits were given a single oral dose of 35 mg·kg-1 of self-prepared rhubarb acid enteric-coated sustained-release pellets (test preparation) and the sodium alginate solution of rhein (reference preparation) in a crossover study. HPLC was applied to determine the plasma concentrations of rhein before and 0.5,1,2,3,4,6,8,9,10,12,24,28,32,36 h after administration of the medicine. The pharmacokinetic parameters were calculatedand statistically analyzed. Result: The plasma concentration curves of both the sustained-release matrix pellets and the sodium alginate solution of rhein had one peak. The main pharmacokinetic parameters of test preparation were as follows: tmax(9.73±0.68)h,Cmax(3.00±0.81) g·L-1, AUC0~∞(59.21±4.35) mg·L -1·h-1,MRT(21.12±1.12)h; and those of reference preparation were as follows: tmax(4.17±0.12)h,cmax(2.71±0.74) g·L-1, AUC0~∞(39.72±3.11) mg·L-1·h-1,MRT(9.33±2.08)h respectively. Conclusion: Compared with the sodium alginate solution of rhein,the self-prepared rhubarb acid enteric-coated sustained-release pellets have better bioavailability.

Method: Rats were treated with 0.81, 2.43, 7.29 g·kg-1 once daily for 21 days, serum were collected for determination of alanine aminotransferase (ALT), aspartate aminotransferase (AST), urea nitrogen (BUN), creatinine (Cr), and liver was removed, then cytosols and microsomes isolated from liver were prepared by differential centrifugation according to the standard procedure. Antioxidant enzmes catalase (CAT), superoxide dismutase (SOD), glutathione peroxidose (GSH-Px), glutathione reductase (Gr) in cytosols and glutathione-S-transferase (GST), uridinediphosphoglucuronate glucuronosyl transferase (UGT) in microsomes were determined by UV-V is spectrophotometer. Result: C. kwangsiensis water extract had no effect on weight, liversomatic indexes, ALT and AST activities under different treatment compared with control group (P>0.05), but significantly decreased the level of Cr (P<0.01) in all groups, middle and high doses could also decreased BUN level (P<0.01). Compared with the control group,C. kwangsiensis had no effect on CAT and GR in cytosols. The groups of 2.43 g·kg-1 (P<0.01) and 7.29 g·kg-1 (P<0.05) significantly increased the activities of SOD. Furthermore, all of the C. kwangsiensis groups significantly increased the activities of GSH-Px (P<0.01). Compared with the control group, all of C. kwangsiensis groups had no effect on the activities of UGT. All groups of C. kwangsiensis, significantly induced the activities of GST (P<0.05) in microsomes. Conclusion: C. kwangsiensis have no toxicity on liver and kidney in rats, on the contrary, C. kwangsiensis may have a protective effect on kidney. C. kwangsiensis can induce the detoxication enzyames in cytosols and microsomes of rat liver, it have antioxidant function and increase the detoxication of liver.

      Abstract:Objective: This project aimed to find if prescription for tonifying kidney fed during embryonic development can prevent impaired glucose tolerance(IGT) caused by feeding high fat diet and its influence on insulin and leptin. Method: The pregnant rats were randomly divided into 5 groups. The normal control group and model group were fed by normal saline on the 1st day,ig,20 mL·kg-1.In other 3 experiment groups,the pregnant rats were fed respectively with Zuogui Wan (ZGW, a prescription for reinforcing kidney Yin) or Yougui Wan(YGW,a prescription for reinforcing kidney Yang)or Bazhen decoction (BZD, a prescription for supplementing Qi and Blood),ig,20 g·kg-1,20 mL·kg-1, at the same time. After birth, the offsping rats in the other four groups except the control group were fed with high fat diet in 6th week. After 12 weeks of high fat diet, the fasting blood glucose(FBG),2 h blood glucose(2 hBG),fasting serum insulin and serum leptin concentrations were measured. Result: 2 hBG of ZGW group was significant lower than 2 hBG of model group(P<0.05);2 hBG of ZGW group and YGW group was not significantly different from model group.HOMA-IR of ZGW group,YGW group and BZD group was significantly lower than HOMA-IR of model group(P<0.01,P<0.01,P<0.05).Leptin of ZGW group and YGW group was significantly lower than leptin of model group (P<0.01,P<0.01). Leptin of BZD group was not significantly different from model group. Conclusion: ZGW and YGW during embryonic development can all withstand the adverse changes of serum insulin,and ZGW can prevent IGT caused by high fat diet, but YGW can not.BZD during embryonic development can also withstand the adverse changes of serum insulin and can not withstand the adverse changes of serum leptin,which is not as good as ZGW and YGW, and can not prevent IGT caused by high fat diet.

      Abstract:Objective: To study the effect of Panax notoginseng saponins(PNS) on the inflammatory factors and microglia cell of senescence-accelerated mouse prone 8 (SAMP8) mice. Method: SAMP8 mice were randomly divided into four groups: PNS high-dosage group, PNS low-dosage group,huperzine A group and model control group. The high-dosage group and low-dosage group were treated with 200, 100 mg·kg-1 PNS respectively per day and the huperzine A group was treated with 0.3 mg·kg-1 huperzine A by intragastric administration,per day for 8 consecutive weeks.The serum cytokines of SAMP8 like interleukin(IL)-1β, IL-6, tumor necrosis factor-α(TNF-α), complement 1q(C1q) were measured by ELISA. The morphological changes of microglia cells were observed by light microscopy with HE staining. Result: The level of IL-1β in the high-dose group was lower than that in the model group (P<0.05), and compared to the model group, the content of IL-6 in PNS high, low-dose groups showed significantly different(P<0.05). The level of C1q in the blank group was higher than that in the high-dose group and positive group. HE staining showed that microglia in the hippocampus of the model group was over-activation, and swallowed nerve cells, while the microglia in the treatment groups was not activated. Conclusion: PNS can reduce the inflammatory response in SAMP8 mice.

    • Effect of Compatibility of Astragali Radix and Salviae Miltiorrhizae Radix et Rhizoma Extract on Pathological Changes of Myocardium in Rats after Myocardial Infarction

      Abstract:Objective: To study the effect of compatibility of Astragali Radix and Salviae Miltiorrhizae Radix et Rhizoma extract on pathological changes of myocardium in rats after myocardial infarction (MI). Method: Left coronary artery of Sprague-Dawley rats were ligated to make MI models. The rats were randomly subjected to MI model group, astragalus group, Salviae Miltiorrhizae Radix et Rhizoma group, compatibility of Astragali Radix and Salviae Miltiorrhizae Radix et Rhizoma and sham-operated group,each group consisted of 8 rats. All the treatment groups were orally given 20 mg·kg-1 ·d-1 drug extract, and the MI group and sham-operated group were fed 0.9% sodium chloride 20 mL·kg-1 ·d-1. 8 weeks later, the rats were sacrificed, the hemodynamic changes in rats were determined, and the segmental heart samples were collected for hematoxylin and eosin staining, masson staining and histological evaluation on expression of protein kinase D1(PKD1). Result: Compared with the MI group, the left ventricular systolic pressure(LVSP) and maximal decrease and increase rate of pressure in left ventricle of Astragali Radix group, Salviae Miltiorrhizae Radix et Rhizoma group, compatibility of Astragali Radix and Salviae Miltiorrhizae Radix et Rhizoma or sham-operated group increased significantly (P<0.01), while the left ventricular end diastolic pressure(LVEDP) decreased significantly (P<0.01). There were serious myocardial necrosis, increased fibroblasts, accompanied by inflammatory cell infiltration in the rat of MI group, and slightly fuzzy cell morphology, nuclear hypertrophy, mild proliferation of fibroblasts, inflammatory cells decreasing in the Astragali Radix or Salviae Miltiorrhizae Radix et Rhizoma group, while clearer cell morphology, rare fibroblasts and inflammatory cells in the compatibility of Astragali Radix and Salviae Miltiorrhizae Radix et Rhizoma group, according to the HE staining analysis. The masson staining results showed that the MI group consisted of more myocardial collagen tissue, yet each treatment group consist of mainly red myocardial tissue, intermingled collagen tissue. The PKD1 protein expression in the cytoplasm of myocardial tissue of the MI group increased significantly (P<0.01), compared to all the treatment group, despite of decreasing obviously, but still visible in the Astragali Radix or Salviae Miltiorrhizae Radix et Rhizoma group. And the PKD1 protein expression in the compatibility of Astragali Radix and Salviae Miltiorrhizae Radix et Rhizoma group declined significantly compared to the Astragali Radix or Salviae Miltiorrhizae Radix et Rhizoma group(P<0.05). Conclusion: Our study reveals that the compatibility of Astragali Radix and Salviae Miltiorrhizae Radix et Rhizoma can significantly ameliorate the abnormal hemorheological indicators and pathologic lesions after myocardial infarction, which may be related to the expression regulation of PKD1 protein in myocardial tissue.

      Abstract:Objective: To observe the effect of Kuijieling decoction containing serum on intestinal trefoil factor(ITF) mRNA and mucin 2(MUC2) protein expression in Caco-2 injury model. Method: Three trinitrobenzene sulfonic acid (TNBS) enema was used for replication of ulcerative colitis (UC) rat model. Cell injury model was established by serum of UC model rats, ITF mRNA and MUC2 protein expression were detected by real time fluorescent quantitative-PCR (RT-PCR) and immunohistochemistry. Result: ITF mRNA (1.20±0.16) and MUC2 protein expression levels(11.22±2.37) in model serum group were higher than that in normal group (1.00±0.11,8.62±3.16) (P<0.05,P<0.01); Compared with the model serum group, ITF mRNA(1.76±0.37) and MUC2 protein expression levels (13.94±2.59) in Kuijieling decoction containing serum group increased significantly (P<0.05, P<0.05). Conclusion: Kuijieling decoction containing serum could promote ITF mRNA and MUC2 protein expression in Caco-2 cell injury model.

      Abstract:Objective: To study the character of hyperlipidemia concurrent cerebral ischemia and analyze the influence of hyperlipidemia inflammation factor on cerebral ischemia by comparing inflammation change in different time change between hyperlipidemia and cerebral ischemia model and normal rat. Method: Dividing experimental rats into normal control, normal sham operation group, day 3, day 7 after normal ischemia and hyperlipid, sham operation group, day 3, day 7 after ischemia with hyperlipidemia. Based on time is day 3 or day 7 after cerebral ischemia. Building classic hyperlipidemia rat model with high fat diet, to confirm a successful model and detect the concentration of serum lipid.Constructing middle cerebral artery occlusion model, and observe the brain ischemia injury of rat. Detect the concentration of monocyte chemotactic protein-1(MCP-1), tumor necrosis factor(TNF-α), creactive protein(CRP) and superoxide dismutase(SOD) in serum between groups by ELISA. Result: The concentration of MCP-1, TNF-α and CRP were increased in day 3 and day 7 after cerebral ischemia model. Compared to normal cerebral ischemia group, in hyperlipidemia concurrent cerebral ischemia group, the concentration of MCP-1, TNF-α were higher with significant difference in day 3 and in day 7 after ischemia (P<0.05). The concentration of CRP were higher with significant difference in day 3 and were prominently decreased in day 7 after ischemia (P<0.05). The concentration of SOD was prominently decreased in the serum after cerebral ischemia in hyperlipidemia and normal groups, compared to sham operation groups and hyperlipidemia sham operation groups. SOD was prominently decreased in day 3 hyperlipidemia concurrent cerebral ischemia (P<0.05). Conclusion: Under the condition of hyperlipidemia, the inflammatory factors have not only accumulated phenomenon, but also specific expression in the difference of cerebral ischemia period, which is contributed to the recovery of the lesion in recovery phase.

      Abstract:Objective: To study the effect of Xiaoyong Kuidekang on expressions of gastrin (GAS) and motilin (MTL) in rat ulcer model and to explore the mechanism of gastric ulcer treated by Xiaoyong Kuidekang. Method: The rats were randomly divided into 5 groups randomly: normal group,sham group,ulcer model group,Xiaoyong Kuidekang group and Omeprazole group.Acetic acid was applied to a certain part of the gastric mucosa to establish the ulcer model. The Xiaoyong Kuidekang and Omeprazole treatment group had been implemented with drugs since the 2nd day after the operation, the serum was collected on the 11 th day and the content of GAS and MTL were measured by ELISA method. Result: The GAS and MTL content of serum in ulcer model group was higher than that in the control group and the sham group (P<0.01), the GAS and MTL content of serum was lower in the Xiaoyong Kuidekang group and Omeprazole group than those in the ulcer model group (P<0.01). Conclusion: The Xiaoyong Kuidekang may be mediated by decreased the GAS and MTL content of serum,so as to achieve the protection of gastric mucosa and promote ulcer healing effect.

    • Protective Effect of Saponins from Bupleurum Chinense on Ethanol-induced Gastric Lesion

      Abstract:Objective: This study was aimed at investigating the effects and mechanism of saponins from bupleurum chinense on ethanol-induced gastric mucosal injury in rats. Method: The rats were divided into control group, model group, bupleurum chinense saponins group (0.047 g·kg-1) and omeprazole group (0.012 g·kg-1) respectively. After two weeks treatment, the experimental gastric damages were established by intragastric ethanol.The protective effects of bupleurum chinense saponins were evaluated by calculating lesion indices and observing pathological changes; meanwhile, the contents prostaglandin E2(PGE2), superoxide dismutase(SOD) and malondialdehyde(MDA) in serum were measured with euzymelinked immunosorbent assay to determine the protecting mechanism. Result: The lesion indices of bupleurum chinense saponins group were significantly lower than the model group(P<0.01). There was no damage was detected in most of rats in treatment group except for small damages in a few rats, and the integral between bupleurum chinense saponins and treatment groups showed statistical differences(P<0.05). The contents of PGE2, SOD in serum of bupleurum chinense saponins group were higher while MDA was lower than the model group(all P<0.01). Conclusion: bupleurum chinense saponins show obvious protective effect on gastric mucosa damage, and its mechanism may be through restraining the gastric mucosa acid secretion and counteracting the injury of free radical.

      Abstract:Objective: To study the effects of L-dopa methyl ester (LDME) on the apoptotic cells the expression of c-fos mRNA, and c-fos positive cells in retina of monocular deprivation cats, then explore its mechanism on amblyopia. Method: Thirty normal cats were randomly divided into 6 groups (5 cats in each group): low dose (LDMEL), medium dose (LDMEM), and high dose of L-dopa methyl ester (LDMEH), positive control group (PC), normal control group (NC) and model contro(MC). The kittens in each group were visually deprived by suturing the lids of the left eyes to establish the monocular deprivation amblyopia, then eye was opened and treated with drug after 12 weeks. LDME (20,40,80 mg·kg-1), L-dopa (40 mg·kg-1) and saline were given to the corresponding animals for 30 days. The TUNEL assay was used to detect apoptosis in retina, and the in situ hybridization and immunohistochemistry techniques were used to detect the expression of c-fos mRNA and c-fos protein. Result: Compared to the model control group, LDME significantly lessened the apoptotic index in retina (P<0.05 or P<0.01), and markedly up-regulated the expression of c-fos mRNA in retina (P<0.05 or P<0.01), meanwhile,elevated the amount of c-fos positive cells (P<0.05 or P<0.01). Conclusion: Levodopa methyl ester can effectively improve the retinal damage caused by deprivation amblyopia, and the mechanisms may be linked to inhibiting apoptosis and up-regulating the expression of c-fos.

      Abstract:Objective: The effect of Pinelliae Rhizoma and Fritillariae Thunbergli Bulbus on antioxidant activity of Wutou decoction (WTD) was studied to validate the scientific of aconite incompatibility. Method: The total antioxidant activity of different samples was determined by ferric reducing antioxidant power (FRAP) assay and DPPH·scavenging assay. The different antioxidant activities of WTD were compared to the WTD added with Pinelliae Rhizoma (RP) and Sichuan fritillary (SF) respectively. Moreover, the antioxidant activities of Monkshood Prepared (PM), PM with opposing herbal remedies and PM with the other herb medicine in WTD. Result: The antioxidant activities of WTD were inhibited by RP and SF. The inhibition activity of SF was more obviously. In the combination between WTD and ZF or PM and ZF, the total antioxidant activity was both enhanced. Conclusion: The antioxidant activities were decreased in the combination between WTD and RP or WTD and SF.

      Abstract:Objective: To explore the effects of the total organic acid of Thladiantha dubia (FTDBP) on activity of isolated rat uterus smooth muscle in vitro. Method: The effect of the FTDBP on activity of the isolated rat uterus smooth muscle of non-pregnant rats eight weeks old was recorded by Maclab/4e four channels physiological recorder. Four antagonists, propranolol hydrochloride tablets (3.4×10-7mol·L-1),diphenhydramine hydrochloride tablets (2×10-6 mol·L-1),ranitidine hydrochloride tablets (2×10-5 mol·L-1) and atropine sulfate tablets (2×10-5 mol·L-1) were used to study their mechanism respectively. Result: Compare with model group, the FTDBP markedly inhibited the tension, frequency and activity of uterus of rat in vitro.And the IC50 of FTDBP was 0.100 24 g·L-1. Conclusion: The effect of FTDBP on activity of uterine smooth muscle in rats is mainly associated with M receptor but not H1 receptor, H2 receptor or β receptor.

    • Comparison on Effect of Anemia Animal Model and HPLC Fingerprint Chromatogram between Polygoni Multiflori Radix and Polygoni Multiflori Radix Preparata

      Abstract:Objective: Tocomparie effect of blood deficiency and HPLC fingerprint chromatogram between Polygoni Multiflori Radix and Polygoni Multiflori Radix Preparata, and to findout the material foundationof effects. Method: According to random design, rats were divided into 4 groups. Except the normal control group, the other groups were subcutaneously injected acetyl phenyl hydrazine 0.015 g·kg-1 and 0.03 g·kg-1 in the 3rd day and the 6th day. From the 6th day, rats were intraperitoneally injected CTX for 3 day. During the period of modeling, treatment groups received corresponding drugs at dose of 2 g·kg-1. Then, the effect of Polygoni Multiflori Radix and Polygoni Multiflori Radix Preparata were observed,and the fingerprint chromatogram of the water extracted from Polygoni Multiflori Radix and Polygoni Multiflori Radix Preparata, respectively, were established by HPLC. Result: Compared with the normal control group, the results showed that the red blood cell count(RBC), hemoglobin(Hgb), hernatocrit(HCT) figures in model group rats declined significantly(P<0.05). Compared with the model group, the results showed that Polygoni Multiflori Radix could not increase RBC and Hgb in anemia model rats, but the HCT was increased(P<0.05), while Polygoni Multiflori Radix Preparata could rise RBC Hgb and HCT significantly in anemia model rats (P<0.01 or P<0.05). The fingerprint chromatograms result revealed that there were differences not only in chemical consistence but in n determination of constituents between two herbals. Conclusion: The results illustrate that Polygoni Multiflori Radix Preparata has significant blood-nourishing ability. Additionally, the result of the fingerprint chromatogram indicates that the product of Maillard reaction and moderate drug properties are both the possible reason why Polygoni Multiflori Radix Preparata has the better efficacy on blood deficiency.

    • Study on Inhibition of Gardeniae Fructus Extracts on Alpha-glucosidase

      Abstract:Objective: To study the inhibitory effects of Gardeniae Fructus extracts on alpha-glucosidase activity. Method: The inhibitory activities of each extracted parts of Gardenia jasminoides against alpha-glucosidase were tested by the colorimetry of PNPG, the reaction system of which included 67 mmol·L-1 potassium phosphate buffer 870 μL, 1 g·L-1 glutathione solution 25 μL, 2.74 U·mL-1 α-glucosidase solution 35 μL, 15 mmol·L-1 PNPG solution 70 μL, 0.1 mol·L-1 Na2CO3 solution 4 mL. Result: At concentrations of 50, 100 g·L-1, Gardeniae Fructus total extract, iridoids and gardenia yellow reduced significantly the activities of alpha-glucosidase with inhibitory rates of 61.47%,83.20%; 34.27%,55.73%; 78.13%, 96.93%. Conclusion: Gardeniae Fructus extracts have the inhibitory effects on alpha-glucosidase, and the active position is probably Iridoids and Gardenia yellow.

    • Effect of TSG and PDTC on Apoptosis of HUVECs Induced by H2O2

      Abstract:Objective: To study the anti-apoptotic effect and mechanism of 2, 3, 5, 4'-tetrahydroxystilbene-2-O-β-D-glucoside (TSG) combined with pyrrolidine dithiocar-bamate (PDTC) on the apoptosis of human umbilical vein endothelial cells (HUVECs) induced by H2O2. Method: HUVECs were divided into five groups: control group, H2O2 group (300 μmol·L-1), TSG group (10 μmol·L-1 TSG + 300 μmol·L-1 H2O2), TSG and PDTC combination group (75 μmol·L-1 PDTC + 10 μmol·L-1 TSG + 300 μmol·L-1 H2O2 group), PDTC group (75 μmol·L-1 PDTC +300 μmol·L-1 H2O2). The viability of HUVECs was detected by MTT assay. The morphology of apoptosis was observed by Hoechst 33258 staining and the rate of apoptosis was determined by flow cytometry. The protein expressions of NF-κB, Caspase-3 were detected by Western-blot. Result: Compared with control group, H2O2 significantly increased the apoptosis, reduced the cell viability, and enhanced the expressions of NF-κB and Caspase-3 (P<0.01). The proliferation rate increased significantly (P<0.01) while the apoptosis rate decreased obviously in TSG or PDTC pretreatment group, and the expression of NF-κB and Caspase-3 down regulated (P<0.05) compared with H2O2 group (P<0.01). Combination of PDTC and TSG raised the cell viability and attenuated the cell apoptosis rates than pretreatment of TSG or PDTC alone, and the expression of NF-κB and Caspase-3 decreased significantly (P<0.01). Conclusion: Both PDTC and TSG pretreatment could inhibit HUVECs apoptosis induced by H2O2, and the combination of PDTC and TSG can lead to the synergistic effect. The mechanism was associated with the expressions of NF-κB and Caspase-3.

RA model rats were randomly divided into model group, positive control group, high dosage TSTR group, middle dosage TSTR group and low dosage TSTR group.TSTR groups were fed with TSTR(40, 80, 160 mg·kg-1·d-1), positive control group were fed with tripterygium(12 mg·kg-1·d-1). After intragastric administration for 21 days, serum levels of OPG and RANKL were evaluated by ELISA. Result: The levels of RANKL of the three TSTR groups (98.74±10.17),(87.81±17.78),(77.75±14.61) pmol·L-1 were significantly lower than the model group (120.80±12.14) pmol·L-1(P<0.05,P<0.01), while the levels of OPG (71.67±2.62),(77.50±0.93),(91.04±7.26) pmol·L-1 were significantly higher than the model group (53.64±3.46) pmol·L-1(P<0.05,P<0.01). Compared to the model group (0.43±0.06), the ratio of OPG/RANKL (0.71±0.08,0.89±0.17,1.13±0.24) were increased significantly in the three TSTR groups(P<0.05,P<0.01). Conclusion: The prevention and therapeutic effect of TSTR on rheumatoid arthritis may be via the up-regulation of OPG and down-regulation of RANKL. This would affect the function of osteoclast.

    • Influence of Jianpi Yiqi Huoxue Decoction on Vasomotor Factors Expression in Vascular Endothelium of Spontaneously Hypertensive Rats

      Abstract:Objective: To study the influence of Jianpi Yiqi Huoxue decoction on systolic pressure and vascular endothelium vasomotor factors of spontaneously hypertensive rats(SHR). Method: Sixty 24-week-old SHRs were randomly divided into six groups: SHR group (distilled water), perindopril group (perindopril 0.4 mg·kg-1·d-1), perindopril plus herb group (perindopril 0.4 mg·kg-1·d-1+20 g·kg-1·d-1 of herb), high, medium and low dose group (40,20,10 g·kg-1·d-1).of Jianpi Yiqi Huoxue decoction and Wistar kyoto rats(WKY) group. The changes of systolic pressure were measured by BP-98A intelligent sphygmomanometer. After six weeks of treatment, the concentration of nitric oxide(NO), prostacycline(PGI2),endothelium derived hyperpolarizing factor(EDHF), angiotensin Ⅱ(AngⅡ), and endothelin(ET-1) in plasma of the rats were determined. Result: The systolic pressure of the perindopril group and the perindopril plus herb group was prominently decreased, compared with that of the SHR group (P<0.01). The concentration of NO, PGI2,and EDHF of the SHR group was lower than those of the WKY group, the perindopril group, the perindopril plus herb group, and the two herb dose groups(P<0.01). The concentration of ET-1, AngⅡ in the SHR was higher than those in the WKY group(P<0.05), and was significantly higher than those in the perindopril group, the perindopril plus herb group, and the three herb dose groups. Conclusion: The Jianpi Yiqi Huoxue decoction may improve functions of blood vessels, and reduce systolic pressure by regulating the balance between the secretion and release of vasomotor factors in vascular endothelium so as to modify the vascular endothelial function obstacles caused by hypertension.

      Abstract:Objective: To observe the effect of Simiao Yong'an decoction(SMYA) on blood lipid and hemorheology in thromboangitis obliterans(TAO) rats. Method: Rats were randomly divided into 6 groups:control group, model group, mailuoning group(2.7 g·kg-1), low, medium and high dose groups of SMYA(24.3,16.2,8.1 g·kg-1). The drugs were administrated to rats for 15 days. The rat TAO model was made by injecting sodium laurate into the femoral artery of rats, and the level of serum lipid and blood rheology were tested. Result: SMYA could obviously lower triglycerides(TG), total cholesterol(TC), oxidized-low density lipoprotein(OX-LDL), blood viscosity and erythrocyte sedimentation rate values, and increase high density lipoprotein (HDL). Conclusion: SMYA can adjust fatty substance metabolism and lower blood lipid and improve hemorrheology of TAO rats,and has therapentic effect on TAO.

      Abstract:Objective: To study the antioxidant activity of different extracts from Rourea microphylla in Guangxi. Method: EE (95%ethanol), AE(acetone)and EAE (ethyl acetate) extracts were obtained by cold-extraction. The antioxidant activities were studied using DPPH(1,1-diphenyl-2-picrylhydrazyl radical 2,2-Dipheny l-1-(2,4,6-trinitrophenyl) hydrazyl)assay, 2,2'-azinobis-(3-ethylbenzthiazoline-6-sulphonate) (ABTS+) assay and reducing power assay by UV-VIS (Ultraviolet and visible spectrophotometry) spectrophotometry. Result: The EE of R. microphylla exhibited the most outstanding antioxidant activities. The IC50 values of EE in DPPH assay and ABTS+ assay were 0.66,0.60 g·L-1, respectively. The reducing power of all extracts was in the sequence of EE>EAE>AE, which was in accordance with the total flavonoid(TF)content. Conclusion: All the extracts especially EE extract of R. microphylla could be employed as natural antioxidants and health care products.

    • Experimental Study of Reversion of Puerariae Radix Lobatae Flavonoids on Pancreas Injury Induced by Streptozotocin in Mice

      Abstract:Objective: To investigate the experimental study of reversion of Puerariae Radix Lobatae flavonoids on pancreas injury induced by streptozotocin in mice. Method: Being established diabetic mice by intraperitoneal injection of streptozotocin (150 mg·kg-1) firstly, and then the mice were randomly divided into 5 groups: model group, dimethyldiguanide group (80 mg·kg-1), low-, moderate-and high-dose groups of Puerariae Radix Lobatae flavonoids (80, 160, 240 mg·kg-1). The drugs were intragastrically given to mice daily for 14 days. The level of fasting blood glucose (FBG) was tested by glucose oxidase method. The levels of fasting serum insulin (FIns) and insulin resistance (IR) were measured via radioimmunoassay (RIA).The activities of glutathione-peroxidase (GSH-Px), superoxide dismutase (SOD) in pancreas tissue, and the contents of reactive oxygen species (ROS) and malondialdehyde (MDA) were using ELISA test. The pathologic changes of pancreatic cells were observed by HE staining. The expression of heme oxygenase-1 (HO-1) in pancreas was through Western blot analysis. Result: Compared to model control group, Puerariae Radix Lobatae flavonoids effectively lowered the blood-glucose level in diabetic mice (P<0.01). The insulin level in serum was restored, and IR condition was improved (P<0.01). The activities of GSH-Px, SOD in pancreas tissue were elevated, while the content of ROS, MDA was lessened (P<0.01). And the islet beta cells injure was reversed. The HO-1 expression was down-regulated in pancreatic tissue (P<0.05). Conclusion: The findings demonstrate that Puerariae Radix Lobatae flavonoids mediate protection against pancreas injury induced by STZ in mice, though the mechanisms involved in amelioration of metabolism in blood glucose and suppression of oxidative stress.

    • Effects of Ethanol Extraction from Cassiae Semen on Leptin and NPY in Hyperlipidemia Rats

      Abstract:Objective: To observe effects of ethanol extraction from Cassiae Semen on leptin and neuropeptide Y(NPY) in hyperlipidemia rats. Method: The rat hyperlipidemia model was established by feeding high calorie-diet. After modeling, the selected rats were randomly divided into six groups: normal control group, model group, positive control group, low dose group, medium dose group and high dose group. Positive control group was administrated Xuezhikang capsule at dose of 0.15 g·kg-1. Low dose group, medium dose group and high dose group were administrated at different concentrations of ethanol extraction of Cassiae Semen, which was respectively 4,8,16 g·kg-1. All groups were applied by intragastric administration for 4 weeks. Except normal control group, the remains were fed with high calorie-diet. After the last administration, the serum content of total cholesterol (TC), triglyceride(TG), low-density lipoprotein-cholesterol(LDL-C), high-density lipoprotein-cholesterol(HDL-C) and glucose(Glu) were detected by automated chemistry analyzer. And the content of leptin and NPY was determined by ELISA. Result: The content of TC, TG, LDL-C and Glu was decreased, and the content of HDL-C was increased in medium dose group and high dose group of ethanol extraction of Cassiae Semen, compared with model group(P<0.01). At the same time, the content of leptin and NPY was decreased in medium dose group and high dose group(P<0.01). Conclusion: Ethanol extraction of Cassiae Semen can regulate the metabolic process of blood lipids and blood glucose levels of hyperlipidemia model rats, which might be contributed to its effect on leptin-hypothalamic neuropeptide Y chain.

      Abstract:Objective: To observe the effect of Mongolian medicine GEI Wang-9 on acute liver failure induced by carbon tetrachloride CCl4. Method: methods 30% carbon tetrachloride was used to induce model of acute liver failure. After 24 h,indexes alanine aminotransferase(ALT),aspartate aminotransferase(AST),nitric oxide(NO),active of nitric oxide composite enzyme (NOS) of serum and liver index and pepsin in the stomach homogenate were detected in normal group,model group,Mongolian medicine GEI Wang-9 low,middle,high dose groups(1.3,2.6,3.9 g·kg-1), respectively. Result: In the model group, compared with normal group, serum AST, ALT extremely increased significantly (all P<0.01); NO,nitric oxide composite enzyme and liver indexall increased significantly (P<0.05). Compared with model group, GEI Wang-9 low doses decreased significantly the serum NO,nitric oxide composite enzymeall (P<0.05). GEI Wang-9, middle doses decreased significantly the serum AST,ALT(P<0.01, P<0.05, respectively); and NO,nitric oxide composite enzyme and liver index reduced significantly (P<0.05). High doses reduced significantly the serum AST,ALT(P<0.01, P<0.05,respectively); NO,nitric oxide composite enzyme and liver index reduced significantly (P<0.05). Conclusion: Mongolian medicine GEI Wang-9 has protective actions on acute liver failure induced by carbon tetrachloride.

    • Influences of Tongluo Huatan Capsule on Neuron Apoptosis and Expression of Bax and Bcl-2 in Rats with Cerebral Ischemic Injury

      Abstract:Objective: To investigate the effect and mechanism of Tongluo Huatan(TLHT) capsule on neuron apoptosis in cerebral ischemic injury. Method: Cerebral ischemia was induced by permanent middle cerebral artery occlusion(MCAO). After operation, the rats were stratified randomly divided into seven groups according to their Longa scores as normal group, sham group, model group, nimodipine group, high dose TLHT capsule group, medium dose TLHT capsule group and low dose TLHT capsule group, respectively given distilled water, nimodipine and Tongluo Huatan capsule. The neuroprotecive effect was detected by Narrow-Alley Corner Test at the 1,2,3,5, 7 days after the operation respectively. The HE staining was applied to observe the pathologic change of ischemic cerebral cortex. The TUNEL technology was used to test the level of neuron apoptosis in the ischemic penumbra, and the immunohistochemistry method was applied to detect the expression of Bcl-2 associated x protein(Bax) and B cell lymphoma/leukemia-2(Bcl-2) in the ischemic penumbra. Result: The scores of Narrow-Alley Corner Test of model group were significantly increased compared with normal group and sham group, while those of nimodipine group and TLHT capsule groups were reduced compared with model group. Neuron apoptosis of ischemic penumbra of model group was increased compared with normal group and sham group. The expression of Bax and Bcl-2 in ischemic penumbra of model group was increased compared with normal group and sham group. The level of neuron apoptosis and the expression of Bax in ischemic penumbra of nimodipine group and TLHT capsule groups were decreased, while the expression of Bcl-2 was increased compared with model group. Conclusion: TLHT capsule can promote the recovery of neurological dysfunction of brain damage after ischemic injury in rats, alleviate the pathological damage of neurons, which may be related to its functions of promoting expression of Bcl-2 expression and inhibiting of the expression of Bax in ischemic penumbra, thereby reducing the level of apoptosis.

      Abstract:Objective: To observe the effects of Buyang Huanwu decoction(BYHWD) and Xinglou Chengqi decoction(XLCQD) on the nerves functional rehabilitation and its protective effect on aminotransferase, creatinine(Cr) and blood urea nitrogen(BUN) were evaluated for Liver and kidney function in rats with cerebral ischemia. Method: Rats were divided randomly into sham,model,nimodipine,BYHWD and XLCQD groups. Rat focal cerebral ischemia model was established by middle cerebral artery occlusion (MCAO) with nylon thread.Rats were administrated with corresponding drugs by gavage before establishing model. After operation of cerebral ischemia, rats were given the corresponding drugs once a day for 14 days.Rats neurological score (NS) was evaluated and blood was taken out at 14 d and 28 d after operation,and then the rat's liver and kidney function were determined. Result: In each model group,rats NS decreased,aspartate aminotransferase(AST),alanine aminotransferase(ALT),Cr,BUN increased. In comparison with that of rats in each model group, NS of rats in each treatment group raised, and AST,ALT,Cr,BUN reduced obviously except 14 d in BYHWD group. In comparison with that in nimodipine and XLCQD group,the NS increased in 28 d BYHWD group,Cr in 28 d BYHWD group reduced Significantly. NS of rats obviously ascended in each treatment group and AST,ALT decreased in 28 d BYHWD group. Conclusion: After cerebral ischemia the neurological function of rats restorated slowly. It is indicated that the treatment of XLCQD and BYHWD can Promote the recovery of neurological function injuried by cerebral ischemia and BYHWD has the more significant roles in the recovery of neurological function. Cerebral ischemia can lead to liver and kidney function damage, BYHWD and XLCQD play a different role in different time after cerebral ischemia, and the role of BYHWD is more comprehensive and significant.

    • Effects of Xiaochaihu Decoction on VIP,Gastric Emptying and Small Intestine Advance Rate in Functional Dyspepsia Rats

      Abstract:Objective: To observe the effects of Xiaochaihu decoction on vasoactive intestinal peptide(VIP),gastric emptying and small intestine advance rate in functional dyspepsia (FD) rats. Method: 48 rats were randomly divided into four groups: blank group, model group, Xiaochaihu decoction group and domperidone group, each group had 12 rats. FD models were established by chronic restraint stress and excessive fatigue and irregular feeding, once per day for continuous 3 weeks. Then feed the rats in different groups with normal saline, normal saline, Xiaochaihu decoction and Domperidone liquor respectively by intragastric administration for continuous 14 days. After 14 days intragastric administration, VIP of each rat was detected. Gastric emptying and small intestine advance rate of each rat were tested. Result: Compared to the blank group, gastrointestinal Hormone VIP and gastric residue obviously increased while small intestine advance rate significantly decreased in model group (P<0.05 or P<0.01). Compared to the model group, gastrointestinal hormone VIP and gastric residue obviously decreased while small intestine advance rate significantly increased in the Xiaochaihu decoction and domperidone group (P<0.05 or P<0.01). Conclusion: Xiaochaihu decoction can reduce gastrointestinal hormone VIP, advance gastric emptying and small intestine advance rate in functional dyspepsia rats.

      Abstract:Objective: Observe the pulmonary vascular pathological change in chronic obstructive pulmonary disease(COPD) rat models of application for activating blood circulation and eliminating stasis drugs, explore the vascular remodeling of COPD. Method: Make COPD models by droping lipopolysaccharide (LPS) into tracheal and passive smoking method. From 29th to 42th days, group H are gave intraperitoneal injection of Chuanqing solution 8 mL·kg-1·day-1; group C are gave volumetric saline. Execute the three groups rats in sixth weeks, observe the pathological changes of pulmonary vessels in rat models applied activating blood circulation and eliminating stasis drugs. Result: Compared to group N there are characteristically changes in group C and H,but different in degree. Compared to group N and C, vascular smooth muscle cells of group H have mild hyperplasia, the wall was thickened slightly, the ratio of blood vessel wall/blood vessel wall and hemal wall area/vascular area are significantly (P<0.01).Under the electron microscopy observation, the endothelial cells, the basement membrane, mitochondria and shelf small body of group N are normal; group C appear obvious changes. The pinocytotic vesicles of endothelial cells increased,mitochondrion edema or vacuoles degenerate. The nuclei have become obvious tendency of becoming pyknotic. Many nucleation white blood cells are blocked in the capillary lumen; endothelial cells, the basement membrane, mitochondria and lamellar body in group H are basically normal. There are a few nucleation white blood cells in the capillary lumens. Conclusion: Traditional Chinese medicine for activating blood circulation and eliminating stasis can slow or improve the blood vessels remodeling of COPD. It guides the treat should accord to its different stages in the process of development and combined with illness, use the drugs for activating blood circulation and eliminating stasis flexibly, give a scientific understanding of the importance in control of COPD to improve the clinical effect.

      Abstract:Objective: To investigate the anti-tumor mechanism of compound Shuang-zhu (BELY) anti-tumor effect in vitro and in vivo. Method: In experimental evaluation, eight groups were setup: control group, negative control group, six different concentrations of BELY groups (100,80,60,40,20,10, 5 μg·mL-1). CCK-8 kit was used to detect the proliferative effect of BELY on K562, MDA-MB-231, LoVo and HepG2 cells. Sixty health kunming mice was used to establish transplant tumor models of murine sarcoma S180 and of H22. Then they were randamly divided into six groups, including control group (NS), DMSO (1%) group, cisplatin group (1 mg·kg-1), three different concentration (100,200, 400 mg·kg-1) of BELY groups to observe the effect of BELY on tumor-bearing mice. ELISA was employed to detect of cytokines interleukin-2(IL-2), interleukin-12(IL-6), interleukin-12(IL-12) and TNF-alpha to reflect the effect of BELY on immune function of tumor-bearing mice. T cell subsets of CD4+and CD8+ in peripheral blood of tumor-bearing mice were measured by flow cytometry. Result: BELY showed a certain degree of inhibition on HepG2 and K562, LoVo, MDA231 cells in dose-effect relationship. BELY could significantly inhibit the growth of the murine sarcoma S180 and of H22, and improve the thymus index, spleen index and body weight of the tumor-bearing mice, BELY could significantly increase serum CD4+/CD8+. Compared with the blank control group, P<0.01 or P<0.05. Conclusion: BELY can not only directly inhibit tumor cells, but also enhance immune to achieve the anti-tumor action bidirectionally.

    • Analgesic and Anti-inflammatory Effect of the Alkaloid in Chelidonium majus Extract

      Abstract:Objective: To study the anti-inflammatory, analgesic effects of Chelidonium majus extract. Method: Writhing method, hot-plate method and formalin pain model were used to evaluate the analgesic effect. The inhibition of auricle edema induced by xylene and mouse peritoneal capillary permeability experiment were used to assess the anti-inflammatory effect. Naloxone test was used to interpret the analgesic mechanisms of the extract. Result: High dose group of C. majus extract significantly decreased the twisting times in mice induced by acetic acid(P<0.05),and obviously inhibited the swelling of mouse pinna induced by xylene(P<0.01). Conclusion: C. majus extract has significant analgesic and anti-inflammatory effects. Its analgesic effects are mainly peripheral and not antagonized by Naloxone.

      Abstract:Objective: To study the therapeutic action and mechanism of total flavonoids of Clerodendranthus spicatus (TFC) on rats models of chronic abacterial prostatitis. Method: Sixty healthy adult male SD rats were randomly divided into six groups, 10 rats in each group: blank control group, model group, positive control group (250 mg·kg-1·d-1), TFC low-dose, middle-dose and high-dose group (100, 200, 400 mg·kg-1), respectively. Except for normal group, the rats models of Chronic abacterial prostatitis were builded by subcutaneous injection of oestradiol monobenzoate at the dose of 0.25 mg·kg-1 after castration, and then treated with TFC at 100, 200, 400 mg·kg-1 by oral administration whileblank and model group were given normal saline, once a day for 30 days. White blood cell(WBC) count, lecithin corpuscle density and prostate index in prostatic fluid were examined, and the expressions of tumor necrosis factor(TNF-α) and interleukin-8(IL-8) in the serum and prostate tissues of the rats were detected by ELISA. Result: Compared with model group, WBC count and prostate index were reduced and lecithin corpuscle density were increased in prostatic fluid by TFC, which showed significant statistical differences(P<0.01 or P<0.05), and the levels of TNF-α and IL-8 in the serum and prostate tissues of the rats were reduced. Conclusion: TFC has therapeutic effects on chronic abacterial prostatitis inrats models, and the mechanism may be related to decreasing the levels of TNF-α and IL-8 in the serum and prostate tissues.

    • Study on Chronic Toxicity of Water Extraction Components from Evodia Fructus in Rats

      Abstract:Objective: To observe the chronic toxicity damage of the water extraction components from Evodia Fructus in rats. Method: The water extraction components with different doses from Evodia Fructus were separately orally given to rats, and the high, middle and low lose-group were separately 8.0,4.0,2.0 g·kg-1 calculated by crude herbs, the general conditions were observed and the related indexes were detected, such as the routine control of blood, hepatic function, renal function, the metabolism condition of lipids and the glycometabolism indexes; the rats were dissected and precisionly weigh the weight of heart, liver, spleen, lung, renal,and calculated the ratio of organ to body; the histopathologic examination of hepatic tissue were examined. The remaining rats were not administrated, and recovered for 20 days, the recovery condition of rats were observed,and the above indexes were detected. Result: The water extraction components from Evodia Fructus with different doses could induce the body weight decreasing in rats, the level of alanine aminotrans ferase(ALT),aspartate aminotransferase(AST),alkaline phosphatase(ALP) and total protein(TPC) serum obviously increased, albumin(ALB) and the ratio of ALB to GLB in serum obviously decreased, the liver weight and the ratio of liver to body increased, and the hepatic tissue damage was obvious in the histopathologic examination of hepatic tissue;the above-mentioned changes gradually aggravated with dose increasing, and it was the obvious discrepancy compared with control group. The influence on the content of total bilirubin(TBI),blood urea nitrogen(BUN), glucose(GLU),total cholesterol(CHO) and creatinine(Cr) was not obvious, and the hepatic toxicity degree had certain dose indepdence relatonship. The ALT level of high dose group in the above indexes of recovered rats had significant difference compared with control group, and the other indexes had no significant difference. Conclusion: The long term administration of water extraction components from Evodia Fructus at the certain dose in rats could induce obvious cumulative toxicity, and the mainly toxical injury sites were in liver, but after recovery period observation, the toxicity was basically recovered.

      Abstract:Objective: To investigate the effect of Xiaoke Wan and Xuezhikang on mannan binding lectin (MBL) in patients with type2 diabetes mellitus and hyperlipidemia. Method: Eighty patients with type2 diabetes mellitus combined with hyperlipidemia were randomly divided into observation group and control group (n=40 each). Observation group was firstly received Xiaoke Wan, 10 pills, before breakfast and lunch. Patients in observation group were also orally received Xuezhikang capsules, 2 capsules, bid. Control group was firstly received glibenclamide tablet, 2.5 mg before breakfast and lunch. Patients in control group were also orally received simvastatin, 20 mg daily. The amount of Xiaoke Wan and glibenclamide were adjusted according to blood glucose level in the two groups. The treatment course of the two groups lasted 12 weeks. Additional 20 normal persons were grouped as the healthy control group. Fasting blood glucose (FBG), level of postprandial 2 h blood glucose (2 hPG), glycated hemoglobin (HbAlc), total cholesterol (TC), triglyceride (TG), low density lipoprotein chotesterol(LDL-C) and high density lipoprotein chotesterol(HDL-C) and MBL were tested before and after treatment. Result: After treatment, FBG and 2 h PBG in the two groups were significantly lower than those before treatment, but there was no significant difference between the two groups. After treatment, HbA1c in the two groups was decreased significantly, and that in observation group decreased more (P<0.05). After treatment, there was no significant difference in blood glucose level between the two groups, LDL-C in control group was still higher than normal control group (P<0.05). After treatment, MBL in observation group was (0.71±0.25) mg·L-1, which was lower than that in normal control group (P<0.05), but significantly higher than that before treatment (P<0.01). MBL in control group was (0.40±0.21) mg·L-1, which was lower than that before treatment. Conclusion: Xiaoke Wan combined with Xuezhikang can effectively control blood glucose and lipids in the patients with type2 diabetes mellitus and hyperlipidemia, raise MBL level, and help to control inflammation.

      Abstract:Objective: To observe the efficacy and safety of rheumatoid arthritis treated with Busheng Quhan Zhiwan decoction and external bathing. Method: One hundred patients were divided into treatment group (Busheng Quhan Zhiwan decoction and external bathing) and control group (Celebrex 0.2 g, qd ig, methotrexate 10 mg qw ig, leflunomide 20 mg qd ig, total glucosides of paeonia 0.6 g, bid ig and symptomatic Chinese medicinal decoctions). After 24 weeks treatment, erythrocyte sedimentation rate (ESR), C-reactive protein (CRP), Rheumatoid factor (RF) and clinical scales was measured and safety was evaluated. Result: There was no significant difference in the effective rate of ACR 20 between two groups. The traditional Chinese medicine syndrome scores, joint pressure pain index number, joint swell index number,early morning stiffness, sufferers' break pain, 20-metre walking times, equally hold dint, Health Assessment Questionnaire(HAQ), ESR, RF and CRP of both groups was significantly improved after the treatments, but there were no significant difference between two groups. The common side effect of two groups was digestive system reaction while blood system impairment was found in the control group. Conclusion: Rheumatoid arthritis treated with Busheng Quhan Zhiwan decoction and external bathing had the same efficacy in clinical improvement and inflammation reduction with few adverse reactions, compared with the integrative therapy with both Chinese and western medicine. Busheng Quhan Zhiwan decoction and external bathing was suggested for clinical aplication.

      Abstract:Objective: To observe the clinical effects of Yifei capsule plus Tankejing powder on stable-stage patients with chronic obstructive pulmonary disease (COPD). Method: Ninety-eight subjects were randomly divided into observation group and control group (n=49 each). Both groups were adopted the conventional non-drug therapy, and were given drugs used for dissolveing sputum and antispasmodic on demand. The observation group was given Yifei capsule, 4 capsules tid, all for 12 days. Tankejing powder (0.2 g each time, 3-6 times daily) was given when abundance of phlegm and heavier cough occurred. The lung function, St George's respiratory disease questionnaire (SGRQ) and BODE index were measured to evaluated the quality of life. Result: The observation group showed significant improvement compared with the control group in the following indexes: forced expiratory volume in first second (FEV1), the ratio of FEV1 and forced vital capacity. There was significant difference between the two groups (P<0.05). The scores of SGRQ and the three dimensions of respiratory symptoms, activity and the affects of disease in the observation group were more lower than those in the control group (P<0.05). The scores of the three dimensions of FEV1, modified medical research council scale (mMRC) and 6 min walking distance (6 MWD) and BODE index were more lower than those in the control group (P<0.05). Conclusion: Yifei capsule plus Tankejing powder can significantly improve the lung function, clinical symptoms and BODE index in patients with COPD, and it can significantly improve the quality of life for patients.

      Abstract:Objective: To investigate the effect of Gansu granule on patients with chronic hepatitis B (CHB). Method: Eighty-six subjects were randomly divided into observation group and control group (n=43 each). Pegasys were given to the control group, 180 μg each time,1 time each week. Based on the pegasys treatment, Gansu granule was added to the observation group, 9 g, tid, all for 24 weeks. Interlukin-21 (IL-21) level, hepatitis B virus-DNA (HBV-DNA) and lung function were quantified and measured. Result: The level of HBV-DNA in both groups after the treatment were lower than those before, the observation group were more lower than that in the control group (P<0.05). IL-21 level in both groups after the treatment were higher than before, the level in observation group was higher than that in the control group (P<0.01). Aspartate aminotransferase (AST), alanine aminotransferase (ALT), γ-glutamete transpeptidase (GGT) and total bilirubin (Tbil) in the observation group after the treatment were lower than those in the control group (P<0.05, P<0.01), and albumin (Alb) of the observation group after the treatment were higher than that in the control group (P<0.05). Conclusion: Gansu granule can elevate IL-21 level for the patients with CHB. Gansu granule has anti-HBV function and could protect liver function.

      Abstract:Objective: To observe the effect of salvia miltiorrhiza and ligustrazine injection in combination with traditional Chinese medicine enema on chronic kidney disease (CKD), endothelin (ET), 6-keto-prostaglandin F1α (6-keto-PGF1α) and thromboxane B2 (TXB2). Method: Seventy patients with stage 3-4 CKD were randomly divided into observation group and control group (n=35 each). Control group was received western medicine therapy and rhubarb fluid enema. 200 mL rhubarb fluid were given via retention enema for 1-2 h and inserted into the intestine 20cm daily. The patients in observation group, on the basis of control group, were received salvia miltiorrhiza and ligustrazine injection 15 mL, diluted with 100 mL saline, iv, for 10 days. After two days' rest, the patients were received another course. There were two courses. Serum creatine (SCr), blood urea nitrogen (BUN), ET, 6-Keto-PGF1α and TXB2 were tested before and after treatment. Result: The total effective rate of observation group (71.43%) was higher than control group (45.71%) with a significant difference (P<0.05). BUN and SCr in observation group were lower than control group (P<0.05,P<0.01); After treatment, ET and TXB2 were decreased, and 6-keto-PGF1α increased in two growps. ET and TXB2 in observation group were lower than those in control group, however, 6-keto-PGF1α in observation group was higher than that in control group (P<0.05). Conclusion: Salvia miltiorrhiza and ligustrazine injection can decrease BUN and SCr in patients with stage 3-4 CKD, improve clinical efficacy, ameliorate hypercoagulative state and protect vascular endothelial function.

      Abstract:Objective: To investigate the effect of triple therapy of Chinese medicine for acute cerebral infarction and its influence on cerebral blood flow. Method: Two hundred and eighty-six patients with acute cerebral infarction were randomly divided into two groups. The observation group was accepted triple therapy of Chinese medicine, including Zhongfeng Fang 1 dose daily; Zhichang Dizhu Zhongfeng Fang 1 dose daily and ginkgo leaf injection 10 mL daily. The control group was treated with ginkgo leaf injection 10 mL daily, citicoline 0.5 g iv daily. All treatment lasted for 14 days. Recovery of neurological function and the changes of cerebral blood flow were evaluated and analyzed. Result: Compared with the control group, the total efficiency of the observation group was significantly higher (88.8% vs. 74.1%, P<0.05). The mean cerebral blood flow comparison of two groups after therapy showed statistically difference [(663.5±99.3) mL·min-1 vs (615.47±96.5) mL·min-1, P<0.05]. Conclusion: Triple therapy of Chinese medicine for acute cerebral infarction results in better cerebral blood flow recovery, and helps to improve the recovery of neurological function, which worthy of further clinical application.

      Abstract:Objective: To investigate the effect of modified Guizhi Fuling decoction combined swith routine western medicine therapy on polycystic ovarian syndrome (PCOS) and leptin (LP) in patients. Method: Ninety-four patients with PCOS were randomly divided into observation group (n=49) and control group (n=45). Patients in control group orally took ethinylestradiol and cyproterone acetate tablet, 1 tablet daily; metformin hydrochloride tablet, 0.5 g tid, from the third day of menstruation. There was a discontinuation at menstruation. Observation group was given modified Guizhi Fuling decoction, 1 dose daily, on the basis of control group. The treatment course of the two groups lasted 3 menstruation cycles. The patients' body mass index (BMI), fasting glucose and fasting insulin were recorded. Five indexes of endocrine were observed by radioimmunoassay. Serum LP was tested by enzyme linked immunosorbent assay. Result: After treatment, follicle stimulating hormone (FSH), luteinizing hormone (LH), estrodiol 2 (E2), testosterone (T) and prolactin (PRL) in the two groups were decreased than those before treatment. LH, E2 and T in observation group were lower than those in the control group (P<0.05); BMI in the two groups were decreased compared with those before treatment, and BMI in observation group were lower than that in control group (P<0.05); Serum LP in observation group was (9.68±2.59) μg·L-1, which was significantly lower than (10.75±2.67) μg·L-1 in control group (P<0.05). Conclusion: Modified Guizhi Fuling decoction can improve insulin resistance by significantly decreasing LH, E2, T and LP and reducing BMI.

      Abstract:Objective: To observe clinical curative effect of Qiaohe plus decoction (QPD) for treating acute pharyngitis. Method: One-hundred and twenty-four patients with acute pharyngitis were divided into two groups at random(62 cases by QPD,1 dose daily; 62 cases by cefalexin capsules, 0.5 g, tid). Result: The total effective rates of treatment group and control group were 96.77% and 82.26% respectively,difference between the two groups was significant (P<0.05). The adverse reaction in treatment group and control group were 3.23% and 11.29% respectively, difference between the two groups had statistical significance (P<0.05). Conclusion: QPD has a better efficacy for the treatment of acute pharyngitis,and shows less side effect.

    • Clinical Observation for Treating Chronic Urticaria Based on Spleen Differentiation

      Abstract:Objective: To observe the effect of treating chronic urticarial (CU) based on spleen differentiation. Method: This prospective, noncomparative interventional case series observed 72 patients with CU which were divided into three groups based on spleen differentiation: the group with deficiency of spleen (group 1) was given modified Shenling Baizhu San, the group with Qi deficiency of spleen and lungs (group 2) was given modified Yupingfeng San, and the group with deficiency of heart and spleen syndrome (group 3) was given modified Guipi Tang. Items such as changes in clinical symptoms and signs, recurrence after treatment, and side effects were examined. Result: After the treatment, satisfactory effects showed that total efficacy rate of group 1 was 92.31%, group 2 was 91.67%, group 3 was 90.91%. No significant deviation between the groups was found. And after 2 month follow-up for the cured patients, 2 patients in group 1 showed relapse, and 1 in group 2, 2 in group 3.So, no significant deviation was found for relapse. There were no side effect or abnormal phenomenon was found on the items of blood routine, urine routine, liver function and kidney function as well. Conclusion: Treating chronic urticarial based on spleen differentiation is a safe and effective therapeutic procedure, which is worth the further study and clinical application.

      Abstract:Objective: To observe the efficacy of Chinese herbal medicine on constipation induced by diabetes according to differentiation of syndrome and its effect on blood glucose and glycosylated hemoglobin. Method: One hundred and twenty cases of constipation induced by diabetes randomly divided into two groups with 60 patients in each group. The treatment group was treated with Chinese herbal medicine decoction based on the routine treatment while the control group was treated with mosapride and Vitamin B12 for 14 days. Stool,blood glucose and glycosylated hemoglobin before and after treatment were investigated. Result: The total effective rate of constipation was 93.5% in the treatment group, but only 74.2% in the control group (P<0.01). The blood sugar level before and after meal and glycosylated hemoglobin in the two groups were decreased. Treatment group was significantly better compared with the control group (P<0.05). Conclusion: Chinese herbal medicine based on syndrome differentiation shows positive effect on releasing constipation induced by diabetes, and helps to improve blood glucose.

      Abstract:Objective: To observe the clinical effects of Xuebijing injection on the treatment of patients with old hypostatic pneumonia. Method: Sixty-four subjects were randomly divided into observation group and control group (n=32 each). The control group was given 1.5 g drugs combined with cefoperazone sodium and sulbactam (2:1), which were diluted by 100 mL 5% glucose injection (bid); and they were given ambroxol hydrochloride injection which contained 30 mg ambroxol hydrochloride diluted by 100 mL normal saline meanwhile (bid). On the basis of the control group intervention, the treatment group was given 50 mL Xuebijing injection diluted by 100 mL normal saline by intravenous drip (bid). All the intervention in two groups lasted 14 days. Cough and expectoration, body temperature, blood picture and lung condition by X-ray were observed. Result: The total symptom scores of the observation group in the 4, 7 and 14 day were lower than those in the control group (P<0.05, P<0.01).In the observation group, the time for body temperature to restore normal, cough and expectoration disappeared time, the time for lung to restore normal and the time for blood picture back to normal were less than those in control group (P<0.05). The clinical effects of the observation group were superior than that of control group (P<0.05). Conclusion: Xuebijing injection can significantly improve clinical signs and symptoms for patients with old hypostatic pneumonia and enhance clinical effects so as to promote the recovery of disease.

    • Effect of Shenkang Injection as an Adjunctive Treatment on Early Diabetic Nephropathy and on Patient's Oxidative Stress

      Abstract:Objective: To observe the effect of Shenkang injection as an adjunctive treatment on early diabetic nephropathy (DN) and on patients' oxidative stress. Method: Sixty-six DN patients were randomly divided into observation group and control group (n=33 each). The patients in control group were received tight control of diet, blood glucose and blood pressure to maintain normal levels. They were orally taken enalaprilat, 5 mg daily. The patients in observation group, on the basis of control group, received Shenkang injection 100 mL mixed with 300 mL 5%glucose, iv,gtt, 1 time daily. The treatment course of the two groups lasted 4 weeks. Urinary albumin excretion rates (UAER), β2-microglobulin (β2-MG), albumin to creatinine ratio (ACR), oxidative stress indexes malondialdehyde (MDA), superoxide dismutase (SOD) and total antioxidant capacity in the two groups were observed. Result: After treatment, UAER, β2-MG and ACR in observation group were lower than those in control group (P<0.01); SOD and T-AOC in observation group were significantly increased and higher than those in control group (P<0.01). MDA in observation group was significantly decreased and lower than that in control group (P<0.01). The total effective rate of observation group (84.84%) was higher than that in control group (63.64%) with a significant difference (P<0.01). Conclusion: Shenkang injection can decrease UAER, β2-MG and ACR in patients with early diabetic nephropathy and improve clinical efficacy, the mechanism of which may be associated with reducing oxidative stress and enhancing anti oxidative stress ability.

    • Yunnan Baiyao for Promoting Postoperative Rehabilitation of Cervical Intraepithelial Neoplasia by Cold Knife Conization

      Abstract:Objective: To observe the clinical effects of Yunnan Baiyao for promoting postoperative rehabilitation of cervical intraepithelial neoplasia by cold knife conization (CKC). Method: Ninety-six postoperatives of cervical intraepithelial neoplasia Ⅲ by CKC were randomly divided into observation group and control group (n=48 each). CKC postoperative wound was fulgerized and packed with gauze strip for the control group and conventional anti-infection treatment was given for 4 days; CKC postoperative wound was fulgerized and covered with Yunnan Baiyao powder and packed with gauze strip for the observation group and conventional anti-infection treatment was given for 4 days. Yunnan Baiyao capsule was added to the observation group, 2 softgels each time, tid, all for 5 days. Postoperative vaginal drainage time, drainage quantity, vaginal bleeding time, amount of bleeding, decrustation of cervical scab and cervical wound healing was observed in 1, 2,5,8 weeks. Result: Postoperative vaginal bleeding time and average amount of bleeding of the observation group were lower than the control group (P<0.01); Postoperative vaginal drainage time and average drainage quantity in the observation group were lower than those in control group (P<0.01). For the observation group, there was 23 cases after operating 5 weeks and 45 cases after operating 8 weeks of wound healing which were superior to 12 cases and 34 cases of the same period in the control group (P<0.05). Cleaning degree of vagina after 5 weeks in the observation group was obviously superior to the control group (P<0.01); there was no obvious difference between the curative effect of the two groups. Conclusion: Postoperatives of cervical intraepithelial neoplasia Ⅲ by CKC treated with electrocautery and covered wound with Yunnan Baiyao powder, and supplemented with Yunnan Baiyao capsule orally could reduce postoperative complications and promote recovery.

      Abstract:Objective: To investigate the clinical effects of Naomaitai capsules plus compound cerebroprotein hydrolysate tablets on old patients with convalescing cerebral infarction. Method: Seventy two subjects were randomly divided into observation group and control group (n=36 each). Both groups were given aspirin enteric-coated tablets to antiplatelet aggregation and were adopted the basic treatment for controlling blood glucose and blood pressure and regulating blood fat. The control group were given compound cerebroprotein hydrolysate tablets, 4 tablets each time, tid; Naomaitai capsules was added to the observation group, 2 capsules each time, tid, all for 8 weeks. Mental state and daily living ability of the two groups were evaluated by mini-mental status examination (MMSE) and activity of daily living scale (ADL). Result: The observation group was higher than the control group in the scores of MMSE (P<0.01) and was lower than the control groups in the scores of ADL (P<0.05); the total effective rate for convalescent stroke in the observation group was superior to the control group (83.33% vs 61.11%) (P<0.05). The total effective rate for treating traditional Chinese medicine symptom in the observation group was superior to the control group (91.67% vs 66.67%) (P<0.05). Conclusion: Naomaitai capsule can significantly improve cognitive function and daily living ability of patients with convalescing cerebral infarction, and has a significant effect on improving the neurologic function of them, thus it can enhance the curative effect for treating disease; it also has obvious improvement on 'Fengtanyuxue' syndrome and can relieve the clinical symptoms so as to improve the curative effect.

    • Analysis on Clinical Trials of Chinese Medicinal Injection Used in Stroke Concerning Rational and Safe Medication

      Abstract:Objective: To evaluate proper and safe medication in acute phase of ischemic stroke treated with Chinese medicinal injection. Method: Clinical studies involving acute stage of ischemic stroke treated with Chinese medicinal injection published in Chinese from 2012 January to November were electronically searched from CBM, CNKI, and CMJD and assessed by the inclusive and exclusive criteria. The data were extracted, grouped by the category, administration, dialectic medication, combination and safety evolution and then analyzed. Result: Eighty-three papers were included finally. 18 kinds of Chinese medicinal injection were used altogether. All were administrated by intravenous injection once per day, while the daily dose, first use time, and treatment course were different. None of injections was applied by the principle of symptom differentiation. The combination treatment included platelet aggregation inhibitors, anticoagulants, anti-fibrinogen, neuroprotectors, dehydrant, thrombolysis, rehabilitation and high pressure oxygen, except Chinese medicinal injection and decoction of Chinese herbs. Fifty-five studies conducted safety assessment and 40 of them found no adverse reaction. Conclusion: The unrational application of Chinese medical injections existsin in Chinical research. The principle of symptom differentiation and strict conduction with therapeutic indication are suggested in research and clinical application of Chinese medicinal injection. More attention should be paid to safety monitoring and side effect reporting.

      Abstract:Objective: It is to explore the effect of Aidi Injecttion on the expression of radiotherapy for lung cancer patients. Method: Fourty cases of lung cancer were randomly divided into treatment group (20 cases) and control group (20 cases).The two groups were given three-dimensional conformal radiation therapy (3D CRT).Meanwhile,the treatment group was added Aidi injecttio.There were some indices compared before and after radiotherapy including the serum interleukin-6(IL-6), the tumor necrosis factor-α(TNF-α) and the transforming growth factor-β(TGF-β).And then,the incidence of the radiation pneumonia were also compared between two groups. Result: After the end of treatment,the IL-6, the TNF-α and the TGF-β of the treatment group were lower than those of the control group. There was significant difference(P<0.05). Nevertheless there were no significat differences between two groups about incidence of the radiation pneumonitis. Conclusion: Aidi injecttion could relieve the progress of Radiation pneumonitis through decreasing levels of the IL-6,the TNF-α and the TGF-β,thus,prevention of the occurrence of radiation pneumonia.

      Abstract:Objective: To comparative analysis flurbiprofen ester joint sufentanil, and used alone sufentanil, in the patients after intestinal surgery produced by the analgesic effect and metabolic toning effect. Method: Will be in february 2011-July 2012 received intestinal cancer radical operation of 80 patients as experimental investigation of the sample, random divided into sufentanil analgesic group (group A, n=40) and fluorine than flurbiprofen ester joint sufentanil analgesic group (group B, n=40). Due to A, B group of patients after surgery after 3 days shall not eat, can only through the week intravenous nutrition supplement. In this time logging A, B group of patients in the stage of fast the tumor necrosis factor-α(TNF-α), body temperature numerical, cortisol, nitrogen balance, the integral effect of analgesia, interleukin-6(IL-6) and adrenal line changes. Result: The two groups of analgesic score difference was statistically significant. In the third day A, B group compared with patients who the analgesic effect of 1 day all appeared obviously improved. Postoperative body temperature, negative nitrogen balance, TNF-α and IL-6 change group B than a group obviously. But adrenaline numerical and cortex leaven numerical and no apparent change. Conclusion: Compared with the use of a single sufentanil on postoperative analgesia, sufentanil and flurbiprofen ester mixed analgesic effect and no obvious advantages, but sufentanil and flurbiprofen ester mixture can play a good postoperative conditioning effect.

      Abstract:Objective: To discuss the clinical analysis of integrated traditional Chinese and western medicine in the treatment of upper gastrointestinal hemorrhage. Method: From January 2010 to December 2011 in our hospital, the 136 cases of upper gastrointestinal hemorrhage were randomLy divided into two groups,67 cases patients were given Conventional treatment coupled with Losec injection treatment as the control group, 69 cases patients are based on the control group coupled with compound traditional Chinese medicine treatment as the research group. Then compared with hemostatic time, effective time, treatment effect, blood transfusion amount and the adverse reaction rate of two groups. Treating for a week continually. Result: Compared with the total effective rate of the control group was 77.6%, the total effective rate of the research group was 94.2%, which was improved effectly, there were significant differences (P<0.05); Compared with hemostatic time(10.68±2.84) h, effective time (9.49±1.64) h and blood transfusion amount(2.86±1.53) U of the control group. all indexes of research group was improved effectly, there were significant differences; Compared the the adverse reaction rate of the control group was 7.6%, the adverse reaction rate of the research group was 5.8%, which was not improved effectly, there were no significant differences. Conclusion: Integrated traditional Chinese and western medicine in the treatment of upper gastrointestinal hemorrhage are better than western medicine treatment, which has effective curative effect. The adverse reaction rate is low, so it is worthy of popularization.

      Abstract:Objective: Observed Fuzheng Xiaoji decoction combined with chemotherapy drugs clinical efficacy for the treatment of advanced colon cancer. Method: Pathological diagnosis of 91 patients with Advanced colon cancer were randomLy divided into two groups,the control group of 30 patients,FOLFOX chemotherapy alone;treatment group,61 cases,FOLFOX chemotherapy with the control group,in conjunction with Fuzheng Xiaoji decoction,150 mL of morning and evening services.Both groups were given the same antiemetics and symptomatic treatment program. Result: Recent efficacy:treatment group complete remission(CR) 9 patients (14.8%),partial remission(PR) 22 cases of (36.1%),50.8% efficiency;CR 4 patients of the control group (13.3%),PR in 10 patients (33.3%),46.7% efficiency.The two groups was statistically no significant difference.Peripheral blood after treatment,observation and comparison of physical (KPS),changes in immune function,the treatment group can reduce the impact of chemotherapy on the peripheral blood,immune function,KPS score,the difference was statistically significant (P<0.05, 0.01). Conclusion: Fuzheng Xiaoji Decoction combined with chemotherapy can reduce the side effects of chemotherapy, and to improve the quality of life, improve the therapeutic effect.

      Abstract:Objective: To explore combine traditional Chinese and western medicine treatment for the influence of coronary heart disease patients plasma brain natriuretic(BNP) level of the change. Method: Select 120 cases from the patients with coronary artery disease.in January 2010 to December 2012 Rrandomly divided into control group were 60 cases given the conventional western medicine treatment, and 60 cases in the research group. Control group on the basis of the treatment with Chinese medicine,the period of treatment are 2 weeks.Compared between the two groups before and after treatment and curative effect of two groups of plasma BNP index change. Result: Compared with control group(68.3%),the effect of the rearch group(86.7%) is obvious increase, significant difference, there is statistical significance (P<0.05); aftert the treatment, compare with the two groups of patients plasma BNP level,the rearch team obviously improved, significant difference, there is statistical significance (P<0.01). Conclusion: raditional Chinese and western medicine treatment of coronary heart disease (CHD) efficacy is obvious, is better than that of pure western medicine treatment, can significantly improve the plasma BNP level. Worth clinical promotion.

    • Effect of Compound Danshen Droppills on Vascular Endothelial Function in Early Diabetic Retinopathy Patients

      Abstract:Objective: To investigate the changes of vascular endothelial function in early diabetic retinopathy patients and the effect of compound Danshen droppills on intervention. Method: Fixty-five patients with early diabetic retinopathy were selected and divided into two groups(routine group and Danshen droppills group).routine group was treated with insulin; the Danshen droppills group was was treated with Compound Danshen droppills (10# Tid)combined with insulin. 31 healthy as control group. After 8 weeks treatment, the endothelium-dependent flow-mediated dilatation(FMD) of brachial artery was determined with ultrasound, and the levels of fasting blood glucose(FBG), glycosylated hemoglobin(HbA1c),cholesterol (TC), triglyceride (TG), high sensitivity C-reactive protein (hs-CRP), intercellular adhesion molecule-1(ICAM-1)and endothelin-1(ET-1) in three groups were measured respectively before and after treatment. The change of fluorescein angiography in early diabetic retinopathy patients were observed before and after treatment. Result: All patients with early diabetic retinopathy showed that the FBG, HbA1c,TC,TG,hs-CRP, ICAM-1 and ET-1 was higher than that of the controlled group(P<0.05), the FMD was lower than the controlled group(P<0.05). After treatment fundus fluorescence angiography in Danshen droppills group was obviously improved than befor therapy,the hs-CRP, ICAM-1,ET-1 in the Danshen droppills group[(8.76±1.26)mg·L-1, (407.82±87.45) μg·L-1,(96.68±10.69) ng·L-1] was decreased more significantly than routine group[(18.78±2.42)mg·L-1,(597.79±60.74) ng·L-1,(184.5±10.78) ng·L-1] (P<0.05).After treatment the FMD in the Danshen droppills group was increased more significantly than routine group [(11.88±3.57)% vs (9.17±0.54)%](P<0.05). Conclusion: The compound Danshen droppills can effectively reduce the body's inflammationrecovery and recovery vascular endothelial function in patients with early diabetic retinopathy.

      Abstract:Objective: To evaluate the curative efficacy of Fuyang Xuanbi decoction on treatment of elderly nerve root cervical spondylosis. Method: Total 98 patients were randomly assigned two groups,48 cases were in the treatment group and 50 cases in the control group. Patients in treatment group were treated with Fuyang Xuanbi decoction and traction,while those of control group were treated with Fuyang Xuanbi decoction only. Then compared the efficacy between them after 20 days. Result: The efficient rate in treatment group was 93.8% and that in control group was 80.0%. The efficacy in treatment group was superior to that in control group (P<0.05). Conclusion: The efficacy of Fuyang Xuanbi decoction and traction to treat elderly nerve root cervical spondylosis is significant.

    • Study on Compatibility Law of Chinese Herbs with Neutral-nature Based on the Data Base

      Abstract:Objective: By analyzing the used law of different nature herbs in 1 307 prescriptions, research the compatibility law of chinese herbs with neutral-nature. Method: Screened 1 307 prescriptions from two books, science of prescription, and Pharmacopoeia of the People's Republic of China, judged the nature of herbs according to Pharmacopoeia, established the data base with the obtained data named Data analysis system of Chinese heabs and prescription. Research the compatibility law of chinese herbs with neutral-nature with the data base, defined the common heabs whose used frequency was upper limit 10% actored as sovereign, minister, assistant and courier, and defined the major herbs whose used frequency was upper limit 25%. Result: It is the used frequencies of heabs with different nature used as different actors in 1 307 prescriptions, that cold 42%, neutral 15%, hot 43% separately used as sovereign heab, cold 40%, neutral 17%, hot 43% separately used as minister heab, cold 37%, neutral 20%, hot 43% separately used as assistant heab, and cold 17%, neutral 66%, hot 17% separately used as assistant courier heab. Screened the common and major heabs with nentral nature used as sovereign, minister, assistant and courier. Conclusion: The feature of heab with neutral nature in compatibility. is less used as sovereign, minister heabs, more used as assistant and courier heabs. The screened commoned and majored heabs with neutral nature will provide a useful reference.

Result: received 234 literatures accorded to the standard, from the statistical analysis,in clinical the using of the Astragalus drug frequency is 33.0%,72.7% use oral mode of administration; the using of the Sophora drug frequency is 19.7%,79.3% use enema mode of administration; Derived from the statistical results, the clinical treatment of UC drugs and drug delivery is according with the consistency of design ideas. Conclusion: The design ideas of the studying agree with nearly 20 years of UC clinical medicine practical.So speculate the designed and developed by preparations safe and effective.This conclusion is also consistent with previous studies of this group.

      Abstract:Chinese medicine believes that the cause of postmenopausal ovarian dysfunction and the decrease in estrogen is kidney deficiency, including kidney Yin deficiency, kidney Yang deficiency and kidney Yin and Yang deficiency syndrome. We can use the kidney yang-tonifying medicinal to treat the disorder of lipid metabolism and related diseases caused by estrogen reduce and kidney yang deficiency. On the one hand, it can warm kidney Yang, improve ovarian function, increase the estrogen, on the other hand, it can warm and tonify kidney Yang, enhanced gasification to educe lipid-lowering effect. Consult the articles, we found that many kidney yang-tonifying drugs not only have estrogen-like effects, but also have the regulating effect of lipid metabolism, such as Herba Epimedii, Cornu Cervi Pantotrichum, Cortex Eucommiae, Fructus Psoraleae, Semen Cuscutae, Fructus Cnidii, Herba Cistanches, Semen Astragali Complanati. Some kidney yang-tonifying drugs have the regulating effect of lipid metabolism, such as Cordyceps, Cortex Cinnamomi, Fructus Rosae Laevigatae. Some kidney yang-tonifying drugs have estrogen-like effects, such as Placenta Hominis, Radix Dipsaci, Gecko. Summarize the kidney yang estrogen-like effects and lipid regulating effect mechanisms can provide the basis for depth study of the estrogen-like effects and lipid-regulating mechanisms of the kidney yang-tonifying drugs.
